The engrailed-2 gene, EN2, a murine homolog of the Drosophila homeobox gene engrailed (EN), is required for midbrain and cerebellum development and dorsal/ventral patterning of the limbs as well as apical ectodermal ridge formation. In Drosophila, the EN gene plays an important role during development in segmentation, where it is required for the formation of posterior compartments. Human EN-1 and EN-2 are homeodomain-containing proteins and have been implicated in the control of pattern formation during development of the central nervous system. Different mutations in the mouse homologs, EN-1 and EN-2, produce different developmental defects that frequently are lethal. EN-1 is highly expressed by essentially all dopaminergic neurons in the substantia nigra and ventral tegmentum. EN-1 and EN-2 regulate expression of a-synuclein, a gene that is genetically linked to Parkinson’s disease. During early brain development mouse EN-2 is expressed in a broad band across most of the mid-hindbrain region. EN-2 is also expressed in mouse myoblasts and has been assiciated with cerebellar hypoplasia.

Emx1 and Emx2 are homeobox proteins expressed in the developing vertebrate brain. Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin (PCDH) gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHGA9 (Protocadherin gamma-A9) is a 932 amino acid that is one of 22 proteins encoded by the protocadherin gamma cluster. The protocadherein gamma cluster consists of three subfamilies (A, B and C) and PCDHGA9 is a member of the gamma subfamily A. PCDHGA9 is a type I transmembrane receptor containing six cadherin motifs and is expressed in the central nervous system where it localizes to synapses. Members of the gamma cluster of protocadherins are essential for neuronal survival. There are two isoforms of PCDHGA9 that are produced as a result of alternative splicing events.

Humanin, an endogenous anti-apoptotic peptide against Alzheimer disease-related insults, consists of 24 amino acids. The secreted protein is a neuroprotective factor against death induced by several different types of Alzheimer's disease genes. Humanin protects neuronal cells from damage caused by Alzheimer's disease genes, specifically APP (amyloid precursor protein). Humanin acts as a ligand for formyl peptide receptor-like 1 (FPRL1) with APP and utilizes its neuroprotective effects by inhibiting FPRL1 access to APP. The peptide prevents Bax translocation from cytosol to mitochondria. Humanin expression levels may be dependent on defects in energy production in muscles with mitochondrial abnormalities. The peptide has been detected in muscles of patients with the mitochondrial disease chronic progressive external ophthalmoplegia (CPEO). Humanin is mainly expressed in the kidney, heart, liver, testis and skeletal muscles.
