Cell surface glycoprotein CD200 Receptor 1 (CD200R1) is the receptor for the CD200 (OX-2) membrane glycoprotein. CD200R1 contains one C2- type Ig-like domain and one V-type Ig-like domain within its extracellular domain and a PTB-signaling motif in cytoplasmic domain. CD200R1 and CD200 associate via their respective N-terminal Ig-like domains. CD200R1 is restricted primarily to mast cells, basophils, macrophages, and dendritic cells. It propagates inhibitory signals despite its lacking a cytoplasmic ITIM (immunoreceptor tyrosinebased inhibitory motif). The receptor-substrate interaction may function as a myeloid downregulatory signal.

Tumor necrosis factor receptor superfamily member 12A(TNFRSF12A) is also known as Fibroblast growth factor-inducible immediate-early response protein 14, FN14, CD266 antigen and tweak-receptor. TNFRSF12A is a single-pass type I membrane protein, including a 27 aa signal peptide, a 53 aa extracellular domain, a 21 aa transmembrane domain and a 28 aa cytoplasmic domain. TNFRSF12A is highly expressed in heart, placenta and kidney. TNFRSF12A can be induced by FGF1 and phorbol ester. TNFRSF12A binds to TWEAK/TNFSF12A to initiate a signal transduction cascade, causing different cellular responses such as cell death, cell proliferation, and angiogenesis.

Cxcl1, also called Gro, Gro1, Mgsa or Scyb1, is short for growth-regulated alpha protein. The protein belongs to the intercrine alpha (chemokine CxC) family. The N-terminal processed form KC(5-72) of the protein is produced by proteolytic cleavage after secretion from bone marrow stromal cells, and shows a highly enhanced hematopoietic activity. Cxcl1 has chemotactic activity for neutrophils, and contributes to neutrophil activation during inflammation. Hematoregulatory chemokine, in vitro, suppresses hematopoietic progenitor cell proliferation.

Complement regulatory protein CD46 is also known as membrane Cofactor Protein (MCP), is a type I membrane protein and is a regulatory part of the complement system. CD46 is expressed by all cells except erythrocytes. MCP acts as a cofactor for complement factor I, a serine protease which protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue, and also acts as a costimulatory factor for T-cells which induces the differentiation of CD4+ into T-regulatory 1 cells. In T-cells by binding to CD46, A number of viral and bacterial pathogens seem to exploit this property and directly induce an immunosuppressive phenotype. Defects in CD46 are a cause of susceptibility to hemolytic uremic syndrome atypical type 2 (AHUS2).

Leukocyte differentiation antigen CD84 is also known as SLAM family member 5 (SLAMF5), which belongs to immunoglobulin (Ig) superfamily. CD84 / SLAMF5 contains one Ig-like C2-type (immunoglobulin-like) domain. CD84 plays a role as adhesion receptor functioning by homophilic interactions and by clustering. CD84 / SLAMF5 increases proliferative responses of activated T-cells and SH2D1A/SAP does not seen be required for this process. Homophilic interactions enhance interferon gamma/IFNG secretion in lymphocytes and induce platelet stimulation via a SH2D1A/SAP-dependent pathway.

LMIR1, also termed CD300a, is a type I transmembrane glycoprotein with a single IgV-like extracellular domain and an extended membrane proximal region that links the immunoglobulin (Ig) and transmembrane domains and belongs to the immunoglobulin superfamily. The intracellular domain of LMIR1 contains several immunoreceptor tyrosine-based inhibition motifs (ITIMs). When cross-linked, it will be tyrosine phosphorylated and capable of recruiting tyrosine phosphatases (SHP-1, SHP-2) and inositol polyphosphate 5-phosphatase, SHIP. LMIR1 will regulate mast cell-mediated inflammatory responses. LMIR1 is broadly expressed on myeloid and lymphoid cells, and its expression is differentially regulated depending on the cell type.
