The RPA-T4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation, together with the MHC class II. The RPA-T4 antibody is capable of blocking HIV binding and inhibiting syncytium formation, by binding to the D1 domain of the CD4 antigen. The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.

Description:
Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

Leptin receptor (LEPR) is also known as LEP-R, cluster of differentiation 295 (CD295), OB-R and B219, is a single-transmembrane-domain receptor of the gp130 family of cytokine receptors. Leptin receptor exists as homodimer and binds Leptin with high affinity, thus mediates the biological function of the adipocyte-specific hormone Leptin. Anthrax Toxin Receptor 1 (ANTXR1) is a single-pass type I membrane protein that belongs to the ATR family. ANTXR1 contains one VWFA domain and binds PA through the VWA domain. ANTXR1 is highly expressed in tumor endothelial cells. ANTXR1 plays a role in cell attachment and migration. Chloride Intracellular Channel Protein 4 (CLIC4) is a 253 amino acid single-pass membrane protein that localizes to both the nucleus and the cytoplasm and contains one GST C-terminal domain. CLIC4 is expressed in various tissues and exhibits an intracellular vesicular pattern in Panc-1 cells (pancreatic cancer cells). CLIC4 acts as a monomer which is able to form selective ion channels in target proteins, thus facilitating the transport of chloride and other ions. CLIC4 is believed to have a role in apoptosis and is able to translocate to the nucleus under stress conditions. CLIC4 has alternate cellular functions like a potential role in angiogenesis or in maintaining apical-basolateral membrane polarity during mitosis and cytokinesis.

T-Cell Antigen CD7 is a single-pass type I membrane protein that that belongs to the the immunoglobulin superfamily. Human CD7 is synthesized as a 240 amino acid precursor that contains a 25 amino acid signal sequence and a 215 amino acid mature chain with a Ig-like (immunoglobulin-like) domain. CD7 is normally expressed on all T-lymphocytes, NK-cells, pre-B lymphocytes and pleuripotent hematopoietic stem cells. CD7 plays an essential role in T-cell interactions, T-cell/B-cell interaction during early lymphoid development, T- and NK-cell activation and cytokine production. CD7 has been shown to interact with PIK3R1and SECTM1. However, the function of the CD7 protein in the immune system is still largely unknown.

The molecular cloning and characterisation of one member of the thioredoxin superfamily, designated as TMX2.The TMX2 cDNA consists of 1644 nucleotides and contains an open reading frame encoding a protein of 372 amino acids with a predicted molecular mass of 42.5 kDa and an isoelectric point of 8.94. The TMX2 protein may possess an N-terminal signal peptide, a potential transmembrane domain, an Myb DNA-binding domain repeat signature, a thioredoxin consensus pattern, an endoplasmic reticulum (ER) membrane retention signal (KKXX-like motif), and a dileucine motif in the tail.
