HS1 which is hematopoietic lineage cell-specific protein 1, is a substrate of protein tyrosine kinases in lymphocytes, it binds to F-actin, and promotes Arp2/3 complex-mediated actin polymerization. However, the mechanism for the interaction between HS1 and F-actin has not yet been fully characterized. HS1 contains 3.5 tandem repeats, a coiled-coil region, and an SH3 domain at the C terminus. Unlike cortactin, which is closely related to HS1 and requires absolutely the repeat domain for F-actin binding, an HS1 mutant with deletion of the repeat domain maintains a significant F-actin binding activity. Deletion of the coiled-coil region abolished the ability of HS1 to bind to actin filaments and to activate the Arp2/3 complex for actin nucleation and actin branching.

XAF-1 Antibody: XAF-1 binds to XIAP, an inhibitor of caspases-3, -7, and -9, and triggers its relocation from the cytosol to the nucleus. Overexpression of XAF-1 results in the neutralization of XIAP's ability to inhibit cell death. XAF-1 is normally expressed in all adult and fetal tissues but was found to be present in very low levels in a variety of cancer cell lines. In contrast, XIAP levels have been shown to be high in a majority of cell lines. Low XAF-1 and high basal expression of XIAP may therefore play a critical role in maintaining survival of cancer cell lines. Both IFN-alpha2 and IFN-beta can induce XAF-1 mRNA in all cells examined but induction of XAF-1 protein (as observed by immunoblot analysis) was seen only in cell lines sensitive to the apoptotic effects of IFNs.

BAG-1 Antibody: Bcl-2-associated athanogene 1 (BAG-1) was first identified as an anti-apoptotic bcl-2-binding protein. Later it was found to bind the molecular chaperones Hsp70 and Hsc70 through its carboxy-terminal sequence (termed the Bag domain), resulting in the inhibition of the refolding activity of these chaperones. It is thought that by binding and inhibiting these molecular chaperones, BAG-1is able to modulate the expression level of proteins requiring chaperones to fold correctly. One such group of proteins that are affected is glucocorticoid receptors. Other reports have suggested that the level of BAG-1 expression correlates with the aggressiveness of various cancers. Multiple isoforms of BAG-1 are known to exist.
