Description:
SMN1 localizes to both the cytoplasm and the nucleus. Within the nucleus, the protein localizes to subnuclear bodies called gems which are found near coiled bodies containing high concentrations of small ribonucleoproteins (snRNPs). This protein forms heteromeric complexes with proteins such as SIP1 and GEMIN4, and also interacts with several proteins known to be involved in the biogenesis of snRNPs, such as hnRNP U protein and the small nucleolar RNA binding protein.This gene is part of a 500 kb inverted duplication on chromosome 5q13. This duplicated region contains at least four genes and repetitive elements which make it prone to rearrangements and deletions. The repetitiveness and complexity of the sequence have also caused difficulty in determining the organization of this genomic region. The telomeric and centromeric copies of this gene are nearly identical and encode the same protein. However, mutations in this gene, the telomeric copy, are associated with spinal muscular atrophy; mutations in the centromeric copy do not lead to disease. The centromeric copy may be a modifier of disease caused by mutation in the telomeric copy. The critical sequence difference between the two genes is a single nucleotide in exon 7 which is thought to be an exon splice enhancer. It is thought that gene conversion events may involve the two genes, leading to varying copy numbers of each gene. The protein encoded by this gene localizes to both the cytoplasm and the nucleus. ORAI1 Monoclonal Antibody: Antigen stimulation of immune cells triggers Ca++ entry through Ca++ release-activated Ca++ (CRAC) channels. ORAI1 is a recently identified four-transmembrane spanning protein that is an essential component of CRAC. A missense mutation in this protein in humans is the cause of one fo rm of hereditary severe combined immune deficiency (SCID) which results in ablated T-cell Ca++ entry. It has been suggested that ORAI1 functions as a highly selective Ca++ plasma membrane channel that is gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca++ sensor. ORAI1 often migrates at a higher than expected molecular weight in SDS-PAGE. This antibody is predicted to have no cross-reactivity to ORAI2 or ORAI3.

Fatty acid-binding protein 2 (FABP2), is also known as Fatty acid-binding protein, intestinal (FABPI), Intestinal-type fatty acid-binding protein (I-FABP). FABP2 belongs to the calycin superfamily and Fatty-acid binding protein (FABP) family. FABP2 / FABPI is expressed in the small intestine and at much lower levels in the large intestine and is highest expressed in the jejunum. FABP are thought to play a role in the intracellular transport of long-chain fatty acids and their acyl-CoA esters. FABP2 is probably involved in triglyceride-rich lipoprotein synthesis. FABP2 binds saturated long-chain fatty acids with a high affinity, but binds with a lower affinity to unsaturated long-chain fatty acids. FABP2 may also help maintain energy homeostasis by functioning as a lipid sensor.

NALP2 Antibody: NALP2 belongs to a family of cytoplasmic proteins that have been implicated in cell responses to apoptotic and inflammatory stimuli. Unlike the prototypical NALP protein NALP1, NALP2 only contains a NACHT domain, leucine rich repeat (LRR), and pyrin-containing domain (PYD). This protein interacts with the adapter protein ASC in addition to CARD8 and caspase-1 to form an inflammasome with high proIL-1 beta -processing activity and is thought to function as a modulator of NF-kappa B and procaspase-1 activation in macrophages. It has also been suggested that NALP2, in addition to other NALP family members, can act as innate sensors of pathogens in a manner similar to the toll-like receptors (TLRs). At least two alternatively spliced transcript variants encoding distinct isoforms have been found for NALP2.

STIM2 Antibody: In T lymphocytes, the sole pathway for Ca++ entry following antigen-receptor binding is through store-operated Ca++-release-activated Ca++ (CRAC) channels. These channels are made up of the pore-forming subunit ORAI1 and the stromal interaction molecule 1 (STIM1), a protein that functions as a Ca++ sensor and activates the CRAC channels, migrating to the plasma membrane from endoplasmic reticulum (ER)-like sites which act as the Ca++ store. A related molecule, STIM2, acts to inhibit the STIM1-mediated store-operated Ca++ entry, and can form complexes with STIM1, suggesting they may play a coordinated role in controlling Ca++ entry. At least three isoforms of STIM2 are known to exist. This STIM2 antibody is predicted to have no cross-reactivity to STIM1.

LXR-A Antibody: LXR-A belongs to the Liver X Receptor family that encodes highly homologous transcription factors. Like the highly homologous LXR-B, LXR-A forms heterodimers with the retinoic acid receptor RXRalpha, function as sensors for cellular oxysterols which when activated, increase the expression of genes that control sterol and fatty acid metabolism and homeostasis. Recent experiments have indicated that the LXRs can also modulate both innate and adaptive immune responses. Human and mouse tumors produce LXR ligands that inhibit CCR7 expression on maturing dendritic cells (DCs), thereby allowing tumor immunoescape. In mouse models, it was shown that ablating LXR-A signaling led to an immune-mediated strong inhibition of tumor growth, suggesting that manipulation of this pathway may be a viable anti-cancer approach.
