Crystallins are the major proteins of the vertebrate eye lens, where they maintain the transparency and refractive index of the lens. Crystallins are divided into alpha, beta, and gamma families, and the beta and gamma-crystallins also comprise a superfamily. Crystallins usually contain seven distinctive protein regions, including four homologous motifs, a connecting peptide, and N- and C-terminal extensions. gamma-crystallins are structural proteins in the lens, and they exists as monomers which typically lack connecting peptides and terminal extensions. The gamma-crystallins include seven closely related gamma A, gamma B, gamma C, gamma D, gamma E, gamma F, and gamma G-crystallin, as well as the gamma N and gamma S-crystallin genes. The gamma-crystallins are differentially regulated after early development, and are involved in cataract formation as a result of either age-related protein degradation or genetic mutation.

It also plays an important part in transmitting extracellular signals to the cytoskeleton and the membrane. DLG5 which can interact with MPP1 and CTNNB1, is primarily expressed in prostate and placenta.

In addition, the protein encoded by this gene binds caveolin-3, a skeletal muscle membrane protein which is important in the formation of caveolae. Cathelicidins are a family of antimicrobial proteins found in the peroxidase-negative granules of neutrophils. Along with the family of proteins known as defensins, cathelicidins participate in the first line of defense by preventing local infection and systemic invasion of microbes. FALL-39 precursor (FALL-39 peptide antibiotic, cationic anti-microbial protein, CAMP, CAP-18, HSD26) is a cathelicidin anti-microbial protein that contains the antibacterial peptide LL-37 (amino acids 134-170). In contrast to the defensins, which are cysteine-rich peptides that fold in ∫-pleated sheets, LL-37 is a cysteine-free peptide that can adopt an amphipathic å-helical conformation. LL-37 binds to bacterial lipopolysaccharides (LPS) and is a potent chemotactic factor for recruiting mast cells to sites of inflammation. LL-37 is present in inflammatory skin diseases that include psoriasis, sub-acute lupus erthematosus, dermatitis and nickel contact hypersensitivity. It is not found in normal skin epidermis. The secreted protein is expressed primarily in bone marrow, testis and neutrophils. The mouse and rat ortholog, CRAMP (cathelin-related antimicrobial peptide), is also part of the cathelicidin family of host defense peptides. These include precursors of potent antimicrobial peptides that direct antimicrobial activity against various microbial pathogens and also activate mesenchymal cells during wound repair. CRAMP is expressed in testis, spleen, stomach and intestine.

Leukocyte cell-derived chemotaxin 2 (LECT2) is a secreted protein with a neutrophil chemotactic activity. LECT2 is highly expressed in liver and shows diffuse immunostaining within the cytoplasm of hepatocytes. The LECT2 protein consists of 133 amino acids and three intramolecular disulfide bonds, and homologues of LECT2 have been widely identified in many vertebrates. LECT2 has a multifunctional role that extends from cell growth, differentiation, damage/repair process and carcinogenesis to autoimmune diseases. LECT2 expression is specifically induced in liver by β-catenin signaling. Serum LECT2 levels have been shown to increase in response to liver recovery, suggesting LECT2 may be used as a prognostic indicator.

Protocadherins are a subfamily of cadherins, a large group of related glycoproteins that mediate calcium-dependent cell-to-cell adhesion via a homophilic mechanism. Involved in a variety of functions, protocadherins help to regulate neural development and synapse formation. PCDH1 is a 1,026 amino acid single-pass type I membrane protein that contains seven cadherin domains and is a member of the protocadherin family. Localized to cell-cell and cell-matrix boundaries and expressed at high levels in brain and neuro-glial cells, PCDH1 is thought to be involved in cell adhesion and cell-cell interactions and may play a role in neuronal development. PCDH1 contains a C-terminal cytoplasmic region, an extracellular region and a transmembrane region, and is expressed as two isoforms due to alternative splicing events.
