Description:
Sox-30 encodes a member of the Sox (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. Sox-30 is expressed in testis and may act as a transcriptional regulator after forming a protein complex with other proteins. Sox-30 may be involved in the differentiation of developing male germ cells. Two transcript variants encoding distinct isoforms have been identified for the human Sox-30 gene. Sox family transcription factors influence cell differentiation, development and sex determination. Sox-30 contains a unique DNA binding domain, known as the high mobility group (HMG) box, that is related to that of the testis determining gene, SRY. The highly complex group of Sox genes cluster at a minimum of 40 different loci that rapidly diverged in various animal lineages. Several Sox genes have been identified, and members of this family have been shown to be conserved during evolution and to play key roles during animal development.

c-Src tyrosine kinase plays a critical role in signal transduction downstream of growth factor receptors, integrins and G protein-coupled receptors. We used stable isotope labelling with amino acids in cell culture (SILAC) approach to identify additional substrates of c-Src tyrosine kinase in human embryonic kidney 293T cells. We have identified 10 known substrates and interactors of c-Src and Src family kinases along with 26 novel substrates. We have experimentally validated 4 of the novel proteins (NICE-4, RNA binding motif 10, FUSE-binding protein 1 and TRK-fused gene) as direct substrates of c-Src using in vitro kinase assays and cotransfection experiments. Significantly, using a c-Src specific inhibitor, we were also able to implicate 3 novel substrates (RNA binding motif 10, EWS1 and Bcl-2 associated transcription factor) in PDGF signaling. Finally, to identify the exact tyrosine residues that are phosphorylated by c-Src on the novel c-Src substrates, we designed custom peptide microarrays containing all possible tyrosine-containing peptides (312 unique peptides) and their mutant counterparts containing a Tyr --> Phe substitution from 14 of the identified substrates. Using this platform, we identified 34 peptides that are phosphorylated by c-Src. We have demonstrated that SILAC-based quantitative proteomics approach is suitable for identification of substrates of nonreceptor tyrosine kinases and can be coupled with peptide microarrays for high-throughput identification of substrate phosphopeptides.

Glycosylation of asparagine residues in Asn-X-Ser/Thr motifs in proteins commonly occur in the lumen of the endoplasmic reticulum (ER). Glucosidase I catalyzes the first step in the N-linked oligosaccharide processing pathway. It specifically removes the distal alpha 1,2-linked glucose residue from the Glc3-Man9-GlcNAc2 oligosaccharide precursor. Glucosidase I contains a short cytosolic tail, a single pass transmembrane domain and a large C-terminal catalytic domain located on the luminal side of the ER. Mutations in the gene encoding Glucosidase I result in the congenital disorder glycosylation (CDG-IIb), which is characterized by generalized hypotonia, dysmorphic features, hepatomegaly, hypoventilation, feeding problems, seizures and death. Two point mutations in the Glucosidase I gene have been identified and result in amino acid substitutions, namely Arg486Thr and Phe652Leu, that affect polypeptide folding and active site formation.

CCDC42 (coiled-coil domain containing 42) is a 316 amino acid protein encoded by a gene that maps to human chromosome 17p13.1. Encoding over 1,200 genes, chromosome 17 comprises over 2.5% of the human genome. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome.

Localized to the nucleus, ZNF206 is thought to play a role in transcriptional regulation events. The gene encoding ZNF206 maps to human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The GAN gene is located on chromosome 16 and, with mutation, may lead to giant axonal neuropathy, a nervous system disorder characterized by increasing malfunction with growth. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, which is a gastrointestinal inflammatory condition.

Rab proteins, a family of Ras-related small GTP-binding proteins, play a key role in regulating intracellular vesicle trafficking (1?). Rab GDP dissociation inhibitor (Rab GDI or GDI2) forms a soluble complex with Rab proteins and thereby prevents the exchange of GDP for GTP (1?). In mammals, there exist two major isoforms, Rab GDI alpha (also known as XAP-4) and Rab GDI beta (1,4). While the mammalian Rab GDI beta-genes are ubiquitously expressed, the Rab GDI ?genes are predominantly brain-specific (1). Since it is expressed predominantly in neural and sensory tissues, Rab GDI a may serve a specific function in neural signal transmission (5). The gene sequences for the Rab GDI proteins are extremely conserved in evolution, with substantial homology preserved across three eukaryotic kingdoms (5).
