Description:
Stratifin (SFN) belongs to the 14-3-3 family of proteins that act as important regulators of intracelluar signal transduction through their ability to bind specific motifs phosphorylated on serine or threonine. There are at least seven isoforms that have been identified in mammals (beta, gamma, epsilon, sigma, zeta, tau and eta). SFN can detected in many tissues, highly expressed in stratified squamous keratinizing epithelium. SFN is indicated as an epithelial cell marker and serves as a tumor suppressor whose expression can be down regulated by methylation. In addition, SFN plays a key role in maintaining the G2 checkpoint in cells and preventing mitotic death.

Description:
IL-16 was initially identified as a chemotactic cytokine (1), but is now known to possess a wide range of activities. Later studies have more fully characterized IL-16 as an immunomodulatory cytokine that contributes to the regulatory process of CD4+ T cell recruitment and activation at sites of inflammation in association with asthma and several autoimmune diseases (2). The precursor of IL-16 (pro-IL-16) is thought to be cleaved towards the C-terminal region by Caspase-3, releasing a 20 kDa active form that binds to and signals through CD4 (3,4). Besides acting as a chemotactic cytokine, IL-16 is thought to also be involved in the regulation of T cell proliferation (5) and multiple infectious, immune-mediated, and autoimmune inflammatory disorders including irritable bowel syndrome, systemic lupus erythematosus, and neurodegenerative disorders (6). At least two isoforms of IL-16 are known to exist; the longer isoform (also known as NIL-16) is detected only in neurons of the cerebellum and hippocampus (7). This recombinant protein fragment is derrived from this longer isoform.

Reacts with a protein of ~66 kDa, identified as bovine serum albumin (BSA). It is a high affinity antibody and can be used for detection of traces of BSA. Bovine serum albumin (BSA) is an abundant plasma protein in cows that is important for maintaining osmotic pressure in blood plasma for proper distribution of body fluids between intravascular compartments and body tissues. BSA is a common buffer component for immunoglobulin type assays due to good solubility characteristics for water, Ca2+, Na+, K+, fatty acids, hormones and bilirubin. BSA makes up about half of the protein in plasma and represents the most stable and soluble protein in the plasma. It is a suitable reagent for laboratories developing immunoassays, mostly due to its availability, solubility and the numerous functional groups present for coupling. The BSA component contains several lysines that are capable of reacting with conjugation sites of linkers, making it applicable as a carrier protein for antigenic compounds.

Signal Regulatory Protein alpha (SIRP alpha) is a monomeric approximately 90 kD type I transmembrane glycoprotein. The 504 amino acid human SIRP alpha contains two Ig-like C1-type domains and one Ig-like V-type domain. SIRP alpha can express in various tissues, mainly on brain and myeloid cells, including macrophages, neutrophils, dendritic and Langerhans cells. It also can detect in neurons, smooth muscle and endothelial cells. SIRPA is an immunoglobulin-like cell surface receptor for CD47. SIRP alpha acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. SIRP alpha shows adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases such as Sjoegren syndrome, Rheumatoid arthritis (RA), Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE). BAFF has also increased levels in some lymphoid cancers.

This mAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. It recognises human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

Recognises a 67 kDa transmembrane protein which is identified as CD5. The CD5 antigen is found on 95% of thymocytes and 72% of peripheral blood lymphocytes. In lymph nodes, the main reactivity is observed in T cell areas. Anti-CD5 is a pan T-cell marker that also reacts with a range of neoplastic B-cells, e.g. chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL), mantle cell lymphoma, and a subset (~10%) of diffuse large B-cell lymphoma. CD5 aberrant expression is useful in making a diagnosis of mature T-cell neoplasms. Anti-CD5 detection is diagnostic in CLL/SLL within a panel of other B-cell markers, especially one that includes anti-CD23. Anti-CD5 is also very useful in differentiating among mature small lymphoid cell malignancies. In addition, anti-CD5 can be used in distinguishing thymic carcinoma (+) from thymoma (-). Anti-CD5 does not react with granulocytes or monocytes.
