Description:
This mAb reacts with a 200 kDa protein, identified as heavy subunit of neurofilaments. Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Carcinoids, neuroendocrine carcinomas of the skin, and cell carcinomas of the lung also express neurofilament.

Description:
Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognises two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.

This antibody recognises a glycoprotein of 110 kDa, which is identified as CD68. antibody to CD68 is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. CD68 antibody reacts with myeloid precursors and peripheral blood granulocytes. It also reacts with plasmacytoid T cells, which are supposed to be of monocyte/macrophage origin. CD68 shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Lymphomas are negative or show few granules.

Description:
A disulphide-linked heterodimer, consisting of mb-1 (or CD79a) and B29 (or CD79b) polypeptides, is non-covalently associated with membrane-bound immunoglobulins on B cells. This complex of mb-1 and B29 polypeptides and immunoglobulin constitute the B-cell Ag receptor. CD79a first appears at pre B-cell stage, early in maturation, and persists until the plasma cell stage where it is found as an intracellular component. It is found in the majority of acute leukemias of B-cell precursors, lines and lymphomas, and in some myelomas. It is not present in myeloid or Tcell lines. CD79a antibody is generally used to complement CD20 mAb, especially for mature B-cell lymphomas after treatment with Rituximab (anti-CD20). This CD79a antibody will stain many of the same lymphomas as CD20 mAb, but also is more likely to stain B-lymphoblastic lymphoma/leukemia than is CD20 mAb. CD79a antibody also stains more cases of plasma cell myeloma and occasionally some types of endothelial cells as well.

This antibody recognises a protein of 33-37 kDa, identified as CD20 (Workshop V; Code CD20.12). The antibody recognises the extracellular domain of CD20. The epitope is similar to or identical to that recognized by other CD20 antibodies including Leu-16 and B1. This antibody can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood, Bcell localization in tissues and B lymphocyte purification by immunosorbent methods. CD20 is a non-Ig differentiation antigen of Bcells and its expression is restricted to normal and neoplastic Bcells, being absent from all other leukocytes and tissues. It is expressed by pre Bcells and persists during all stages of Bcell maturation but is lost upon terminal differentiation into plasma cells. Protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting Bcells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as a calcium channel involved in Bcell activation and cell cycle progression.

This antibody recognises a single chain, transmembrane, heavily glycosylated protein of 90 to 120 kDa, which is identified as CD34. Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi’s sarcoma, but shows low specificity.

Description:
The antibody recognises a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesised as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin’s disease, CD30 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This antibody distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. About one third of the CD30 positive lymphomas lack the leukocyte common antigen (CD45).
