Interleukin-22 (IL-22), also known as IL-10 related T cell derived inducible factor (ILTIF) was initially identified as a gene induced by IL-9 in mouse T cells and mast cells. IL-22 has been shown to activate STAT1 and STAT3 in several hepatoma cell lines and upregulate the production of acute phase proteins. IL-22 is produced by normal T cells upon anti-CD3 stimulation in humans. Mouse IL-22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL-22 may be involved in inflammatory responses. The functional IL-22 receptor complex consists of two receptor subunits, IL-22R(CRF29) and IL-10Rbeta(CRF24), belonging to the class II cytokine receptor family.

Lymphocyte cytosolic protein 2（LCP2）contains a SAM domain and a SH2 domain. It is highly expressed in spleen, thymus and peripheral blood leukocytes, T-cell and monocytic cell lines, but expressed at lower level in B-cell lines. LCP2 was originally identified as a substrate of the ZAP-70 protein tyrosine kinase following T cell receptor (TCR) ligation in the leukemic T cell line Jurkat. It is phosphorylated after T-cell receptor activation by ZAP70, ITK and TXK, which leads to the up-regulation of Th1 preferred cytokine IL-2 during post-translational modification. Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

T cell immunoreceptor with Ig and ITIM domains (TIGIT), also called VSIG9 and Vstm3, is a member of the CD28 family within the Ig superfamily of proteins. TIGIT contains an immunoglobulin variable domain, a transmembrane domain and an immunoreceptor tyrosine-based inhibitory motif (ITIM), and is expressed on regulatory, memory, activated T cells and NK cells. TIGIT binds to CD155(PVR) that appear on dendritic cells (DC), macrophages and endothelium with high affinity, and CD112(PVRL2) with lower affinity, but not CD113 (PVRL3). TIGIT-Fc fusion protein could interact with PVR on DC and enhance the secretion of IL-10, but inhibit the macrophage activation. Mice lacking TIGIT show increased T cell responses and susceptibility to autoimmune challenges, while knockdown of TIGIT with siRNA in human memory T cells did not affect T cell responses.

Complement 3 (C3) is the most abundant component of the complement cascade and the convergent point for all three major complement activation pathways: namely classical, alternative and mannose-binding lectin pathways.
