The PC61.5 antibody specifically reacts with mouse CD25, the 55 kDa low-affinity Interleukin-2 Receptor alpha chain (IL-2R alpha), expressed on early progenitors of T and B lineage, and on B and T cells. Together with CD122 (IL-2 Receptor beta) and CD 132 (IL-2 Receptor gamma c, the common gamma chain), CD25 forms high-affinity receptor complexes for IL-2. Resting B and T cells and natural killer cells do not express IL-2R alpha. Cd25 is also expressed on the dendritic cells, and it enhances lymphocyte differentiation and activation.The PC61.5 antibody block the binding of IL-2 to both high-affinity and low-affinity receptors.

CDIP Antibody: The p53 tumor-suppressor gene integrates numerous signals that control cell life and death; loss of its functions contributes to the development of most cancers. CDIP is a novel pro-apoptotic target gene whose inhibition abrogates p53-mediated apoptotic responses. Overexpression of CDIP induced apoptosis in transfected cells while siRNA suppression of caspase-8 mRNA blocked this CDIP-induced apoptosis, indicating that the CDIP-dependent apoptosis pathway proceeds through extrinsic cell death pathway. CDIP may thus represent a novel target for drug design to maximize p53 response and sensitize tumor cells to cancer therapy. Multiple isoforms of CDIP are known to exist.

MAPKAP1 Antibody: MAPKAP1 was initially identified as the human homolog of S. pombe SIN1. Recent evidence has shown that it identical to Mip1, a protein that interacts with MEKK2, a member of the mitogen-activated protein kinase (MAPK) intracellular signaling network. MAPKAP1 is thought to prevent MEKK2 activation and dimerization by forming a complex with the inactive and non-phosphorylated MEKK2, thereby blocking the JNK1/2, ERK1/2, p38 and ERK5 MAPKs. MAPKAP1 has also been shown to play a role in the TOR signaling process, a pathway that is involved in controlling cell growth and proliferation in response to environmental cues such as nutrients, growth factors and hormones. Experiments showed that MAPKAP1 helped to maintain the TOR/rictor assembly but not the TOR/RAPTOR complex, which allowed specific phosphorylation of Akt, a kinase that is believed to couple the growth factor-PI3K signaling pathway to the nutrient-regulated TOR signaling pathway. Multiple alternatively spliced isoforms of MAPKAP1 have been identified.

DDX3 Antibody: DDX3 contains all of the motifs of the DEAD-box family of RNA helicases, including the Asp-Glu-Ala-Asp sequence that gives the protein family its name and distinguishes it from other RNA helicases. DDX3 is localized to the X chromosome and has a highly conserved functional homolog (DBY) on the Y chromosome. DDX3 is thought to be involved in RNA splicing, RNA transport, and translation initiation. The J43.1 monoclonal antibody specifically reacts with mouse CD279, also known as PD-1 (programmed death-1), a 50 to 55 kDa glycoprotein of the Ig superfamily. The PD-1 ligands, PD-L1 (B7-H1) and PD-L2 (B7-H2) are members of the B7 family. Pd-1 contains an Immunoreceptor Tyrosine-based Inhibitory Motif (ITIM) and influences the peripheral tolerances and autoimmune diseases in mice. PD-1 is transiently expressed on CD4/CD8 thymocytes, it is upregulated in apoptotic cells, and it is expressed by activated myeloid and T and B cells. The binding of PD-1 to its ligands is blocked by the J43 antibody, which also enhances contact hypersensitivity and exacerbates acute Graft-versus-host disease, Experimental autoimmune encephalomyelitis and NOD diabetes. PD-1 seems to downregulate the immune response, as the development of splenomegaly and breakdown of peripheral tolerance in PD-1 deficient mice suggests.
