Description:
Involved in autophagic vesicle formation. Conjugation with ATG12, through a ubiquitin-like conjugating system involving ATG7 as an E1-like activating enzyme and ATG1 as an E2-like conjugating enzyme, is essential for its function. The ATG12-ATG5 conjugate acts as an E3-like enzyme which is required for lipidation of ATG8 family proteins and their association to the vesicle membranes. Involved in mitochondrial quality control after oxidative damage, and in subsequent cellular longevity. The ATG12-ATG5 conjugate also negatively regulates the innate antiviral immune response by blocking the type I IFN production pathway through direct association with RARRES3 and MAVS. Also plays a role in translation or delivery of incoming viral RNA to the translation apparatus. Plays a critical role in multiple aspects of lymphocyte development and is essential for both B and T lymphocyte survival and proliferation. Required for optimal processing and presentation of antigens for MHC II. Involved in the maintenance of axon morphology and membrane structures, as well as in normal adipocyte differentiation. Promotes primary ciliogenesis through removal of OFD1 from centriolar satellites and degradation of IFT2 via the autophagic pathway. May play an important role in the apoptotic process, possibly within the modified cytoskeleton. Its expression is a relatively late event in the apoptotic process, occurring downstream of caspase activity. Plays a crucial role in IFN-gamma-induced autophagic cell death by interacting with FADD.

Description:
Phosphatidylethanolamine (PtdEtn) is a major membrane phospholipid which serves to play a primary role in cell membrane structure and is also involved in cell division, cell signaling, activation, phagocytosis and autophagy. PCYT2 (Phosphorylethanolamine transferase), also known as Ethanolamine-phosphate cytidylyltransferase, is a 389 amino acid protein that catalyzes the formation of CDP-ethanolamine from ethanolamine. This product combined with diacylglycerol form phosphatidylethanolamine via the de novo Kennedy pathway. PCYT2 is expressed at highest levels in heart, liver and skeletal muscle. Elevated levels of MyoD, reduced content of Sp1 and a changed ratio of Sp1 to Sp3 all together stimulate upregulation of PCTY2 transcription during C2C12 muscle cell differentiation. Disruption of the PCYT2 gene in mice leads to death after embryo implantation, establishing the necessity of PCYT2 for murine development.

Description:
Protein tyrosine kinase that is part of several cell surface receptor complexes, but that apparently needs a coreceptor for ligand binding. Essential component of a neuregulin-receptor complex, although neuregulins do not interact with it alone. GP30 is a potential ligand for this receptor. Regulates outgrowth and stabilization of peripheral microtubules (MTs). Upon ERBB2 activation, the MEMO1-RHOA-DIAPH1 signaling pathway elicits the phosphorylation and thus the inhibition of GSK3B at cell membrane. This prevents the phosphorylation of APC and CLASP2, allowing its association with the cell membrane. In turn, membrane-bound APC allows the localization of MACF1 to the cell membrane, which is required for microtubule capture and stabilization. In the nucleus is involved in transcriptional regulation. Associates with the 5'-TCAAATTC-3' sequence in the PTGS2/COX-2 promoter and activates its transcription. Implicated in transcriptional activation of CDKN1A; the function involves STAT3 and SRC. Involved in the transcription of rRNA genes by RNA Pol I and enhances protein synthesis and cell growth.

Description:
Ancillary protein that assembles as a beta subunit with a voltage-gated potassium channel complex of pore-forming alpha subunits. Modulates the gating kinetics and enhances stability of the channel complex. Assembled with KCNB1 modulates the gating characteristics of the delayed rectifier voltage-dependent potassium channel KCNB1 (PubMed:12954870). Associated with KCNC4/Kv3.4 is proposed to form the subthreshold voltage-gated potassium channel in skeletal muscle and to establish the resting membrane potential (RMP) in muscle cells. Associated with KCNQ1/KCLQT1 may form the intestinal cAMP-stimulated potassium channel involved in chloride secretion that produces a current with nearly instantaneous activation with a linear current-voltage relationship.

Description:
DNA damage results in the arrest of cell cycle progression, allowing the damaged DNA to be repaired prior to replication. Checkpoints exist at several cell cycle phase transitions to maintain this genetic integrity. Rad9, Rad17, Rad24 and Mec3 are involved in activating the G1 and G2 checkpoints (1–4). Pol2 (also known as Dun2), encoding the catalytic subunit of DNA polymerase epsilon, plays a role in activating the S phase checkpoint (5). The protein kinase Rad53 (also designated Spk1, Mec2 or Sad1) is essential for both G2 and S phase arrest. Activation of Rad53 is regulated by Mec1 (also known as Esr1 and Sad3), a homolog of the human ATM protein (6). Pds1 and Mad2 both regulate checkpoints associated with incomplete spindle replication (7,8). Dun1, another protein kinase, plays a role in transducing the DNA damage signal (9).

Description:
SETBP1 (SET binding protein 1), also known as SEB, is a 1,542 amino acid nuclear protein that contains three AT hook DNA-binding domains, one SKI homology region and a C-terminal SET-binding domain, which is followed by three PPLPPPPP repeats. SETBP1 may be involved in SET-related tumorigenesis and leukemogenesis by regulating the transforming activity of SKI in the nucleus or suppressing SET function. As a widely expressed protein, SETBP1 is encoded by a gene that maps to human chromosome 18, which houses over 300 protein-coding genes and contains nearly 76 million bases. There are a variety of diseases associated with defects in chromosome 18-localized genes, some of which include Trisomy 18 (also known as Edwards syndrome), Niemann-Pick disease, hereditary hemorrhagic telangiectasia, erythropoietic protoporphyria and follicular lymphomas.

Sulfonylureas are a class of drugs commonly used in the management of noninsulin-dependent diabetes mellitus. ENSA/ARPP-19, also known as ARPP-19e, is a 121 amino acid protein that localizes to the cytoplasm and belongs to the endosulfine family. ENSA/ARPP-19 is an endogenous regulator of the β-cell K(ATP) channels and reduces K(ATP) channel currents by inhibiting sulfonylurea from binding to the receptor thereby stimulating insulin secretion by pancreatic β cells. It is suggested that ENSA/ARPP-19 expression is decreased in brains of both Alzheimer's disease (AD) and Down's syndrome patients. Widely expressed with high levels in skeletal muscle and brain and lower levels in pancreas, ENSA/ARPP-19 is phosphorylated by PKA and exists as eight alternatively spliced isoforms. ENSA/ARPP-19 is encoded by a gene located on human chromosome 1q21.3.

The collapsin response mediator protein (CRMP) family of five cytosolic phosphoproteins are highly expressed throughout brain development. The functions of CRMPs encompass signal transduction in developmental guidance cues as well as multiple cellular and molecular events involved in apoptosis/proliferation, cell migration, and differentiation. In the adult brain, the expression of CRMPs is dramatically downregulated. However, CRMPs remain expressed in structures that retain their capacity for differentiation and plasticity. The expression of CRMPs is altered in neurodegenerative diseases, and these proteins may have a role in the physiopathology of the adult nervous system.

Description:
UBE20, also known as E2-230K, is a 1,292 amino acid member of the uniquitin-conjugating enzyme family that is involved in protein modification. Expressed predominately in heart and skeletal muscle, UBE2O functions to catalyze the ATP-dependent covalent attachment of ubiquitin to select proteins, thereby targeting the ubiquitinated proteins for proteasomal degradation. The gene encoding UBE2O maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, though specifically it is recognized as a genetic determinant of early onset breast cancer and predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes.

alpha-methylacyl-CoA racemase(AMACR/P504S) is Prostate-specific antigen (PSA) screening for prostate cancer is now widespread in the United States among men of all ages. However PSA has limited specificity because benign disease, including prostatic enlargement and inflammation, can increase PSA levels. Thus, a more specific prostate cancer markers is needed. One such potential marker is AMACR, an enzyme that is involved in peroxisomal beta-oxidation of dietary branched-chain fatty acids. Recent studies have shown that, compared with expression in normal or benign prostate epithelium, AMACR is consistently overexpressed in prostate cancer epithelium, making it a specific marker for cancer cells within the prostate gland. Furthermore, overexpression of AMACR may increase the risk of prostate cancer because its expression is increased in premalignant lesions (prostatic intraepithelial neoplasia).
