Description:
Platelet-derived growth factor (PDGF) is an important regulator of cell growth, proliferation, and angiogenesis. PDGF synthesis is induced by IL-1, IL-6, TNF-α, TGF-β, and EGF signaling. PDGF functions as a mitogenic growth hormone on cells of mesenchymal lineage, such as smooth muscle and glial cells. PDGF is also stored in the alpha-granules of platelets and is released upon adherence to traumatised tissues. PDGF is a dimeric glycoprotein formed by two A chains (AA), two B chains (BB), or as a heterodimer with an A and a B chain (AB). The PDGF dimer binds the cell surface receptor tyrosine kinases PDGFR-α and PDGFR-β.

Cytokines for polarizing mouse T cells into Th9 cells. Th9 cells are a unique subset of T helper cells that produce IL-9. Although the exact function of IL-9 producing T cells is yet to be completely clear it is proposed that these cells could be important in inflammation and helminth infections. Th9 cells can be generated from naive CD4+ T cells in culture by culturing them in the presence of IL-4 and TGF-β 1. It also appears that IFNγ and IL-27 can inhibit the generation of Th9 cells.

Fluor de Lys®-SIRT1 is a fluorogenic, acetylated peptide substrate for SIRT1 (human Sirtuin 1). Based on residues 379-382 of p53 (Arg-His-Lys-Lys(Ac)), a site of regulatory acetylation by the p300 and CBP acetyltransferases (lysines 381, 382), it was the best substrate for SIRT1 from among a panel of substrates patterned on p53, histone H3, and histone H4 acetylation sites. Fluor de Lys®-SIRT1 is deacetylated by SIRT1 (BML-SE239) at a rate of more then 8-fold that of the acetylated lysine substrate, Fluor de Lys® (Prod. No. BML-KI104; acetylated substrates both at 25 µM, 500 µM NAD+). The Km of Fluor de Lys®-SIRT1 for human recombinant Sirtuin 1 is 108 µM (determined at 37°C, 500 µM NAD+). Must be used in conjunction with Fluor de Lys® Developer II (Prod. No. BML-KI176). Fluorescent signal indicates deacetylation of Lys382. Sufficient for 100-200 assays of human recombinant SIRT1 (1U/well, 50-100 µM substrate).

Interleukin 34 (IL-34) was originally identified in humans, by large scale screening of secreted proteins; orthologs have also been found in chimpanzee, murine, rat and chicken. Human IL-34 is synthesized as a 242 amino acid (aa) precursor that contains a 20 aa signal sequence and a 222 aa mature chain. IL-34 is secreted as a homodimer and contains one potential N-glycosylation site. IL-34 increases proliferation of immune cells known as monocytes; it elicits its activity by binding the Colony Stimulating Factor 1 Receptor (CSF-1R). Expression of IL-34 is most abundant in the spleen and is expressed in many other tissues including the heart, brain, liver, kidney, thymus, intestine and lung. Interleukin 21 (IL-21) is a member of the common-gamma chain family of cytokines that are involved in immunoregulation. IL-21 is normally expressed by activated CD4+ T cells. The IL-21 receptor (IL-21R) activates the JAK/STAT signaling pathway and is expressed on T, B, and natural killer (NK) cells. Within the B cell lineage, IL-21 is a switch factor regulating IgG1 and IgG3 antibody production. IL-21 also cooperates with interleukin 4 (IL-4) for the production of multiple antibody classes in B cells. IL-21 has pleiotropic effects on the proliferation, differentiation, and effector functions of B, T, NK, and dendritic cells.
