Alpha-Synuclein (SNCA) is a member of the Synuclein family. SNCA is expressed principally in brain but also expressed in low concentrations in all tissues except liver. SNCA interacts with UCHL1, Phospholipase D and histones. SNCA can include beta- and gamma-synuclein. In addition, SNCA is an important regulatory component of vesicular transport in neuronal cells. It has been suggested that SNCA is related to the pathogenesis of Parkinson's Disease and neurodegenerative disorders. Defects in SNCA will lead to Dementia Lewy Body (DLB).

SERPINI2 is a secreted protein of the serpin family. Serpins are a group of proteins with similar structures that were first identified as a set of proteins able to inhibit proteases. As protease inhibitors, serpins have an array of functions including regulating blood coagulation, fibrinolysis, the complement pathway, angiogenesis, inflammation, tumour suppression, extracellular matrix remodelling, and cell motility. SERPINI2 is expressed in human tissues including pancreas and adipose tissues. Mutations of human SERPINI2 can directly result in conditions such as acinar cell apoptosis and malabsorption.

Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) are closely related secreted ligands which bind with similar affinity to Tie-2. Tie-2 and angiopoietins have been shown to play critical roles in embryogenic angiogenesis and in maintaining the integrity of the adult vasculature. Ang-1 activates Tie-2 signalling on endothelial cells to promote chemotaxis, cell survival, cell sprouting, vessel growth and stabilisation. Ang-2 has been identified as a secreted protein ligand of Tie-2 and has alternatively been reported to be an antagonist for Ang-1 induced Tie-2 signalling as well as an agonist for Tie-2 signalling, depending on the cell context.

Description:
TLR11, an innate sensor for profilin (Toxoplasma gondii), is an intracellular receptor that resides in the endoplasmic reticulum. The 12 membrane-spanning endoplasmic reticulum-resident protein UNC93B1 interacts directly with TLR11 and regulates the activation of dendritic cells in response to profilin (Toxoplasma gondii) and parasitic infection in vivo. TLR12, a previously uncharacterised TLR, also recognises profilin from Toxoplasma gondii. TLR12 is sufficient for recognition of profilin by plasmacytoid dendritic cells (pDCs), whereas TLR11 and TLR12 are both required in macrophages and conventional DCs. In contrast to TLR11, TLR12-deficient mice succumb rapidly to T. gondii infection.

IL-36alpha (IL-1F6), IL-36beta (IL-1F8) and IL-36gamma (IL-1F9) bind to IL-36R (IL-1Rrp2) and IL-1RAcP, activating similar intracellular signals as IL-1. IL-36Ra inhibits the production of proinflammatory cytokines, including IL-12, IL-1beta, IL-6, TNF-alpha and IL-23 induced by IL-36 in BMDC and CD4 T cells. Skin and dendritic cells are targets of the IL-36 interleukins leading to a Th1 response. These cytokines may represent potential targets for immune-mediated inflammatory conditions or, alternatively, could be used as adjuvants in vaccination.

Irisin activates beige fat cells (beige cells have a gene expression pattern distinct from either white or brown fat and are preferentially sensitive to the polypeptide hormone irisin). Irisin is cleaved from the type I membrane protein FNDC5 and improves systemic metabolism by increasing energy expenditure.

Description:
Stromal Cell-Derived Factor-1 (SDF-1) is a chemokine member of the intercrine family. SDF1 is expressed as five isoforms that differ only in the C terminal tail. SDF1 alpha and SDF1 beta are identical except for the four residues present in the C-terminus of SDF1 beta but absent from SDF1 alpha. SDF1 isoforms interact with CXCR4 and CXCR7 receptors on the cell surface, and can also bind syndecan4. SDF1 is known to influence lymphopoiesis, regulate patterning and cell number of neural progenitors, and promote angiogenesis. It also enhances the survival of myeloid progenitor cells.

CD14 is a cell surface glycoprotein that is preferentially expressed on monocytes/macrophages. CD14 is anchored to cells by linkage to glycosylphosphatidylinositol (GPI) and functions as a pattern recognition receptor that binds lipopolysaccharides (LPS) and a variety of ligands derived from different microbial sources. The binding of CD14 with LPS is catalyzed by LPS binding protein (LBP). Toll like receptors have also been implicated in the transduction of CD14-LPS signals. Soluble CD14 can be released from the cell surface by phosphatidyinositolspecific phospholipase C and has been detected in serum and body fluids. High concentrations of soluble CD14 have been shown to inhibit LPS mediated responses. However, soluble CD14 can also potentiate LPS response in cells that do not express cell surface CD14.

CD85 antigen-like family member J (CD85J) is also known as Leukocyte immunoglobulin-like receptor subfamily B member 1 (LILRB1 or LIR-1), Immunoglobulin-like transcript 2 (ILT-2), Monocyte/macrophage immunoglobulin-like receptor 7 (MIR7), which belongs to leukocyte immunoglobulin-like receptor (LIR) family. CD85J / LILRB1 Contains 4 Ig-like C2-type (immunoglobulin-like) domains. CD85J / LIR-1 is expressed predominantly on B-cells and monocytes. CD85J is receptor for class I MHC antigens and recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles and is also receptor for H301/UL18, a human cytomegalovirus class I MHC homolog. Ligand binding results in inhibitory signals and down-regulation of the immune response. CD85J / LILRB1 interaction with HLA-B or HLA-E leads to inhibition of the signal triggered by FCER1A and inhibits serotonin release. CD85J / LILRB1 inhibits FCGR1A-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions.

Carcinoembryonic antigen-related cell adhesion molecule 8 (CEACAM8) is also known as CD66b (Cluster of Differentiation 66b), CD66b, CD67, CGM6, NCA-95, and is one of seven human CEACAM family members within the immunoglobulin superfamily. CEACAM family members are a set of widely expressed proteins involved in several biological functions, including cell adhesion, migration, signal transduction, and the regulation of gene expression. Abnormal overexpression and downregulation of some CEACAMs have been described in tumor cells. In humans, CEACAMs include type I transmembrane proteins (CEACAM1, CEACAM3, and CEACAM4) and GPI-linked molecules (CEACAM5 through CEACAM8). There is no human CEACAM2. CEACAM8 is a single chain, two Ig-like C2-type (immunoglobulin-like) domains and one Ig-like V-type (immunoglobulin-like) domain. It is an activation marker for human granulocytes.
