CTGF belongs to the CCN (CTGF/Cyr61/Cef10/NOVH) protein family, which is comprised of six secreted proteins that reside in the extracellular matrix (ECM). CTGF causes a variety of cellular responses including reduced cell adhesion and enhanced cell migration and proliferation. CTGF has also been shown to be essential for epithelial to mesenchymal transition (EMT), a process whereby normal functioning cells morph into ones that produce mainly scar tissue (of which collagen in the major protein component).
Description:
B18R, also known as B19R in the Copenhagen strain of Vaccinia, is a type I interferon (IFN)-binding protein encoded by the B18R open reading frame in the Western Reserve strain of vaccinia virus. B18R exists in two forms, soluble and membrane-bound form and has a broad species specificity. B18R has high affinity for human IFN-alpha and binds rabbit, bovine, rat, pig, and mouse IFN-alpha and IFN-beta. Secreted B18R binds to uninfected and infected cells. B18R presents at the cell surface and protects cells from the antiviral state. It has shown that binding of soluble recombinant B18R protects cultured cells from IFN and allows vaccinia virus replication.
Description:
C1q R1 is also known as CD93, collectin receptor, and AA4 antigen, belongs to the Group XIV C-Type lectin family which play a role not only in cell–cell adhesion processes but also in host defence. All of them contain a C-type lectin domain, a series of epidermal growth factor like domains (EGF), a highly glycosylated mucin-like domain, a unique transmembrane domain and a short cytoplasmic tail. C1q R1 has also been identified as a hematopoietic stem cell marker.
Description:
Cystatin-D is a protein that in humans is encoded by the CST5 gene. The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a protein found in saliva and tears. The encoded protein may play a protective role against proteinases present in the oral cavity.
Description:
Flagellin is the subunit protein which polymerises to form the filaments of bacterial flagella. It activates the innate immune system through the receptor Toll-like Receptor 5 (TLR5) or the intracellular NLRC4 protein. The Flagellin (TLR5 Mutant)(rec. ) cannot be detected by human and mouse TLR5. It has not been tested on NLRC4.
Description:
Ferritin is a large, iron-storage heteropolymeric protein, which is expressed in most kinds of cells and co-assemble in different proportion in a tissue-specific manner. Ferritin has oligomer of 24 subunits and two types of subunits including light chain(FTL) and heavy chain. Ferritin can remove Fe (Ⅱ) from solution in the presence of oxygen and is very important for iron homeostasis. Iron is absorbed in the ferrous form and deposited as ferric hydroxides after oxidation. Iron is first oxidised to the ferric state for storage as ferric oxyhdroxide whithin the protein shell of ferritin. Thus, ferritin removes excess iron from the cell sap where it could otherwise participate in peroxidation mechanisms. Ferritin also plays a role in delivery of iron to cells and mediates iron uptake in capsule cells of the developing kidney.
Description:
The prolactin receptor (PRLR) is a member of the class I cytokine/lactogen receptor family which mediates the diverse cellular actions of prolactin in several tissues. PRLRs are expressed in normal and neoplastic human breast tissue, and in most breast cancer cells. PRLR contains an extracellular region that binds prolactin, a transmembrane region, and a cytoplasmatic region required for the activation of the Jak2–Stat5 signal transduction pathway by Prl which is essential for transcriptional activation of all known prolactin regulated genes. PRLRs have also been observed in ovarian follicular cells of mice, pigs, sheep, deer, and humans, as well as in luteal tissue in cow and horse ovaries. Furthermore, PRLR knockout mice exhibit failure of embryonic implantation, reduced number of mature oocytes, and low fertilisation rates. Knockout females also display a reduced number of primary follicles.
Description:
Tumor necrosis factor receptor superfamily member 1B(TNFRSF1B) is expressed by the gene TNFRSF1B. The soluble form is produced from the membrane form by proteolytic processing. It can bind to TRAF2, and interacts with BMX. It can act as the receptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha.
Description:
CD5 Antigen-Like (CD5L) is a soluble protein that belongs to group B of the scavenger receptor cysteine-rich (SRCR) superfamily and contains three SRCR domains. CD5L is a secreted glycoprotein and expressed by macrophages presentin lymphoid tissues. It binds to myelomonocytic and lymphoid cells and may play an important role in the regulation of the innate and adaptive immune systems. CD5L functions as a pattern recognition molecule by binding both lipoteichoic acid (LTA) on Gram positive and lipopolysaccharide (LPS) on Gram-negative bacteria and the SRCR domain one of CD5L retains both the LPS and LTA binding activities. Furthermore, CD5L seems to play a role as an inhibitor of apoptosis.
Description:
Complement Factor H-Related 1 (CFHR1) is a 43 kDa secreted member of the factor H family of glycoproteins. The human Complement Factor H protein family consists of the complement and immune regulators factor H, the factor H-like protein 1 (FHL-1) and five factor H-related proteins (CFHR-1 to -5). Members of the H-related protein family are exclusively composed of individually folded protein domains, termed short consensus repeats (SCRs) or complement control modules. FHR1 is produced by hepatocytes and circulates as two differentially glycosylated isoforms (37 kDa and 43 kDa). Mature human FHR1 is 312 amino acids in length. It contains five, approximately 60 aa SCRs that basically constitute the entire molecule. FHR1 may play a role in complement regulation, lipid metabolism and lipoprotein complexes that bind PMNs to LPS.
Description:
Visinin-Like Protein 1 (VILIP) is a a member of the Visinin/Recoverin subfamily of neuronal calcium sensor proteins. VILIP is strongly expressed in the Granule Cells of the Cerebellum where it associates with membranes in a Calcium-dependent manner and modulates intracellular signaling pathways of the central nervous system by directly or indirectly regulating the activity of Adenylyl Cyclase. It has been shown that VILIP regulates the inhibition of rhodopsin phosphorylation in a Calcium-dependent manner in vitro.
Description:
Fms-related tyrosine kinase 3 ligand(Flt3L) is a single-pass type I membrane protein and consists of 232 amino acids. Flt3L is a hematopoietic four helical bundle cytokine, structurally homologous to stem cell factor and colony stimulating factor. Flt3L synergises well with a number of other colony stimulating factors and interleukins. Flt3L stimulates the proliferation and differentiation of various blood cell progentiors by activating FLT3.
Description:
Interleukin-15 receptor subunit alpha, also known as Il15ra, is a high-affinity receptor for interleukin-15. Il15ra associates as a heterotrimer with the IL-2 receptor beta and gamma subunits (Common gamma chain, or gamma c) to initiate signal transduction. It can signal both in cis and trans where IL15R from one subset of cells presents IL15 to neighboring IL2RG-expressing cells. Il15ra is expressed in special cells including a wide variety of Tand B cells and non-lymphoid cells. Human Il15ra shares 45% amino acid sequence homology with the mouse form of the receptor. Eight isoforms of IL-15 R alpha mRNA have been identified, resulting from alternative splicing events involving different exons.Interleukin 15 (IL-15) is a cytokine that regulates T cell and natural killer cell activation and proliferation. IL-15 binds to the alpha subunit of the IL15 receptor (IL-15RA) with high affinity. IL-15 also binds to the beta and gamma chains of the IL-2 receptor, but not the alpha subunit of the IL2 receptor. IL-15 is structurally and functionally related to IL-2. Both cytokines share some subunits of receptors, allowing them to compete for and negatively regulate each other's activity. The number of CD8+ memory T cells is controlled by a balance between IL-15 and IL-2. Despite their many overlapping functional properties, IL-2 and IL-15 are, in fact, quite distinct players in the immune system. IL-15 is constitutively expressed by a wide variety of cell types and tissues, including monocytes, macrophages and DCs.
Description:
CD40 is a member of the TNF receptor superfamily which are single transmembrane-spanning glycoproteins and plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. CD40 contains 4 cysteine-rich repeats in the extracellular domain and is expressed in B cells, dendritic cells, macrophages, endothelial cells and several tumour cell lines. The cognate interaction between CD40 and CD40 ligand (CD154) on T cells activates NF-kappaB, Jun N-terminal kinase and Janus kinase signal transducers and activators of transcription pathways. Several different TRAF proteins (adapter proteins) have been identified to serve as mediators of the signal transduction. In addition, CD40/CD40L interaction is found to be necessary for amyloid-beta-induced microglial activation and thus is thought to be an early event in Alzheimer disease pathogenesis. Defects in CD40 result in hyper-IgM immunodeficiency type 3 (HIGM3), an autosomal recessive disorder characterised by the inability of B cells to undergo isotype switching, as well as an inability to mount an antibody-specific immune response and a lack of germinal center formation.
Description:
Intercellular Adhesion Molecule-1 (ICAM-1, CD54) binds the leukocyte integrins LFA-1 and Mac1. CD54 expression is weak on leukocytes, epithelial and resting endothelial cells, as well as some other cell types, but expression can be stimulated by IFN-gamma, TNF-alpha, IL-1beta and LPS. Soluble CD54 is found in a biologically active form in serum, probably as a result of proteolytic cleavage from the cell surface, and is elevated in patients with various inflammatory syndromes such as septic shock, LAD, cancer and transplantation.
Description:
CD40 Ligand (CD40L), renamed TNFSF5 but now also known as CD154, TRAP and gp39, is a 34-39kDa type II transmembrane glycoprotein that belongs to the TNF superfamily. As with other TNF superfamily members, CD40L will exist as a trimer, both as a membrane bound and soluble form. Multiple mutations and alternate splice forms of CD40L exist, often associated with pathology and leading to truncated or nontrimerizable forms of CD40L. CD40L binds to both CD40 and to integrin alphaIIbbeta3 (CD41). In the cell membrane, it also associates with a unique splice variant of CD28 (CD28i) that may facilitate CD40L signal transduction. CD40L is expressed by monocytes, NK cells, mast cells, basophils, smooth muscle cells, endothelial cells, dendritic cells, activated and resting B cells, plus activated platelets and CD4+ T cells. CD40L ligation of CD40 on dendritic cells (DC) initiates DC maturation and differentiation. CD40L signalling into naive B cells promotes germinal center formation and isotope switching. CD40-CD40L seems to bridge innate and adaptive immune signals.
