Description:   This antibody recognises a 60 kDa protein, identified as the heat shock protein 60 (hsp60). The antibody's epitope is localized between amino acids 383-419 of human HSP60. A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp’s), more appropriately called as stress response proteins (srp’s). HSP60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial HSP60, but also against its mammalian homolog. Clone LK2 antibody, unlike LK1 antibody, recognises both mammalian and bacterial HSP60.

Description:   HIF1 (hypoxia-inducible factor 1), a heterodimeric transcription factor complex central to cellular response to hypoxia, consists of two subunits (alpha and beta) which are basic helix-loop-helix proteins of the PAS (Per, ARNT, Sim) family. Expression of HIF-1 alpha is regulated by cellular oxygen level alterations as well as in oxygen-independent manner via different cytokines (through the PI3K-AKT-mTOR pathway), growth factors, oncogenic activation, or loss of tumor suppressor function etc. In normoxic cells, HIF-1 alpha is proline hydroxylated leading to a conformational change that promotes its binding to the VLH (von Hippel Lindau) protein E3 ligase complex; ubiquitination and followed by rapid proteasomal degradation. Hypoxia as well as chemical hydroxylase inhibitors (desferrioxamine, cobalt etc.) inhibit HIF-1 alpha degradation and lead to its accumulation in the cells, whereas, contrastingly, HIF-1 beta/ARNT (AhR nuclear translocator) remains stable under both conditions. Besides their critical role in hypoxic response, HIFs regulates the transcription of genes responsible for angiogenesis, erythropoiesis/iron-metabolism, glucose metabolism, cell proliferation/survival, adipogenesis, carotid body formation, B lymphocyte development and immune reactions.

Description:   Estrogen receptor (ER) is an important regulator of growth and differentiation in the mammary gland. It is found in two forms, alpha and beta. Both forms are widely expressed, however, alpha is the predominant form in endometrium and breast cancer cells, and beta is more highly expressed in brain, kidney, heart, lung and bone. Activated Estrogen receptor dimerizes and forms both homo- and heterodimers. ER contains five domains: A-F. The A and B domain are able to weakly activate ER in the absence of ligand. The C domain binds DNA and the E domain contains the ligand binding site. Presence of ER in breast tumors indicates an increased likelihood of response to anti-estrogen (e.g. tamoxifen) therapy. This antibody is specific to estrogen receptor alpha and shows minimal cross-reaction with other members of the family.

Description:   Cytokeratin 14 (CK14) belongs to the type I (or A or acidic) subfamily of low molecular weight keratins and exists in combination with keratin 5 (type II or B or basic). Cytokeratin 14 is found in basal cells of squamous epithelia, some glandular epithelia, myoepithelium, and mesothelial cells. antibody to cytokeratin 14 is useful in differentiating squamous cell carcinomas from poorly differentiated epithelial tumors. cytokeratin 14 antibody is one of the specific basal markers for distinguishing between basal and non-basal subtypes of breast carcinomas. Cytokeratin 14 antibody is also a good marker for differentiation of intraductal from invasive salivary duct carcinoma by the positive staining of basal cells surrounding the in-situ neoplasm as well as for differentiation of benign prostate from prostate carcinoma. Furthermore, this antibody has been useful in separating oncocytic tumors of the kidney from its renal mimics, and in identifying metaplastic carcinomas of the breast.

Description:   This antibody is specific to the kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. In general, the ratio of Kappa to Lambda is 3:1. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

Cytokeratin 19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Cytokeratin 19 antibody reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Cytokeratin 19 antibody is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.
