Description:
Fibroblast growth factor-1 (FGF-1), also designated acidic FGF, and fibroblast growth factor-2 (FGF-2), also designated basic FGF, are members of a family of growth factors that stimulate proliferation of cells of mesenchymal, epithelial and neuroectodermal origin. Additional members of the family include the oncogenes FGF3 (Int2) and FGF4 (hst/Kaposi), FGF5, FGF6, FGF7 (KGF), FGF8 (AIGF), FGF9 (GAF) and FGF10 through FGF-23. Members of the family share 30-55% amino acid sequence identity and similar gene structure, and are capable of transforming cultured cells when overexpressed in trans- fected cells. Cellular receptors for FGFs are members of a second multigene family, including four tyrosine kinases designated Flg (FGFR-1), Bek (FGFR-L), TKF and FGFR-3.

It can also form heterodimers with the related proteins CEBP-beta and CEBP-gamma. The encoded protein has been shown to bind to the promoter and modulate the expression of the gene encoding leptin, a protein that plays an important role in body weight homeostasis. Also, the encoded protein can interact with CDK2 and CDK4, thereby inhibiting these kinases and causing growth arrest in cultured cells.

The PK136 monoclonal antibody is specific for the mouse NK1.1, a receptor from the killer cell lectin-like receptor (KLR) family. Nk1.1 is an antigen encoded by the Klrb1c/NKR-P1C gene expressed by the natural killer cells of some selected strains of mice (C57BL, FVB/N, NZB) and encoded by the Klrb1b/NKR-P1B gene expressed on Swiss NIH and SJL mice. Pk136 binds to an epitope common to NKR-P1B and NKR-P1C.The Klrb1 is a family of type II transmembrane C-type lectin receptors. Klrb1c activates the NK-cell cytotoxicity, while Klrb1b inhibits it. PK136 is useful in defining the NK cells and the rare population of NK-T lymphocytes and specific cultured monocytes.

The glycoprotein isolation kit allows isolation of glycoproteins from complex mixtures, including serum, tissue and cultured cell lysates. The procedure uses concanavilin A (ConA) or wheat germ agglutinin (WGA) as a ligand to bind a specific class of glycoprotein from complex protein samples - those containing alpha-lined mannose or terminal glucose residues (in case of ConA) and N-Acetyl glucosamine (GlcNAc) groups or sialic acid (in case of WGA). PRKCDBP Antibody: The protein kinase C delta (PKC-δ) binding protein (PRKCDBP), also known as cavin-3, is a member of the cavin family of proteins that are involved in caveolin formation and regulation. PRKCDBP was initially identified in a screen of cultured cell lines for proteins that were strongly induced by serum starvation. Studies indicate that PRKCDBP binds not only to PKC-δ but also to caveolin-1 and helps regulate caveolin traffic and function. Similar to other members of the cavin family, the expression of PRKCDBP was found to be down-regulated in various cancer cell lines, suggesting a possible tumor suppressor function of PRKCDBP.

Tissue inhibitor of metalloproteinases 1 (TIMP1) and Tissue inhibitor of metalloproteinases 2 (TIMP 2) have similar properties, specifically in inhibiting enzymes of the matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP1 forms a complex of 1:1 stoichiometry with activated interstitial collagenases, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP2 or gelatinase A), and latent and active forms of 92kDa Type IV collagenase (also known as MMP0 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumour cells and normal placental trophoblast cells. TIMP1 is produced in low (pg/mL) levels by most cell types. Treatment of cells with the phorbol ester TPA stimulates production of TIMP1 in some cell types, but the low protein levels produced often require concentration of cell culture media to visualize the bands by Western Blotting.

Epidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells. The EGF precursor is believed to exist as a membrane-bound molecule which is proteolytically cleaved to generate the 53-amino acid peptide hormone that stimulates cells to divide. EGF exerts its actions by binding to the EGFR, a 170 kDa protein. Epidermal growth factor is also a key growth factor regulating cell survival. Through its binding to cell surface receptors, EGF activates an extensive network of signal transduction pathways that include activation of the PI3K/AKT, RAS/ERK and JAK/STAT pathways
