This antibody recognises a protein of 185 kDa, which is identified as HER2/ErbB2. Its epitope is localized in the extracellular domain. ErbB2 is a member of the EGFR family. This antibody is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. Mouse T cell immunoglobulin and mucin domain-containing protein 4, also known as T cell membrane protein 4, TIMD4 and TIM4 is a single-pass type I membrane protein which belongs to the immunoglobulin superfamily and TIM family. TIM4 contains one Ig-like V-type (immunoglobulin-like) domain. It is expressed on dendritic cells and macrophages. TIM4 plays an important role in the proliferation of T helper type 2 (Th2) cells. TIM4 is involved in regulating T cell proliferation and lymphotoxin signalling. It is a ligand for HAVCR1 / TIMD1. The expression of TIM4 in fibroblasts enhanced their ability to engulf apoptotic cells. TIM4 and TIM1 are phosphatidylserine receptors for the engulfment of apoptotic cells and may also be involved in intercellular signalling in which exosomes are involved. Modulation of TIM4 production in dendritic cells (DCs) represents a novel therapeutic approach for the treatment of peanut allergy.

Interleukin 2 (IL2) is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. IL2 has been shown to have antitumor effects in some studies. This is probably mediated by cytotoxic effector cells.

LZTR2 Antibody: LZTR2, also known as RGPR-p117, is a member of the BTB-kelch superfamily and was initially described as a nuclear factor I (NFI) binding protein and transcriptional regulator of the regucalcin gene. LZTR2 is cytoplasmically localized but is thought to translocate to the nucleus, a process mediated by protein kinase C signaling following hormonal stimulation. Recent evidence has suggested that there is a strong correlation of single nucleotide polymorphisms of LZTR2 with obesity in the Japanese population similar to that seen with the TMEM18 gene and the GNPDA2, BDNF, FAIM2, and MC4R genes with obesity in Caucasian populations, suggesting LZTR2 may play a role in metabolism and obesity risk.

Tau is a key microtubule-associated protein that plays an important role in the formation of microtubules in axons (Binder et al. 1985). Six tau isoforms have been identified as products of a single gene produced by alternative mRNA splicing (Goedert 1990). Tau mutations have been implicated in many neurodegenerative disorders such as Alzheimer’s disease (AD), Pick’s disease and progressive supranuclear palsy. It has been well documented that hyperphosphorylated tau is a major component of paired helical filaments in AD brain (Lee 1995). Serine 416 has been demonstrated to be a major phosphorylation site in vitro by CaM kinase II (Steiner at al. 1990).

Calcium ions play a primary role in the regulation of sperm motility. Anti-CATSPER2 belongs to a family of putative cation channels that are specific to spermatozoa and localize to the flagellum. The protein family features a single repeat with six membrane-spanning segments and a predicted calcium-selective pore region. A second, closely linked copy of this gene has been identified. Although multiple transcript variants encoding protein isoforms have been characterized, they seem to be only transcribed from this gene. Additional splice variants have been described but their full-length nature has not been determined.

PSMC3IP plays an important role in meiotic recombination. It stimulates DMC1-mediated strand exchange required for pairing homologous chromosomes during meiosis. The complex PSMC3IP/MND1 binds DNA, stimulates the recombinase activity of DMC1 as well as DMC1 D-loop formation from double-strand DNA. This complex stabilizes presynaptic RAD51 and DMC1 filaments formed on single strand DNA to capture double-strand DNA. This complex stimulates both synaptic and presynaptic critical steps in RAD51 and DMC1-promoted homologous pairing. It may inhibit HIV-1 viral protein TAT activity and modulate the activity of proteasomes through association with PSMC3.

SHISA3 Antibody: SHISA3 plays an essential role in the maturation of presomitic mesoderm cells by individual attenuation of both FGF and WNT signaling. The Shisa family of single-transmembrane proteins is characterized by an N-terminal cysteine-rich domain and a proline-rich C-terminal region. Its founding member, Xenopus Shisa, promotes head development by antagonizing Wnt and FGF signaling. Shisa physically interacted with immature forms of the Wnt receptor Frizzled and the FGF receptor within the ER and inhibited their posttranslational maturation and trafficking to the cell surface. Loss of Shisa function sensitized the neuroectoderm to Wnt signaling and suppressed head formation during gastrulation.
