TNF-related apoptosis-inducing ligand (TRAIL; Apo2L;CD253; TNFSF10) is a type II transmembrane protein of about 34kDa. Like most members of the tumor necrosis factor (TNF) superfamily of cytokines TRAIL can be cleaved at the cell surface by metalloproteases to form a soluble molecule. Active TRAIL forms trimers and specifically binds to five distinct receptors: TRAIL-R1 (DR4; Apo2;CD261; TNFRSF10A), TRAIL-R2 (DR5; KILLER; TRICK2A;TRICK2B; CD262; TNFRSF10B), TRAIL-R3 (DcR1;LIT; TRID; CD263; TNFRSF10C), TRAIL-R4 (DcR2; TRUNDD; CD264; TNFRSF10D), and osteoprotegerin (OPG; OCIF; TNFRSF11B). Trimerised TRAIL triggers apoptosis upon ligation of cell surface TRAIL-R1 and/or TRAIL-R2 by inducing the formation of the so-called multiprotein death-inducing signalling complex (DISC).

Description:
PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signalling pathway leading to the reparation of DNA strand breaks. PARP-1 positively regulates the transcription of MTUS1 and negatively regulates the transcription of MTUS2/TIP150. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production. PARP-1 (E998K mutant) is an inactive form of PARP-1 which can be used as a control compound.

IFNGR1 is phosphorylated at Ser/Thr residues after translation. IFNGR1 is a receptor for interferon gamma, two receptors bind one interferon gama dimer. A genetic variation in IFNGR1 is associated with susceptibility to Helicobacter pylori infection. In addition, defects in IFNGR1 are a cause of mendelian susceptibility to mycobacterial disease, also known as familial disseminated atypical mycobacterial infection.

Description:
Annexin V is an anticoagulant protein that acts as an indirect inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade. Annexin V competes for phosphatidylserine binding sites (e. g. platelets) with prothrombin or coagulation factors and inhibits the activity of phospholipase A1. Antibodies directed against annexin V are found in patients with a disease called the antiphospholipid syndrome(APS), a thrombophilic disease associated with autoantibodies against phospholipid compounds. Annexin V is used as a probe in the 'annexin V affinity assay' to detect cells that have expressed phosphatidylserine on the cell surface, a feature found in apoptosis and other forms of cell death.

BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases such as Sjoegren syndrome, Rheumatoid arthritis (RA), Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE). BAFF has also increased levels in some lymphoid cancers.

Interleukin 23 receptor (IL23R) is a type I cytokine receptor for IL23. IL23 receptor complex is comprised of two subunits, the IL12R beta 1 subunit, which is shared with several cytokines, and a subunit that is unique to IL-23. IL23, after binding to IL23R, activates memory T cells and mediates pro-inflammatory activities in part by the production of IL17 through activation of TH17 lymphocytes. IL23R is expressed on T cells, NK cells, dendritic cells, and macrophages. In fact, polymorphisms of the IL23R gene were reported to be associated with susceptibility to inflammatory diseases and autoimmune diseases such as psoriasis, multiple sclerosis, Graves's ophtalmopathy and inflammatory bowel diseases. The IL23R is known to be critically involved in the carcinogenesis of different malignant tumour.

Colony stimulating factor 1 receptor (CSF1R) is also known as macrophage colony-stimulating factor receptor (M-CSFR), CD115 Cluster of Differentiation 115 (CD115), C-FMS, CSFR, FIM2, FMS, and is a member of the typeIII subfamily of receptor tyrosine kinases (RTKs). CSF1R is a receptor for a cytokine called colony stimulating factor 1, The protein encoded by the CSFR1 gene is the receptor for colony stimulating factor 1, a cytokine which controls the production, differentiation, and function of macrophages. This receptor mediates most, if not all, of the biological effects of this cytokine. Ligand binding activates CSFR1 through a process of oligomerisation and transphosphorylation . Mutations in CSF1R are associated with chronic myelomonocytic leukemia and type M4 acute myeloblastic leukemia. Increased levels of CSF1R1 are found in microglia in Alzheimer's disease and after brain injuries. The increased receptor expression causes microglia to become more active. Both CSF1R, and its ligand colony stimulating factor 1 play an important role in the development of the mammary gland and may be involved in the process of mammary gland carcinogenesis.
