B7H4 is a B7 family member that negatively regulates T cell immunity by inhibiting of T cell proliferation, cytokine production, and cell cycle progression. In vitro, B7H4 inhibits CD4+ and CD8+ T cell proliferation, cytokine production, and generation of alloreactive cytotoxic T-cells (CTLs). In vivo, blockade of endogenous B7-H4 by specific monoclonal antibody promotes T cell responses. B7-H4 ia an important negative regulator of innate immunity through growth inhibition of neutrophils. B7-H4 is expressed on some tumour cancer cells. The role of B7-H4 in tumour progression may be to transform precancerous cells and then protect them from immunosurveillance.

CD40 is a Type I Transmembrane Glycoprotein that belongs to the TNF Receptor Superfamily. CD40 is expressed in B cells, follicular dendritic cells, dendritic cells, activated monocytes, macrophages, endothelial cells, vascular smooth muscle cells, and several tumor cell lines. The extracellular domain of CD40 is characterized by Cysteine rich repeat regions. Interaction of CD40 with its ligand (CD40L) leads to aggregation of CD40 molecules, which in turn interact with cytoplasmic components to initiate signaling pathways. Several different TRAF proteins (adaptor proteins) have been identified to serves as mediators of the signal transduction. CD40 plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

Folate Receptor 1 (FOLR1) is also known as Folate receptor alpha, Folate Binding Protein (FBP), FOLR, and is a member of the folate receptor (FOLR) family. Members of this gene family have a high affinity for folic acid and for several reduced folic acid derivatives, and mediate delivery of 5-methyltetrahydrofolate to the interior of cells. Mature FOLR1 is an N-glycosylated protein that is anchored to the cell surface by a GPI linkage. FOLR1 is predominantly expressed on epithelial cells and is dramatically upregulated on many carcinomas. FOLR1 is internalized to the endosomal system where it dissociates from its ligand before recycling to the cell surface. A soluble form of FOLR1 can be proteolytically shed from the cell surface into the serum and breast milk. Defects in FOLR1 are the cause of neurodegeneration due to cerebral folate transport deficiency (NCFTD). NCFTD is an autosomal recessive disorder resulting from brain-specific folate deficiency early in life.

CD99 is a type I transmembrane glycoprotein and the founding member of the CD99 family of molecules. The extracellular domain of CD99 contains no identifiable motifs, its cytoplasmic region, although short, does have signal transduction capability. Cells known to express CD99 include fibroblasts, neutrophils, T cells, double positive thymocytes, CD34+ stem cells, monocytes and endothelial cells. Two types of CD99 isoforms have been classified. Native human CD99 is referred to as the long, or type I isoform. The best studied type II isoform shows an Asp-Gly substitution for the C terminal 27 amino acids. The type I and II isoforms have distinctive signal transduction pathways (FAKsrc for type I PI3K plus srcERK1/2 for type II), and mediate clearly different biological outcomes. Homophilic interaction between CD99 on the neutrophil and CD99 on the endothelial cell regulates the transendothelial migration of neutrophils during inflammation. Human CD99 has 48% aa sequence identity to mouse CD99.

Intelectin-1(ITLN1) is a secreted protein and contains 1 fibrinogen C-terminal domain. Intelectin-1 is a 40 kDa Ca-dependent galactofuranose-binding lectin that is not a C-type lectin. It is expressed on multiple cell types and appears to participate in insulin signaling and microbe recognition. The protein has no effect on basal glucose uptake but enhances insulin-stimulated glucose uptake in adipocytes. It increases AKT phosphorylation in the absence and presence of insulin and it may play a role in the defense systemagainst microorganisms. It also may specifically recognize carbohydrate chains of pathogens and bacterial components containing galactofuranosy l residues, in a calcium-dependent manner.

Programmed cell death protein 1 (PD-1) is also known as CD279 and PDCD1, is a type I membrane protein and is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 is expressed on the surface of activated T cells, B cells, macrophages, myeloid cells and a subset of thymocytes. PD-1 has two ligands, PD-L1 and PD-L2, which are members of the B7 family. PD-L1 is expressed on almost all murine tumor cell lines, including PA1 myeloma, P815 mastocytoma, and B16 melanoma upon treatment with IFN-V. PD-L2 expression is more restricted and is expressed mainly by DCs and a few tumor lines. PD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN-γ by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. In vitro, treatment of anti-CD3 stimulated T cells with PD-L1-Ig results in reduced T cell proliferation and IFN-γ secretion. Monoclonal antibodies targeting PD-1 that boost the immune system are being developed for the treatment of cancer.
