This antibody reacts with an extracellular domain (close to transmembrane) of NCAM (Neural cell adhesion molecule), also called CD56. Three isoforms of NCAM/CD56 are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. NCAM antibody recognises two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). NCAM antibody plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.

Mouse interleukin-16(IL-16) is a single chain non-glycosylated polypeptide. IL-16 is widely expressed in human tissues including spleen, thymus, lymph nodes, peripheral leukocytes, bone marrow and cerebellum. IL-16 plays an important role instimulating a migratory response in CD4+ lymphocytes, monocytes, and eosinophils,inducing T-lymphocyte expression of interleukin 2 receptor.It was originally identified as a CD8+ T cell-derived chemoattractant for CD4+ cells. In addition to its chemotactic properties, IL-16 has also been shown to suppress HIV-1 replication in vitro and appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. It may act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.

ALKBH2 Antibody: The E. coli AlkB protein protects against the cytotoxicity of methylating agents by repair of the specific DNA lesions generated in single-stranded DNA; ALKBH2 and ALKBH3 are mammalian homologs of AlkB that catalyze the removal of 1-methyladenine and 3-methylcytosine, modifications that left unchecked could lead to cancerous cells. Mutations in both ALKBH2 and ALKBH3 have been observed in pediatric brain tumors indicating that these proteins are important in the prevention of cancer formation. Like the histone demethylase JMJD1A, ALKBH2 is a non-heme iron enzyme that is inhibited by Nickel ions, suggesting that inhibition of ALKBH2 by Nickel ions may play a role in the development of cancer. Conversely, ALKBH2 mRNA and protein levels are increased glioma cells following Photofrin-mediated photodynamic therapy, an adjuvant therapy in cancer treatment, suggesting that down-regulating ALKBH2 expression in cancer cells may enhance the anti-cancer effectiveness of this treatment.

Ephrin Type-A Receptor 7 (EPHA7) is a single-pass type I membrane protein which belongs to the Eph family of transmembrane receptor tyrosine kinases. It contains two fibronectin type-III domains, one protein kinase domain and one SAM (sterile alpha motif) domain. EPHA7 is a receptor for members of the ephrin-A family. Eph receptors are largely expressed throughout the ectoderm, mesoderm, and endoderm of vertebrate embryos. EPHA7 functions as a repulsive guidance molecule during the targeting of retinal axons to the superior colliculus and of neocortical axons to the thalamus. EPHA7 is expressed at a substantial level in most human lung cancers. The high expression of EPHA7 protein may participate in the malignancy transformation, invasion progression and metastasis of primary hepatocellular carcinoma. EPHA7 may involve in smoking related lung carcinogenesis.

C-Type Lectin Domain Family 10 Member A (CLEC10A) is a type II transmembrane C-type lectin that is expressed on immature myleloid dendritic cells and alternatively activated (tolerogenic) macrophages. CLEC10A/MGL binds and internalizes molecules with terminal nonsialylated GalNAc carbohydrates such as the Tn carcinoma antigen. CLEC10A/MGL also binds the GP envelope glycoprotein on Marburg and Ebola viruses and enhances viral entry and infectivity. It constitute a unique class of C-type lectins because of their specificity for galactose and its structural homologues. CLEC10A is thought to participate in the recognition of molecules from both altered self and pathogens due to its monosaccharide specificity for Gal and N-acetylgalactosamine (GalNAc). Human and rat carry a single gene for CLEC10A/MGL, while mouse has two closely related MGL1 and MGL2 genes.

Human serum albumin (HSA), the most abundant protein in human blood plasma, is essential for maintaining osmotic pressure. It is produced in the liver, consists of a single polypeptide chain, with 585 amino acids with 17 tyrosil residues and one tryptophan located in position 214. HSA is organised in three domains, I, II and III, each consisting of two subdomains, A and B. In the physiological states, HSA occurs in two forms – the non-modified HSA with a free thiol group of Cys-34, and the modified (oxidised) form (oHSA), also called human mercaptoalbumin (HMA) and human nonmercaptoalbumin (HNA), respectively. HMA and HNA are in equilibrium, depending on the redox state of Cys-34, and their ratio also varies depending on age and the diseased state. HSA functions primarily as a carrier protein for drugs, steroids, fatty acids, and thyroid hormones, and plays a role in stabilising extracellular fluid volume.

RNAse H2A Antibody: Ribonucleases (RNAses) H are enzymes that hydrolyze the RNA strands of RNA/DNA hybrids. The major role of these enzymes is to remove the RNA strand from the RNA/DNA hybrids that form during DNA replication and repair. RNAse H2 is made up of three subunits; all three are required for RNAse activity. Recent evidence has demonstrated that mutations in RNAse H2A or any of the other subunits result in Aicardi-Goutieres syndrome (AGS), a neurological disorder with similar symptoms to viral brain infections including high levels of IFN-alpha in the cerebral spinal fluid. Similar conditions are observed with mutations in TREX1, a single-stranded DNA exonuclease, suggesting that RNAse H2 and TREX1 may have similar roles, and that mutations in any of these genes lead to an accumulation of intracellular nucleic acids, triggering an inflammatory response through activation of the innate immune system.

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. C-Type Lectin Domain Family 4 Member E (CLEC4E) is a 219 amino acid single-pass type II membrane protein that contains one C-type Lectin domain. It is expressed in monocytes, CLEC4E functions as a downstream target of C/EBP beta and is thought to play a role in the inflammatory response, possibly via transcriptional control of C/EBP beta . CLEC4E may play a role in the response to inflammatory stimuli in peritoneal macrophages and may be involved in immune surveillance processes under transcriptional control of CEBPB. Human CLEC4E shares 67% sequence identity with its mouse counterpart, suggesting a similar function between species. CLEC-4E exists as multiple alternatively spliced isoforms that are encoded by a gene which maps to a natural killer gene complex region on human chromosome 12.
