This MAb recognizes an epitope within the 74-182 C-terminal sequence (11kD peptide fragment) of human serum Cellular Retinol Binding Protein 1 (CRBP 1), a single-chain glycoprotein belonging to the superfamily of hydrophobic molecule transporter proteins, which is responsible for transport of retinol (vitamin A1) from the liver to peripheral target tissues, like the eye, where it mediates the cellular uptake. CRBP 1 is synthesized by hepatic parenchymal cells where it becomes bound to its ligand retinol and is then released into the circulation, where it binds further to the protein transthyretin, to form a transporting complex, which is big enough not to be lost by filtration through the kidney glomeruli. It is detected in nearly all tissues with higher expression in adult ovary, pancreas, pituitary gland, adrenal gland, and fetal liver.

Antibody to fascin-1 is a very sensitive marker for Reed-Sternberg cells and variants in nodular sclerosis, mixed cellularity, and lymphocyte depletion Hodgkin's disease. It is uniformly negative in lymphoid cells, plasma cells, and myeloid cells. Fascin-1 is also expressed in dendritic cells. This marker may be helpful to distinguish between Hodgkin lymphoma and non-Hodgkin lymphoma in difficult cases. Also, the lack of expression of fascin-1 in the neoplastic follicles in follicular lymphoma may be helpful in distinguishing these lymphomas from reactive follicular hyperplasia in which the number of follicular dendritic cells is normal or increased. Antibody to fascin-1 has been suggested as a prognostic marker in neuroendocrine neoplasms of the lung as well as in ovarian cancer. Fascin-1 expression may be induced by Epstein-Barr virus (EBV) infection of B cells with the possibility that viral induction of fascin in lymphoid or other cell types must also be considered in EBV-positive cases.

Recognizes a sialoglycoprotein of 39 kDa, identified as glycophorin A (GPA). It is present on red blood cells (RBC) and erythroid precursor cells. It has been shown that glycophorin acts as the receptor for Sandei virus and parvovirus. Glycophorins A (GPA) and B (GPB), which are single, trans-membrane sialoglycoproteins. GPA is the carrier of blood group M and N specificities, while GPB accounts for S and U specificities. GPA and GPB provide the cells with a large mucin like surface and it has been suggested this provides a barrier to cell fusion, so minimizing aggregation between red blood cells in the circulation.

Recognizes a 60 kDa protein, identified as the heat shock protein 60 (hsp60). Its epitope is localized between aa 383-419 of human hsp60. A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp), more appropriately called as stress response proteins (srp). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone LK1, unlike LK2, recognizes only the mammalian (not bacterial) hsp60 and is useful in distinguishing hsp60 from mammals and bacteria.

Recognizes a protein of 12 kDa, identified as β-microglobulin. β-microglobulin non-covalently associates with the 44 kDa chain to form the HLA Class I antigen complex. Human β-2 microglobulin associated with HLA Class I antigens is expressed on many types of cells including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial cells, and epithelial cells. It is absent on erythrocytes. This MAb is specific to human β-2 microglobulin and does not react with non-human primate cells. This antibody reacts with all cell types excluding erythrocytes. The detection of β-2 microglobulin in body fluids has been used as a tumor marker and for monitoring patients with HIV infection.

This antibody recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures, all cells that have left the basal layer express it. Involucrin expression is altered in pathological conditions: in psoriasis and other benign epidermal hyperplasias, involucrin expression begins closer to the basal layer than normal; expression is abnormal in squamous cell carcinomas and premalignant lesions, and is reduced in severe dysplasias of the larynx and cervix.

This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.

This MAb reacts with the HLA-DR antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts.

CD309, also known as VEGFR2, KDR3, and Flk-1 (mouse), is a type I transmembrane glycoprotein. It is a member of the CSF-1/PDGF receptor family of type III tyrosine kinase receptors. Human VEGFR2 is mainly expressed by endothelial cells, embryonic tissues, and megakaryocytes. It plays an important role in the regulation of angiogenesis, vasculogenesis, and vascular permeability. The ligands of VEGFR2 include VEGF-A, VEGF-C, VEGF-D, and VEGF splice isoforms. Ligation of VEGFR2 with its ligands results in the receptor dimerization and auto-phosphorylation, stimulating endothelial cell proliferation and migration.

This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb reacts with nonspecific cross-reacting antigen (NCA) and shows a cross-reaction with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 β chain (CD8 α/β) or as a homodimer (CD8 α/α). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.
