Description:
Thrombopoietin (TPO) is a key regulator of megakaryocytopoiesis and thrombopoiesis in vitro and <i>in vivo</i>. TPO stimulates the proliferation and maturation of megakaryocytes and has an important role in regulating the level of circulating platelets <i>in vivo</i>. TPO also promotes the survival, self-renewal, and expansion of hematopoietic stem cells and primitive multilineage progenitor cells. It is commonly used with other cytokines such as stem cell factor (SCF) and Flt3/Flk-2 Ligand to promote expansion of primitive hematopoietic cells in culture (Hitchcock andamp; Kaushansky). The TPO receptor, c-Mpl, is expressed at all stages of megakaryopoiesis, from hematopoietic stem and progenitor cells to mature platelets.

Description:
CUB domain-containing protein 1(CDCP1) is a transmembrane glycoprotein with a large extracellular domain (ECD) containing two CUB domains, and a smaller intracellular domain (ICD) containing five tyrosines. CDCP1 is widely expressed in human epithelial tissues, but its phosphorylation is only seen in mitotically detached or shedding cells, consistent with its role in the negative regulation of cell adhesion. The tyrosine phosphorylation of CDCP1 in cultured cells occurs when cells are induced to detach by trypsinor EDTA, or seen spontaneously during mitotic detachment. The overexpression of CDCP1 leads to the loss of cell adhesion and a detached phenotype.

The Mammalian β-Galactosidase Assay Reagent Kit is a simple and versatile assay to measure β-galactosidase activity in cultured mammalian cells. Cells grown on a plate may be lysed and tested for β-galactosidase activity by adding a single all-in-one lysis-and-assay solution. Peflin has a longer N-terminal hydrophobic domain than any other member of the family, and it contains nine nonapeptide (A/PPGGPYGGP) repeats. Western blot analysis demonstrated that peflin (30 kDa) was expressed in various nonadherent and adherent cultured human cell lines, including Jurkat, HL60, HeLa, and HT1080. Peflin may play basic roles in Ca(2+) signaling irrespective of cell types.
