Receptor-activated non-selective cation channel involved in detection of sensations such as coolness, by being activated by cold temperature below 25 degrees Celsius. Activated by icilin, eucalyptol, menthol, cold and modulation of intracellular pH. Involved in menthol sensation. Permeable for monovalent cations sodium, potassium, and cesium and divalent cation calcium. Temperature sensing is tightly linked to voltage-dependent gating. Activated upon depolarization, changes in temperature resulting in graded shifts of its voltage-dependent activation curves. The chemical agonists menthol functions as a gating modifier, shifting activation curves towards physiological membrane potentials. Temperature sensitivity arises from a tenfold difference in the activation energies associated with voltage-dependent opening and closing.

GP350 meditates EBV attachment to B lymphocytes via binding to the cell surface receptor CR2 (also known as C3d receptor or CD21) (Nemerow <i>et al.</i>; Tanner <i>et al.</i>). Viral peptide pools are useful for a broad range of applications, including vaccine development, immunological research, and diagnostic assay development.

Description:
This antibody is specific to Complement 4d (C4d) and it reacts with the secreted as well as cell-bound protein. C4d is a degradation product of the activated complement factor C4b. Complement 4b is typically activated by binding of antibodies to specific target molecules. Following activation and degradation of the C4 molecule, thio-ester groups are exposed, which allow transient, covalent binding of the degradation product C4d to endothelial cell surfaces and extracellular matrix components of vascular basement membranes near the sites of C4 activation. The presence of C4d in peritubular capillaries is a key indicator for acute humoral (i.e. antibody-mediated) rejection of kidney, heart, pancreas and lung allografts. As an established marker of antibody-mediated acute renal allograft rejection and its proclivity for endothelium, this component can be detected in peritubular capillaries in chronic renal allograft rejection as well as hyperacute rejection, acute vascular rejection, acute cellular rejection, and borderline rejection. C4d has been shown to be a significant predictor of transplant kidney graft survival. C4d antibody, combined with antibody to C3d, can be utilized as a tool for diagnosis of allograft rejection that may warrant a prompt and aggressive anti-rejection treatment.

MEK6 is a member of MAPKK protein kinase family. By using degenerate oligonucleotide primers from the conserved kinase domains of MKK3 and MKK4 two human cDNAs and 1 murine cDNA encoding closely related proteins of the MKK family were cloned. The two human clones appear to be different isoforms of the same gene generated by differential splicing: the shorter clone was designated MKK6, encodes a 278-amino acid protein, while the longer clone, designated MKK6b, encodes a 334-amino acid protein. MKK6 is about 80% identical to MKK3 and 40% identical to MKK4. 1.7-kb human MKK6 transcript is highly expressed in skeletal muscle, while an MKK6b-specific probe detected mRNA bands of 1.8, 2.4, and 4.5 kb that are enriched in heart, skeletal muscle, pancreas and liver. MKK6 plays an important role in intracellular signaling pathways leading toward activation of the p38 MAP kinase. MEK6 phosphorylates and activates p38 in response to inflammatory cytokines or environmental stress. As an essential component of p38 MAPK mediated signal transduction pathway, this gene is involved in many cellular processes such as stress induced cell cycle arrest, transcription activation and apoptosis.

Cadherin-6 (CDH6) is a type-II classic cadherin cell-cell adhesion molecules, which are expressed in graded or areal patterns, as well as layer-specific patterns, in the cortical plate. Human Cadherin-6 is synthesized as a 790 aa type I transmembrane glycoprotein that contains a 18 aa signal peptide, a 35 aa propeptide, a 562 aa extracellular region, a 21 aa transmembrane segment, and a 154 aa cytoplasmic domain. There are five cadherin domains of approximately 110 aa each in the extracellular region. Cadherin-6 has high expression in kidney, brain, and cerebellum, and may contribute to the formation of the segmental structure of the early brain, as well as the development of renal proximal tubules. Weak expression is also detected in lung, pancreas, gastric mucosa and cytotrophoblasts. As a classic cadherin, Cadherin-6 will form homodimers and promote intercellular adhesion with itself and, possibly, Cadherin-9 and -14.

Recognises a protein of 42-46 kDa, identified as MAGE-1. This mAb does not cross-react with MAGE-2, -3, -4, -6 -9, -10, -or -12 protein. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8+ T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). Because MAGE-1 is expressed on a significant proportion of human neoplasms of various histological types (melanoma, brain tumors of glial origin, neuroblastoma, non-small cell lung cancer, breast, gastric, colorectal, ovarian, renal cell carcinomas) and not on normal tissues, the encoded antigen may serve as a marker of early detection and target for cancer immunotherapy.

LRFN5 Antibody: LRFN5 is one of a family of five transmembrane glycoproteins that are highly expressed in neuronal tissues. LRFN proteins share leucine-rich repeat (LRR)-immunoglobulin-like (Ig)-fibronectin type III (Fn)-transmembrane domain structure with other members of the LRR-Ig-Fn protein superfamily such as the Slitrk family of proteins. Expression of LRFN1, -3, and -4 mRNA was detected in embryonic neuronal cells, while LRFN2 and LRFN5 expression was primarily restricted to more mature cells. LRFN1, -2, and -4 bound to PDZ domains of postsynaptic PSD95, re-distributing PSD95 to the cell periphery. It has been suggested that the Lrfn proteins play a role in the developing and/or mature vertebrate nervous system.

LGP2 Antibody: Anti-viral innate immune responses are triggered by pathogen-associated molecular patters (such as the accumulation of intracellular nucleic acids resulting from virus infections) and represent the first line of defense against numerous infectious organisms. The Toll-like receptors (TLR) 3, 7, 8, and 9 are expressed in immune cells and function as transmembrane pattern recognition receptors to detect foreign nucleic acids. Other proteins that play similar roles, such as RIG-1 and MDA5 are members of a CARD-helicase family and are expressed in the cytoplasm. A third protein, LGP2, is similar to RIG-1 and MDA5, except for lacking a homologous CARD domain. It is thought to act as an element of negative-feedback regulation of intracellular antiviral signaling. At least four isoforms of LGP2 are known to exist.

CEA is synthesised during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This antibody does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. CEA antibody is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are positive) from pleural mesotheliomas (rarely or weakly positive). CEA antibody positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

It plays a central role in platelet activation and aggregation. The CD51/CD61 is implicated in tumor metastasis and adenoviral infection. The M1/70 monoclonal antibody specifically reacts with the 170 kDa alpha M integrin chain of mouse CD11b from the Mac-1 integrin (CD11b/CD18). Mac-1 binds to C3bi, CD54 (ICAM-1), and fibrinogen, and it is expressed by granulocytes, macrophages, NK cells, myeloid-derived dendritic cells, microglia, activated lymphocytes, and mouse B-1 cells. The expression is up-regulated on activated neutrophils at the same time that L-selectin is shed from the cell surface. This mAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. It recognises human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.
