Description:
Insulin receptor substrates (IRS) are responsible for several insulin related activities, such as glucose homeostasis, cell growth, cell transformation, apoptosis and insulin signal transduction. Serine/threonine phosphorylation of IRS1 has been demonstrated to be a negative regulator of insulin signaling and is responsible for its degradation, although IRS1 degradation pathways are not well understood. IRS1 has also been shown to be constitutively activated in cancers such as breast cancer, Wilm's tumors, and adrenal cortical carcinomas, thus making IRS1 phosphorylation and subsequent degradation an attractive therapeutic target. To date there have been four subtypes identified: IRS1, 2, 3 and 4, with IRS1 being widely expressed.

IKK beta (I-Kappa-B kinase-beta) is a member of the IKK complex which is composed of IKK alpha, IKK beta, IKK gamma and IKAP. Phosphorylation of I-Kappa-B on a serine residue by the IKK complex frees NF-kB from I-Kappa-B and marks it for degradation via ubiquination. IKK beta has been shown to activate NF-kB and phosphorylate IKB alpha and beta. Phosphorylation of 2 sites at the activation loop of IKK beta is essential for activation of IKK by TNF and IL1. Once activated, IKK beta autophosphorylates which in turn decreases IKK activity and prevents prolonged activation of the inflammatory response. Additionally, IKK beta activity can also be regulated by MEKK1.

NPDC-1 (Neural Proliferation Differentiation and Control-1) is expressed in neurons once they have stopped dividing and begun to differentiate. NPDC-1 is transported from the Golgi apparatus via vesicles before becoming internalized by endosomes at the cell membrane. NPDC-1 interacts with Cdk2, D-type cyclins, and the transcription factor E2F1. This interaction can lead to an increased replication time, and might have implications in final neural differentiation and apoptosis. NPDC-1 has been shown to colocalize with synaptic vesicle proteins: synaptophysin, synaptobrevin 2, and Rab3 GEP (Rab3 GTP/GDP exchange protein). One function of NPDC-1 is to regulate retinoic acid-mediated events by directly interacting with retinoid receptors. The amino acid sequence of NPDC-1 is highly conserved between mouse, rat, and human.

Phemx is a member of the tetraspanin (TM4SF) family of proteins that may be involved in transmembrane signal transduction, regulation of cell proliferation, differentiation and motility. Phemx is a multi-pass membrane protein containing intracellular N- and C-terminal domains, four transmembrane domains and two extracellular loops. It is ubiquitously expressed from early embryogenesis through adulthood. Phemx exhibits predominant expression in hematopoietic tissues suggesting a role in hematopoietic-cell function. In association with the Integrin αIIb/Integrin β3 complex, Phemx functions to stabilize arterial thrombi in platelets and regulate “outside-in†signaling. This interaction may be important in the process of wound healing. The gene encoding Phemx is located in an important tumor-suppressor gene region that has been associated with Beckwith-Wiedemann syndrome as well as a variety of cancers.

This is a paternally expressed imprinted gene that encodes transcripts containing two overlapping open reading frames (ORFs), RF1 and RF1/RF2, as well as retroviral-like slippage and pseudoknot elements, which can induce a -1 nucleotide frame-shift. ORF1 encodes a shorter isoform with a CCHC-type zinc finger motif containing a sequence characteristic of gag proteins of most retroviruses and some retrotransposons. The longer isoform is the result of -1 translational frame-shifting leading to translation of a gag/pol-like protein combining RF1 and RF2. It contains the active-site consensus sequence of the protease domain of pol proteins. Additional isoforms resulting from alternatively spliced transcript variants, as well as from use of upstream non-AUG (CUG) start codon, have been reported for this gene.
