Crystallins are the major proteins of the vertebrate eye lens, where they maintain the transparency and refractive index of the lens. Crystallins are divided into alpha, beta, and gamma families, and the beta and gamma-crystallins also comprise a superfamily. Crystallins usually contain seven distinctive protein regions, including four homologous motifs, a connecting peptide, and N- and C-terminal extensions. gamma-crystallins are structural proteins in the lens, and they exists as monomers which typically lack connecting peptides and terminal extensions. The gamma-crystallins include seven closely related gamma A, gamma B, gamma C, gamma D, gamma E, gamma F, and gamma G-crystallin, as well as the gamma N and gamma S-crystallin genes. The gamma-crystallins are differentially regulated after early development, and are involved in cataract formation as a result of either age-related protein degradation or genetic mutation.

Cytokeratin 10 is a heterotetramer of two type I and two type II keratins. Cytokeratin 10 is generally associated with keratin 1. It is seen in all suprabasal cell layers including stratum corneum. A number of alleles are known that mainly differ in the Gly-rich region (positions 490-560). Defects in cytokeratin 10 are a cause of epidermolytic hyperkeratosis (EHK), also known as bullous congenital ichthyosiform erythroderma (BCIE) or bullous erythroderma ichthyosiformis congenita of Brocq. EHK is an hereditary skin disorder characterized by blistering and a marked thickening of the stratum corneum. At birth, affected individuals usually present with redness, blisters and superficial erosions due to cytolysis. Within a few weeks, the erythroderma and blister formation diminish and hyperkeratoses develop. Transmission is autosomal dominant, but most cases are sporadic. Defects in cytokeratin 10 are also a cause of annular epidermolytic ichthyosis (AEI), also known as cyclic ichthyosis with epidermolytic hyperkeratosis. AEI resembles clinical and histologic features of both epidermolytic hyperkeratosis and ichthyosis bullosa of Siemens.

RUSC1 is a 902 amino acid protein that contains a RUN domain and a SH3 domain. RUSC1’s RUN domain is necessary for NGF induced nuclear redistribution. RUSC1 is a putative signaling adapter which may play a role in neuronal differentiation. RUSC1 seems to be involved in signaling pathways that are regulated by the prolonged activation of MAPK. RUSC2 (RUN and SH3 domain containing 2), also known as Iporin, is a 1,516 amino acid cytoplasmic protein that is widely expressed, with highest levels in brain and testis. The RUN domain of RUSC2 is required for interaction with Rab 1A, Rab 1B and GM130. It is thought that RUSC2 may possibly function as a connector between endoplasmic reticulum (ER) derived vesicle targets triggered by the Rab 1 GTPases and a signaling pathway regulated by molecules containing SH3 and/or poly-proline regions.
