Cell Growth Regulator with EF Hand Domain Protein 1 (CGREF1) is a secreted calcium ion binding protein. CGREF1 contains two EF-hand domains and both EF-hands are required for function. Human CGREF1 is synthesized as a 301 amino acid precursor that contains a 19 amino acid signal sequence, and a 282 amino acid mature chain. CGREF1 is probably digested extracellularly by an unknown serine protease generating extremely hydrophobic bioactive peptides. CGREF1 mediates cell-cell adhesion in a calcium-dependent manner. In addition, CGREF1 is able to inhibit growth in several cell lines.

Placental growth factor is a protein that in humans is encoded by the PGF gene. It is a secreted protein and belongs to the PDGF/VEGF growth factor family. The protein is a member of the VEGF (vascular endothelial growth factor) sub-family-a key molecule in angiogenesis and vasculogenesis, in particular during embryogenesis. The main source of PGF during pregnancy is the placental trophoblast. PGF is also expressed in many other tissues, including the villous trophoblast.

Bone Marrow Stromal Antigen 2 (BST2) is a single-pass type II membrane protein that belongs to the tetherin family. BST2 is predominantly expressed in the liver, lung, heart and placenta. BST2 is involved in the sorting of secreted proteins. BST2 is a human cellular protein which inhibits retrovirus infection by preventing the diffusion of virus particles after budding from infected cells. BST2 is initially discovered as an inhibitor to HIV-1 infection in the absence of Vpu, it has also been shown to inhibit the release of other viruses such as retroviruses, filoviruses, arenaviruses, and herpes viruses. BST2 may play a role in B-cell activation in rheumatoid arthritis.

NF-kappaB is a pleiotropic transcription factor present in almost all cell types and is the endpoint of a series of signal transduction events that are initiated by a vast array of stimuli related to many biological processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. NF-kappaB is a homo- or heterodimeric complex formed by the Rel-like domain-containing proteins RELA/p65, RELB, NFKB1/p105, NFKB1/p50, REL and NFKB2/p52. The heterodimeric p65-p50 complex is the most abundant complex. The dimers bind at kappaB sites in the DNA of their target genes and the individual dimers have distinct preferences for different kappaB sites that they can bind with distinguishable affinity and specificity. Different dimer combinations act as transcriptional activators or repressors, respectively. NF-kappaB complexes are held in the cytoplasm in an inactive state complexed with members of the NF-kappaB inhibitor (I-kappaB) family. In a conventional activation pathway, I-kappaB is phosphorylated by I-kappaB kinases (IKKs) in response to different activators, subsequently degraded thus liberating the active NF-kappaB complex which translocates to the nucleus. NF-kappaB heterodimeric p65-p50 and RelB-p50 complexes are transcriptional activators. The NF-kappaB p50-p50 homodimer is a transcriptional repressor, but can act as a transcriptional activator when associated with BCL3.

Transforming Growth Factor beta -1 (TGF beta -1) is a secreted protein which belongs to the TGF- beta family. TGF beta -1 is abundantly expressed in bone, articular cartilage and chondrocytes and is increased in osteoarthritis (OA). TGF beta -1 performs many cellular functions, including the control of cell growth, cell proliferation, cell differentiation and apoptosis. The precursor is cleaved into a latency-associated peptide (LAP) and a mature TGF beta -1 peptide. TGF beta -1 may also form heterodimers with other TGF beta family members. It has been found that TGF beta -1 is frequently upregulated in tumor cells. Mutations in this gene results in Camurati-Engelmann disease.
