Description:
The EGFR subfamily of receptor tyrosine kinases is composed of EGFR, ErbB2, ErbB3 and ErbB4. The EGFR shares 43% - 44% aa sequence identity with the ECD of human EGFR subfamily. All these family members are type I transmembrane glycoproteins with an extracellular ligand binding domain. The extracellular ligand binding domain is containing two cysteine-rich domains separated by a spacer region and a cytoplasmic domain containing a membrane-proximal tyrosine kinase domain. Ligand binding could induce EGFR homodimerisation and heterodimerisation with ErbB2, resulting in cell signalling, heterodimerisation tyrosine phosphorylation and kinase activation. It can bind EGF, amphiregulin, TGF-alpha, betacellulin, epiregulin, HB-EGF, epigen, and so on. Its signalling regulates multiple biological functions including cell proliferation, differentiation, motility, and apoptosis. EGFR can also be recruited to form heterodimers with the ligand-activated ErbB3 or ErbB4. EGFR is overexpressed in different tumours. Several anti-cancer drugs use EGFR as target.

Pigment epithelium-derived factor (PEDF) is a 47 kDa secreted glycoprotein that belongs to the non-inhibitory serpin family group. PEDF is widely expressed in adult and foetal tissues, including brain, spinal cord, plasma, bone, prostate, pancreas, heart and lung. PEDF acts as an angiogenesis inhibitor with neurotrophic, immunomodulation and antitumor properties. It functions as anti-angiogenic agent by counterbalancing the proangiogenic effect of VEGF. PEDF is one of the most abundant proteins released by adipocytes and induces insulin resistance in adipocytes and human skeletal muscle cells. Recently, it has been reported that PEDF is sufficient to maintain the self-renewal of pluripotent human embryonic stem cells.

Interleukin-2 receptor subunit beta (IL2RB) is also known as CD122, which Belongs to the type I cytokine receptor family. Type 4 subfamily. IL2RB contains 1 fibronectin type-III domain. IL2RB / CD122 is involved in T cell-mediated immune responses, is present in 3 forms with respect to ability to bind interleukin 2. Ephrin type-B receptor 2(EPHB2) belongs to the protein kinase superfamily and Ephrin receptor subfamily. EPHB2 contains 1 Eph LBD domain, 2 fibronectin type-III domains, 1 protein kinase domain and 1 SAM domain. Ephrin receptors and their ligands, the ephrins, mediate numerous developmental processes, particularly in the nervous system. Based on their structures and sequence relationships, ephrins are divided into the ephrin-A (EFNA) class, which are anchored to the membrane by a glycosylphosphatidylinositol linkage, and the ephrin-B (EFNB) class, which are transmembrane proteins. HMGB1 was originally discovered as an essential DNA-binding protein for regulating p53, NF-kappaB and other important proteins. It is secreted from activated dentric cells, macrophage and nectrotic cells, and acts as a ligand for RAGE, TLR-2 and TLR-4 expressed on surrounding cells. As a result, HMGB1 activates Rac, CDC42 and NF-kappaB inducing localised innate immunity of damaged tissue, tissue regeneration by recruitment of stem cells and hemostasis by induction of tissue factor expression. HMGB1 is also causative agent of various diseases as it causes localised inflammation such as arteriosclerosis, chronic rheumatoid arthritis and nephritis.
