Description:
Cadherin-3 (CDH3) is a single-pass type I membrane protein that belongs to the cadherin superfamily. CDH3 is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. CDH3 is expressed in some normal epithelial tissues and in some carcinoma cell lines. CDH3 preferentially interacts with themselves in a homophilic manner in connecting cells. CDH3 is involved in loss of heterosygosity events in breast and prostate cancer. Mutations in CDH3 have been associated with congential hypotrichosis with juvenile macular dystrophy.

Description:
The hexokinases utilize Mg-ATP as a phosphoryl donor to catalyze the first step of intracellular glucose metabolism, the conversion of glucose to glucose-6-phosphate. Four hexokinase isoenzymes have been identified, including hexokinase I (HXK I), hexokinase II (HXK II), hexokinase III (HXK III) and hexokinase IV (HXK IV, also designated glucokinase or GCK). Hexokinases I-III each contain an N-terminal cluster of hydrophobic amino acids. Glucokinase lacks the N-terminal hydrophobic cluster. The hydrophobic cluster is thought to be necessary for membrane binding. This is substantiated by the finding that glucokinase has lower affinity for glucose than do the other hexokinases. HXK I has been shown to be expressed in brain, kidney and heart tissues as well as in hepatoma cell lines. HXK II is involved in the uptake and utilization of glucose by adipose and skeletal tissues. Of the hexokinases, HXK III has the highest affinity for glucose. Glucokinase is expressed in pancreatic beta cells where it functions as a glucose sensor, determining the “set point†for insulin secretion.

Tissue Factor (TF) is a single-pass type I membrane glycoprotein member of the tissue factor family. TF expression is highly dependent upon cell type. This factor enables cells to initiate the blood coagulation cascades, and it functions as the high-affinity receptor for the coagulation factor VII. TF initiates blood coagulation by forming a complex with circulating factor VII or VIIa. The complex activates factors IX or X by specific limited protolysis. TF plays a role in normal hemostasis by initiating the cell-surface assembly and propagation of the coagulation protease cascade.

Replication protein A (RPA) is a single-stranded DNA binding protein. Human RPA is a heterotrimeric protein containing subunits of 70, 32 and 14 kDa. AMP/ATP-binding subunit of AMP-activated protein kinase (AMPK), an energy sensor protein kinase that plays a key role in regulating cellular energy metabolism. In response to reduction of intracellular ATP levels, AMPK activates energy-producing pathways and inhibits energy-consuming processes: inhibits protein, carbohydrate and lipid biosynthesis, as well as cell growth and proliferation. AMPK acts via direct phosphorylation of metabolic enzymes, and by longer-term effects via phosphorylation of transcription regulators. Also acts as a regulator of cellular polarity by remodeling the actin cytoskeleton; probably by indirectly activating myosin. Gamma non-catalytic subunit mediates binding to AMP, ADP and ATP, leading to activate or inhibit AMPK: AMP-binding results in allosteric activation of alpha catalytic subunit (PRKAA1 or PRKAA2) both by inducing phosphorylation and preventing dephosphorylation of catalytic subunits. ADP also stimulates phosphorylation, without stimulating already phosphorylated catalytic subunit. ATP promotes dephosphorylation of catalytic subunit, rendering the AMPK enzyme inactive.

Macrophage colony-stimulating factor 1（M-csf）is a single-pass type I membrane protein . It is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. M-CSF affects macrophages and monocytes in several ways, including stimulating increased phagocytic and chemotactic activity, and increased tumour cell cytotoxicity. The role of M-CSF is not only restricted to the monocyte/macrophage cell lineage. By interacting with its membrane receptor, M-CSF also modulates the proliferation of earlier hematopoietic progenitors and influence numerous physiological processes involved in immunology, metabolism, fertility and pregnancy.
