This antibody recognises a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45) called CD45RO (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. CD45RO is found on mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. The CD45RO antibody shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, antibody to CD45RO is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.

Description:
This antibody is specific for a 53 kDa protein, which is identified as p53 suppressor gene product. The large number of prolines in its amino acid sequence causes p53 to migrate slowly in SDS-PAGE, resulting in the amino acid content-estimated 43 kDa protein appearing larger than expected. DO-7 reacts with the mutant as well as the wild form of p53 under denaturing and non-denaturing conditions. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intra-tubular germ cell neoplasia.

Fibroblast growth factor-1 (FGF-1), also designated acidic FGF, and fibroblast growth factor-2 (FGF-2), also designated basic FGF, are members of a family of growth factors that stimulate proliferation of cells of mesenchymal, epithelial and neuroectodermal origin. Its primary function is platelet adhesion, binding to Factor VIII, collagen and platelets, to coagulate blood at the site of wounding. The enzyme VWFCP, or vWF-cleaving protease, facilitates clotting by cutting the protein into subunits, increasing its binding capacity. <p>Deficiency or dysfunction of the protein increases the tendency of wounds to bleed, or to bleed more. Over 300 gene mutations have been identified and classified into three types. Type 1 von Willebrand Factor disease is characterised by reduced amounts in the bloodstream, Type 2 by reduced binding ability and Type 3 by a nonfunctional protein.

This antibody is specific to SUMO1 and shows no cross-reaction with either SUMO2 or SUMO3. The small ubiquitin-related modifier (SUMO) proteins, which include SUMO1, 2 and 3, belong to the ubiquitin-like protein family. Like ubiquitin, the SUMO proteins are synthesised as precursor proteins that undergo processing before conjugation to target proteins. Also, both utilize the E1, E2, and E3 cascade enzymes for conjugation. However, SUMO and ubiquitin differ with respect to targeting. Ubiquitination predominantly targets proteins for degradation, whereas sumoylation targets proteins to a variety of cellular processing, including nuclear transport, transcriptional regulation, apoptosis and protein stability. The unconjugated SUMO1 protein localizes to the nuclear membrane.
