Carbonic anhydrase is a family of metalloenzymes (enzymes that contain one or more metal atoms as a functional component of the enzyme) that catalyze the rapid interconversion of carbon dioxide and water into carbonic acid and protons. It also converts bicarbonate in carbon dioxide and water. This reaction occurs spontaneously in aqueous solution, but does so slowly. Carbonic anhydrase can greatly increase the rate of the reaction, with typical catalytic rates of the different forms of this enzyme ranging between 104 and 106 reactions per second. The active site of most carbonic anhydrases contains a zinc ion.

Description:
Angiopoietin-like protein 3 (Angptl3) functions as a potent lipoprotein lipase inhibitor and is an important component of plasma triglyceride homeostasis. Angptl3 also plays a role in adipose formation and angiogenesis through its interaction with integrin ?v)beta(3). It is secreted by the liver and is functionally defined by the C-terminal fibrinogen (FBN)-like domain and an N-terminal coiled-coil domain. Angptl3 regulates circulating triglyceride levels during different nutritional states thereby mediating the feeding/fasting cycle. A deficiency of Angptl3 results in abnormally low lipid levels, and a repression of the protein may be protective against atherosclerosis. Angptl3 may also play an important role in hyperlipidemia in diabetes.

Description:
The cystatin superfamily is a well-established family of cysteine protease inhibitors. All true cystatins inhibit cysteine peptidases of the papain family, such as cathepsins, while some also inhibit legumain family enzymes. The CRES (cystatin-related epididymal spermatogenic) protein defines a new subgroup in the family 2 cystatins of the cystatin superfamily. CRES proteins lack two of the three consensus sites necessary for the cystatin inhibition of C1 cysteine proteases. They are also preferentially expressed in postmeiotic germ cells, the proximal caput epididymidis, and anterior pituitary gonadotrophs. Therefore, CRES proteins may perform unique and tissue-specific functions in the reproductive and neuroendocrine systems. As a member of the CRES subfamily, Cystatin 8 is a 142 amino acid protein that is expressed in the proximal caput region of the epididymis, where it performs a specialized role during sperm development and maturation.

This protein contains a RING zinc-finger motif and has been shown to possess E3 ubiquitin ligase activity. Expression of this gene in retrovirally transduced T cell hybridoma significantly inhibits activation-induced IL2 and IL4 cytokine production. Induced expression of this gene was observed in anergic CD4(+) T cells, which suggested a role in the induction of anergic phenotype. HIBADH is a 336 amino acid mitochondrial enzyme that catalyzes the NAD+-dependent, reversible oxidization of 3-Hydroxyisobutyrate to methylmalonate semialdehyde, an intermediate of valine catabolism. The enzyme functions as a homodimer between a pH of 7.0 and 10.0, with optimal activity between 8.8 and 9.0. It was previously hypothesized that defects in the gene encoding HIBADH may be the cause of 3-Hydroxyisobutyric aciduria, a rare disorder that is characterized by a variety of clinical manifestations such as neurodevelopmental problems and dysmorphic features. However, it was shown that HIBADH activity was equal in patients with 3-Hydroxyisobutyric aciduria as compared with controls.

Epidermal growth factor (EGF) is an acid- and heat-stable 53 amino acid protein originally found in rodents and humans. It has been shown to be a potent mitogen for a variety of cell types both in vivo and in vitro. EGF binds to the EGF receptor on the surface of cells and mediates intrinsic phosphorylation of the receptor on tyrosine residues. It has been detected in nearly all body fluids, such as urine (urogastrone), saliva, milk and platelet-rich plasma. EGF, TGF?and vaccinia virus growth factor exhibit 30-40% amino acid homology. Several additional members of the EGF/TGF family have been described; these include Cripto, Amphiregulin and the heparin-binding EGF-like growth factor. Amphiregulin and the heparin-binding EGF-like growth factor both bind to the EGF receptor.
