P2RX5 is the receptor for ATP that acts as a ligand gated ion channel.The product of this gene belongs to the family of purinoceptors for ATP. This receptor functions as a ligand-gated ion channel. Several characteristic motifs of ATP-gated channels are present in its primary structure, but, unlike other members of the purinoceptors family, this receptor has only a single transmembrane domain. Three transcript variants encoding distinct isoforms have been identified for this gene.

Les étudiants se familiarisent avec la solubilité des protéines et apprennent comment elle est affectée par des paramètres variés, tels que la température, le pH, le sel et la constante diélectrique. Ils assimilent la précipitation des protéines due au pH, à une grande concentration de sel et en présence de solvants organiques, et la dénaturation des protéines résultant d'une température élevée. De plus, le kit démontre dans quelle mesure les non-protéines, comme les détergents, modifient considérablement les propriétés physiques des molécules de protéines, ce qui explique l'importance de ces détergents dans la solubilisation des protéines. Cette activité de laboratoire passe par l'analyse de trois types de protéines pures. Les étudiants modifient ensuite certaines de ces propriétés avec un détergent et réexaminent les propriétés physiques de ces protéines. Ils doivent alors observer comment les propriétés physiques des molécules de protéines peuvent être exploitées pour la purification et la caractérisation des protéines, et appliquer leurs résultats à un échantillon test d'extrait de tissu complexe.

Killer cell immunoglobulin-like receptor 2DL4(KIR2DL4) is a Single-pass type I membrane protein and contains 2 Ig-like C2-type (immunoglobulin-like) domains.It belongs to the immunoglobulin superfamily. KIR2DL4 is expressed in all NK cells and some T cells. KIR2DL4 activates the cytotoxicity of NK cells, despite the presence of an immunoreceptor tyrosine-based inhibition motif (ITIM) in its cytoplasmic tail. The ITIM was not necessary for activation of lysis by KIR2DL4. The activation signal of KIR2DL4 was sensitive to inhibition by another ITIM-containing receptor. The activation-deficient mutant of KIR2DL4 inhibited the signal delivered by the activating receptor CD16.

Junctional Adhesion Molecule A (JAM-A) is a single-pass type I membrane protein that belongs to the immunoglobulin superfamily. JAM-A contains 2 Ig-like V-type (immunoglobulin-like) domains and Interacts with the ninth PDZ domain. JAM-A is localized to the tight junctions of both epithelial and endothelial cells. JAM-A seems to be involved in epithelial tight junction formation. JAM-A appears early in primordial forms of cell junctions and recruits PARD3. The association of the PARD6-PARD3 complex may prevent the interaction of PARD3 with JAM-A, thereby preventing tight junction assembly. JAM-A plays a role in regulating monocyte transmigration involved in regulating integrity of the epithelial barrier. In the case of orthoreovirus infection, JAM-A serves as receptor for the virus.

Amphoterin-Induced Protein 2 (AMIGO2) is a single-pass type I membrane protein which belongs to the AMIGO family of immunoglobulin superfamily. Mature AMIGO2 contains an Ig-like C2-type (immunoglobulin-like) domain, 6 LRR (leucine-rich) repeats, a LRRCT domain, as well as a LRRNT domain. AMIGO2 is mainly expressed in in breast, ovary, cervix, and uterus, although lower in lung, colon, and rectum. AMIGO2 required for depolarization-dependent survival of cultured cerebellar granule neurons. AMIGO2 may mediate homophilic as well as heterophilic cell-cell interaction with AMIGO1 or AMIGO3. AMIGO2 may contribute to signal transduction through its intracellular domain, and may be required for tumorigenesis of a subset of gastric adenocarcinomas.

Fractalkine(CX3CL1) is a single-pass type I membrane protein and belongs to the intercrine delta family. It consists of an extracellular NH2-terminal domain, a mucin-like stalk, a transmembrane alpha helix, and a short cytoplasmic tail. CX3CL1 exists in two forms: as a membrane-anchored or as a shed 80-95K glycoprotein. Soluble CX3CL1 is generated by limited proteolysis on the cell surface, and a disintegrin and metallopeptidase 10 (ADAM10) and ADAM17/tumour necrosis factor- alpha-converting enzyme (ADAM17/TACE) participate in this shedding. It has been suggested that ADAM10 acts in the constitutive shedding, and ADAM17 acts in response to cell activation. The protein may play a role in regulating leukocyte adhesion and migration processes at the endothelium.

The Rh blood group includes this gene which encodes both the RhC and RhE antigens on a single polypeptide and a second gene which encodes the RhD protein. The classification of Rh-positive and Rh-negative individuals is determined by the presence or absence of the highly immunogenic RhD protein on the surface of erythrocytes. The tetrameric receptor complex for IFN gamma consists of two subunits, IFNGR1 (IFN gamma R alpha) and IFNGR2 (IFN gamma R beta ), through which the dimeric IFN- gamma exerts its biological functions, including antiviral, antiproliferation and immune-modulatory activity in mammals. Both IFNGR1 and IFNGR2 are single transmembrane proteins belonging to the class II cytokine family. FNGR1, widely expressed in most host cells, is essential for IFN gamma binding, receptor trafficking, and signal transduction. IFNGR1 possesses an intracellular Janus tyrosine kinase (JAK) 1 binding site, a signal transducer and activator of transcription 1 (STAT1) binding site. The resulting STAT1 homodimers translocate from the cytoplasm to the nucleus and bind to the interferon-gamma activated sequence (GAS) promoter to induce expression of downstream interferon stimulated genes (ISGs).

Dectin-1 and dectin-2 are type II transmembrane proteins of the C-type lectin family. They contain single carbohydrate recognition domains (CRDs) in their extracellular region. Dectin-1 and dectin-2 are expressed in macrophages and dendritic cells. Dectin-1 recognises beta-glucans with its CRD and signals through its immunoreceptor tyrosine-based activation motif (ITAM)-like motif in the cytoplasmic domain, whereas dectin-2 recognises alpha-mannans and transduces its signal through association with the ITAM-containing Fc receptor gamma-chain.
