Description:
Syntaxins, a family of proteins involved in the fusion of synaptic vesicles with the plasma membrane, display broad tissue distribution and contain carboxy-terminal hydrophobic domains that direct themselves to their respective intracellular compartments. Synaptin 6 is a 255 amino acid protein that is widely expressed, with higher expression levels in brain, lung and kidney. This synaptin co-localizes with vesicle associated membrane protein (VAMP) 4 to tubular and vesicular membranes of the Golgi apparatus. The cytosolic domain of Syntaxin 6 reduces the rate on Glut4 reinternalisation upon insulin withdrawl and is involved in a memrane-trafficking process that removes Glut4 from traffic directed to the plasma membrane. Syntaxin 6 is upregulated in activated macrophages in conjunction with an increase in the secretion of cytokines. The delivery of microdomain-associated lipids and proteins to the cell surface is regulated by Syntaxin 6.

Encoding over 300 genes, chromosome 18 contains about 76 million bases. Trisomy 18, or Edwards syndrome, is the second most common trisomy after Downs syndrome. Symptoms of Edwards syndrome include low birth weight, a variety of physical development defects, heart deformations and breathing difficulty. Translocation between chromosome 18 and 14 is the most common translocation in cancers and occurs in follicular lymphomas. Niemann-Pick disease, hereditary hemorrhagic telangiectasia and erythropoietic protoporphyria are associated with chromosome 18. The TGF∫ modulators, Smad2, Smad4 and Smad7 are encoded by chromosome 18. The FAM59A gene product has been provisionally designated FAM59A pending further characterization.

Synucleins are small soluble proteins expressed primarily in neural tissues and in certain tumors. The family includes 3 known proteins, alpha synuclein, beta synuclein and gamma synuclein. All synucleins have in common a highly conserved alpha helical lipid binding motif with similarity to the class A2 lipid binding domains of the exchangeable apolipoproteins. The alpha and beta synuclein proteins are found primarily in brain tissue, where they are seen mainly in pre synaptic terminals. Alpha synuclein is believed to be a major component of Lewy bodies in Parkinson’s disease. Mutations in alpha synuclein are associated with rare familial cases of early onset Parkinson’s disease, Alzheimer’s disease and several other neurodegenerative illnesses.

The ADP-ribosylation factor (Arf) family comprises a group of structurally and functionally conserved 21 kDa proteins, which are members of the Ras superfamily of regulatory GTP-binding proteins. Arf is involved in intracellular protein traffic to and within the Golgi complex. Arf has a number of disparate activities including maintenance of organelle integrity, assembly of coat proteins, as a co-factor for cholera toxin and as an activator of phospholipase D. Like other small GTPases, Arf is found to be active when bound to GTP and inactive when bound to GDP. Arf’s activation is dependent upon guanine nucleotide exchange factors (GEFs) which increase the rate of exchange of bound GDP with GTP. All GEFs have a highly conserved Sec7 domain. GEF activity lies in the Sec7 domain and this activity has been shown to be inhibited by the fungal metabolite brefeldin-A (BFA). A small group of GEFs which are insensitive to brefeldin-A (BFA) include cytohesin-1 (B2-1), cytohesin-2 (ARNO), cytohesin-3 (ARNO3), and cytohesin-4. All cytohesins function in the cell periphery and contain a pleckstrin homology (PH) domain. The PH domain has been shown to interact with phosphatidylinositol 3,4,5-triphosphate and is believed to promote membrane targeting of the cytohesins. Recruitment of the cytohesins to the membranes can occur in response to tyrosine kinase receptor activation. This response appears to require the activation of phosphatidylinositol 3-kinase (PI 3-kinase).

UFD1L is a member of the UFD1 family of proteins and is a component of the ubiquitin-dependent proteolytic pathway which degrades ubiquitin fusion proteins. This complex, also containing UFD1L, VCP and NPLOC4, binds ubiquitinated proteins and is required for the export of misfolded proteins from the ER to the cytoplasm for disposal. The NPLOC4-UFD1L-VCP complex also regulates spindle disassembly at the end of mitosis and is necessary for the formation of a closed nuclear envelope. UFD1L gene hemizygosity is the cause of some developmental defects including DiGeorge syndrome (DGS), velo-cardio-facial syndrome (VCFS) and Opitz G/BBB syndrome. UFD1L has 2 named isoforms produced by alternative splicing.

Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH10 (protocadherin 10), also known as PCDH19 or OL-PCDH, is a 1,040 amino acid single-pass type I membrane protein that contains six cadherin domains and one transmembrane domain. Expressed at moderate levels in brain, ovary and testis and present at lower levels in all other tissues, PCDH10 functions as a calcium-dependent cell-adhesion protein that may function as a tumor suppressor. When underexpressed, PCDH10 is associated with the progression of various carcinomas, including gastric cancer.
