Mitoferrin 2, also known as MRS3/4 (mitochondrial RNA-splicing protein 3/4 homolog), mitochondrial iron transporter 2, NPD016, MRS4L or SLC25A28 (solute carrier family 25 member 28), is a 364 amino acid multi-pass membrane protein of the mitochondrial inner membrane that mediates iron uptake. Mitoferrin 2 is thought to play a role in heme synthesis of hemoproteins and iron-sulfur cluster assembly. Ubiquitously expressed, Mitoferrin 2 is found at high levels in skeletal muscle, heart, placenta, kidney, lung, liver, brain and pancreas. Mitoferrin 2 is a member of the mitochondrial carrier family and undergoes alternative splicing events to produce four isoforms. Mitoferrin 2 contains three solcar repeats and is encoded by a gene that maps to human chromosome 10q24.2.

Adaptins are heterotetrameric subunits of adaptors, which are complexes involved in the formation of Clathrin-coated pits for vesicle-mediated endocytosis. It is localized at a subset of adherens junctions, specialized cell-cell and cell-matrix associations that are characterized by the presence of filamentous actin at the cytoplasmic face of the junctional complex. In cultured cells, talin is absent from cell-cell junctions and found predominantly at adhesion plaques and in fibrillar streaks underlying cell surface fibronectin. Talin interacts with at least two other proteins that are localized at adhesion plaques, vinculin and integrin. Talin and vinculin have been shown to interact with each other and both have been proposed to be involved in generating the transmembrane connection, between the extracellular matrix and the cytoskeleton, that occurs at adhesion plaques. At physiological ionic strength, talin is an elongate, flexible, monomeric protein with the ability to self-associate into dimers at higher protein concentrations.

Expressed as multiple alternatively spliced isoforms, pantophysin is present in tissues throughout the body where it may play a role in vesicle trafficking and protein transport. The gene encoding pantophysin maps to human chromosome 7, which houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localized to chromosome 7 have been linked to Osteogenesis imperfecta, Williams-Beuren syndrome, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome.

ARID3A, also known as DRIL1 in humans and Bright (for B cell regulator of IgH transcription) in mice, are the mammalian homologs of the Drosophila Dri (dead ringer) protein. ARID3A is developmentally regulated and is expressed in a restricted set of cells, including differentiating cells of the gut and salivary glands. ARID3A represents a member of a unique family of transcriptional activators that shares sequence similarity to proteins of SWI/SNF complexes; it contains an A/T-rich DNA-binding (ARID) domain and a distinct domain involved in tetramerization. The gene encoding ARID3A is linked to a marker of Peutz-Jeghers syndrome, which is an autosomal-dominant disorder characterized by melanocytic macules of the lips, multiple gastrointestinal hamartomatous polyps and an increased risk for various neoplasms, including gastrointestinal cancer. E2FBP1 (E2F-1 binding protein 1) is identical to ARID3A in the carboxy terminal region. E2FBP1 appears to lack DNA binding and transactivation domains, and it functions to regulate the transcription of proteins involved in cell proliferation by binding to the transcription factor E2F-1.

BRCC45 was initially suggested to be a housekeeping protein that is highly expressed in brain and reproductive organs. Later experiments indicated BRCC45 forms a complex with the breast and ovarian predisposition proteins BRCA1 and BRCA2 as well as RAD51 and BRCC36. This complex has a ubiquitin E3 ligase activity and is thought to enhance cellular survival following DNA damage. BRCC45 has also been suggested to function as a death receptor associated anti apoptotic protein by inhibiting the BID induced activation of the mitochondrial apoptotic pathway. Higher levels of BRCC45 were detected in the majority of hepatocellular carcinomas, suggesting that BRCC45 may promote tumorigenesis when overexpressed. At least three isoforms of BRCC45 are known to exist.
