Wnt1 is a secreted protein that signals through the Frizzled family of cell surface receptors and is required for normal embryonic development. Wnt1 activation induces a complex signaling cascade that ultimately leads to the increased expression of over fifty genes. An important component of Wnt1 signaling is the stabilization, and resulting accumulation, of the intracellular signaling protein, beta catenine. Mature human Wnt1 is a glycosylated protein containing 343 amino acid residues.

HOXB8 belongs to the Antp homeobox family. It is a nuclear protein with a homeobox DNA-binding domain. This gene is included in a cluster of homeobox B genes located on chromosome 17. The protein functions as a sequence-specific transcription factor that is involved in development. Increased expression of this gene is associated with colorectal cancer. Mice that have had the murine ortholog of this gene knocked out exhibit an excessive pathologic grooming behavior. Gap junctional intercellular communication is thought to play a key role in development and may also be involved in epilepsy (Aronica et al., 2001). Connexin 43 forms gap-junctional channels and regulates the permeability of these gap junctions to small organic molecules. Permeability of connexin 43 is known to be regulated by phosphorylation at Ser368 by protein kinase C (Yogo et al., 2002; Bao et al., 2004a). Phosphorylation of Ser368 by PKC induces a conformational change of connexin 43 that results in a decrease in gap junction permeability (Bao et al., 2004b).

Leukemia inhibitory factor (LIF) is a member of the interleukin 6 (IL-6) family that is made by a variety of adult and embryonic tissues. LIF signals through the glycoprotein 130 (gp130)/LIF receptor (LIFR) heterodimer to activate STAT3 and MAPK signaling. This gene encodes a protein that binds to the C-terminus of adenovirus E1A proteins. This phosphoprotein is a transcriptional repressor and may play a role during cellular proliferation. This protein and the product of a second closely related gene, CTBP2, can dimerize. Both proteins can also interact with a polycomb group protein complex which participates in regulation of gene expression during development. Alternative splicing of transcripts from this gene results in multiple transcript variants.
