Description:
Recognizes a protein of ~76 kDa, which is identified as Nucleolin (NCL). It is the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11kb. It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly.This MAb can be used to stain the nucleoli in cell or tissue preparations and can be used as a marker of the nucleoli in subcellular fractions. It produces a speckled pattern in the nuclei of cells of normal and malignant cells and may be used to stain the nucleoli of cells in fixed or frozen tissue sections. It can be used with paraformaldehyde fixed frozen tissue or cell preparations and formalin fixed, paraffin-embedded tissue sections.

MAb VU-2G7 reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. The dominant epitope of VU-2G7 has not been established with epitope fingerprinting. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion.

This product is prepared by labeling highly cross-adsorbed donkey anti-rat IgG (H+L) with CF™405S. CF™405S is a blue fluorescent dye with an absorption peak wavelength that nearly coincides with the 405 nm blue diode laser line. In addition, the emission peak wavelength of the dye well centers within the blue detection window of BD flow cytometers. As a result, flow cytometry analysis using CF™405S dye results in much brighter signal than that using Alexa Fluor® 405. To minimize cross-reactivity, the antibody has been adsorbed against bovine, chicken, goat, guinea pig, Syrian hamster, horse, human, mouse, rabbit, and sheep serum proteins .

APC belongs to the family of phycobiliproteins, which are derived from cyanobacteria and eukaryotic algae. APC has absorption maxima at 650 and emission at 660 nm. Because of the high fluorescence quantum yield (~0.68), APC is by far the brightest dye for the 650 nm wavelength region.

Recognizes a 100 kDa glycoprotein, identified as CD10, also known as Common Acute Lymphocytic Leukemia Antigen (CALLA). It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). It is also expressed on the surface of normal early lymphoid progenitor cells, immature B cells within adult bone marrow and germinal center B cells within lymphoid tissue. CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, with especially high expression on the brush border of kidney and gut epithelial cells.

The antibody HEB-29 reacts with human blood group B. The specificity of the antibody HEB-29 was confirmed by comparison of specificity and reactivity to standard reagent using >5.000 samples of blood. MAb HEB-29 shows specific staining of erythrocytes and vascular epithelium of blood group B controls and no staining in group A controls. It is applicable for tissue staining in tumor patients with blood groups B and AB. Blood group antigens are generally defined as molecules formed by sequential addition of saccharides to the carbohydrate side chains of lipids and proteins detected on erythrocytes and certain epithelial cells. The A, B and H antigens are reported to undergo modulation during malignant cellular transformation. Blood group related antigens represent a group of carbohydrate determinants carried on both glycolipids and glycoproteins. They are usually mucin type, and are detected on erythrocytes, certain epithelial cells, and in secretions of certain individuals. Sixteen genetically and biosynthetically distinct but inter related specificities belong to this group of antigens, including A, B, H, Lewis A, Lewis B, Lewis X, Lewis Y, and precursor type 1 chain antigens.

MITF has been shown to be phosphorylated by MAP kinase in response to c-kit activation, resulting in upregulation of MITF transcriptional activity. Mutations of the MITF gene are associated with the autosomal dominant hereditary deafness and pigmentation condition, Waardenburg Syndrome type 2A. Multiple isoforms of MITF exist, including MITF-A, MITF-B, MITF-C, MITF-H, and MITF-M, which differ in the amino-terminal domain and in their expression patterns. The MITF-M isoform is restricted to the melanocyte cell lineage. Anti-MITF, D5, recognizes a nuclear protein, which is expressed in the majority of primary and metastatic epithelioid malignant melanomas as well as in normal melanocytes, benign nevi and dysplastic nevi.

Recognizes a protein of 12 kDa, identified as β-2 microglobulin. Major histocompatibility complex (MHC) class 1 molecules bind to antigens for presentation on the surface of cells. The proteasome is responsible for producing these antigens from the components of foreign pathogens. MHC class 1 molecules consist of an α heavy chain that contains three subdomains (α1, α2, α3) and a non-covalent associating light chain, known as β-2-Microglobulin. β-2-Microglobulin associates with the α3 subdomain of the α heavy chain and forms an immunoglobulin domain-like structure that mediates proper folding and expression of MHC class 1 molecules. The α1 and α2 domains of the α heavy chain form the peptide antigen-binding cleft. Mutations in the β-2-Microglobulin gene can enhance the progression of malignant melanoma phenotypes.

This MAb recognizes a protein of 145 kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. Macrophages usually arise from hematopoietic stem cells in the bone marrow. Under migration into tissues, the monocytes undergo further differentiation to become multifunctional tissue macrophages. They are classified into normal and inflammatory macrophages. Normal macrophages include macrophages in connective tissue (histiocytes), liver (Kupffer's cells), lung (alveolar macrophages), lymph nodes (free and fixed macrophages), spleen (free and fixed macrophages), bone marrow (fixed macrophages), serous fluids (pleural and peritoneal macrophages), skin (histiocytes, Langerhans's cell) and in other tissues. Inflammatory macrophages are present in various exudates. Macrophages are part of the innate immune system, recognizing, engulfing and destroying many potential pathogens including bacteria, pathogenic protozoa, fungi and helminthes. This MAb reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and has been shown as an important marker for identifying macrophages in tissue sections.
