Description:
JAK2 (Janus Activating Kinase 2) is a tyrosine kinase of the non-receptor type, that associates with the intracellular domains of cytokine receptors; JAK2 is the predominant JAK kinase activated in response to several growth factors and cytokines such as IL-3, GM-CSF and erythropoietin; it has been found to be constitutively associated with the prolactin receptor and is required for responses to gamma interferon. Ligand binding to a variety of cell surface receptors (e.g., cytokine, growth factor, GPCRs) leads to an association of those receptors with JAK proteins, which are then activated via phosphorylation on tyrosines 1007 and 1008 in the kinase activation loop. Activated JAK proteins phosphorylate and activate STAT (signal transducers and activators of transcription) proteins, which then dimerize and translocate to the nucleus. Once in the nucleus, STAT proteins bind to DNA and modify the transcription of various genes.

Description:
Plays a role in the regulation of the actin cytoskeleton through its interactions with actin capping protein (CP). May function to target CK2 to the plasma membrane thereby serving as an adapter to facilitate the phosphorylation of CP by protein kinase 2 (CK2). Appears to target ATM to the plasma membrane. Appears to also inhibit tumor cell growth by inhibiting AKT-mediated cell-survival. Also implicated in PI3K-regulated muscle differentiation, the regulation of AP-1 activity (plasma membrane bound AP-1 regulator that translocates to the nucleus) and the promotion of apoptosis induced by tumor necrosis factor TNF. When bound to PKB, it inhibits it probably by decreasing PKB level of phosphorylation.

Description:
This gene encodes a member of the ALEX family of proteins which may play a role in tumor suppression. The encoded protein contains a potential N-terminal transmembrane domain and a single Armadillo (arm) repeat. Other proteins containing the arm repeat are involved in development, maintenance of tissue integrity, and tumorigenesis. This gene is closely localized with other family members on the X chromosome. Three transcript variants encoding the same protein have been identified for this gene. [provided by RefSeq, Jul 2008].

Description:
Humanin, an endogenous anti-apoptotic peptide against Alzheimer disease-related insults, consists of 24 amino acids. The secreted protein is a neuroprotective factor against death induced by several different types of Alzheimer's disease genes. Humanin protects neuronal cells from damage caused by Alzheimer's disease genes, specifically APP (amyloid precursor protein). Humanin acts as a ligand for formyl peptide receptor-like 1 (FPRL1) with APP and utilizes its neuroprotective effects by inhibiting FPRL1 access to APP. The peptide prevents Bax translocation from cytosol to mitochondria. Humanin expression levels may be dependent on defects in energy production in muscles with mitochondrial abnormalities. The peptide has been detected in muscles of patients with the mitochondrial disease chronic progressive external ophthalmoplegia (CPEO). Humanin is mainly expressed in the kidney, heart, liver, testis and skeletal muscles.

Description:
Associates with components of the Mediator and p160 coactivator complexes that play a role as intermediaries transducing regulatory signals from upstream transcriptional activator proteins to basal transcription machinery at the core promoter. Recruited to endogenous nuclear receptor target genes in response to the appropriate hormone. Also functions as a p53 coactivator. May thus play an important role in transcriptional regulation (By similarity). May be involved in apoptosis signaling in the presence of the reinoid CD437. Apoptosis induction involves sequestration of 14-3-3 protein(s) and mediated altered expression of multiple cell cycle regulatory genes including MYC, CCNB1 and CDKN1A. Plays a role in cell cycle progression and/or cell proliferation.

Description:
Histone methyltransferase involved in left-right axis specification in early development and mitosis. Specifically trimethylates 'Lys-9' of histone H3 (H3K9me3). H3K9me3 is a specific tag for epigenetic transcriptional repression that recruits HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. Contributes to H3K9me3 in both the interspersed repetitive elements and centromere-associated repeats. Plays a role in chromosome condensation and segregation during mitosis.

Description:
Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. These receptors are heteromeric protein complexes composed of multiple subunits, arranged to form ligand-gated ion channels. The classification of glutamate receptors is based on their activation by different pharmacologic agonists. The subunit encoded by this gene belongs to a family of AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate)-sensitive glutamate receptors, and is subject to RNA editing (AGA->GGA; R->G). Alternative splicing of this gene results in transcript variants encoding different isoforms, which may vary in their signal transduction properties.

Description:
Receptor for retinoic acid. Retinoic acid receptors bind as heterodimers to their target response elements in response to their ligands, all-trans or 9-cis retinoic acid, and regulate gene expression in various biological processes. The RXR/RAR heterodimers bind to the retinoic acid response elements (RARE) composed of tandem 5'-AGGTCA-3' sites known as DR1-DR5. In the absence or presence of hormone ligand, acts mainly as an activator of gene expression due to weak binding to corepressors. In concert with RARG, required for skeletal growth, matrix homeostasis and growth plate function.

Description:
GBGT1 is a 347 amino acid single-pass type II membrane protein that belongs to the glycosyltransferase 6 family. Localizing to the golgi apparatus membrane, GBGT1 is widely expressed, with high levels found in placenta, ovary and peripheral blood leukocyte, and lower levels expressed in liver, thymus and testis. GBGT1 utilizes manganese as a cofactor, and assists in the addition of N-acetylgalactosamine (GalNAc) in alpha-1,3-linkage to various substrates, resulting in the formation of glycolipids. Glycolipids are present in most eukaryotic cells and may assist in the adherence of certain pathogens. Existing as two alternatively spliced isoforms, the gene encoding GBGT1 maps to human chromosome 9q34.2 and mouse chromosome 2 A3.

Description:
In the central nervous system GABA functions as the main inhibitory transmitter by increasing a Cl-conductance that inhibits neuronal firing. GABA has been shown to activate both ionotropic (GABAA ) and metabotropic (GABAB ) receptors as well as a third class of receptors called GABAC. In addition to GABA receptors, several proteins have been identified as regulators of GABA function, including GAD65, GAD67, GABA transporters and GABARAP (GABAA receptor-associated protein). GABARAP associates with GABAA Rg2 to link GABAA receptors to the cytoskeleton. The GABARAP protein sequence is similar to light chain-3 of microtubule-associated proteins (MAPs) suggesting that it may be a type of MAP or a component of a MAP complex.

Description:
Has a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. Plays an important role in memory formation and synaptic plasticity in the hippocampus (By similarity). Receptor for SIRPA, binding to which prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells. Interaction with SIRPG mediates cell-cell adhesion, enhances superantigen-dependent T-cell-mediated proliferation and costimulates T-cell activation. May play a role in membrane transport and/or integrin dependent signal transduction. May prevent premature elimination of red blood cells. May be involved in membrane permeability changes induced following virus infection.

Description:
Microtubules are components of the Actin cytoskeleton that play crucial roles in cell morphogenesis, cell motility, spindle formation and chromosome movements. Echinoderm microtubule-associated (EML) proteins function to modify the assembly dynamics of microtubules. EML4 (Echinoderm microtubule-associated protein-like 4), also known as EMAPL4, ELP120, C2orf2 or ROPP120, is a 981 amino acid cytoplasmic protein that contains nine WD repeats. Expressed at high levels during mitosis, EML4 is thought to modify the assembly dynamics of microtubules, specifically altering microtubules to become longer and more flexible. Due to a chromosomal inversion with chromosome 2p, EML4 may exist as a fusion protein with ALK (anaplastic lymphoma receptor tyrosine kinase), producing an EML4-ALK fusion complex that plays a role in the pathogenesis of lung cancer.
