Description:
Endothelial Differentiation-Related Factor 1 (EDF1) is a 148 amino acid transcriptional coactivator that contains 1 HTH cro/C1-type DNA-binding domain. It has been postulated that the protein functions as a bridging molecule that interconnects regulatory proteins and the basal transcriptional machinery, thereby modulating the transcription of genes involved in endothelial differentiation. When endothelial cells are induced to differentiate in vitro, EDF1 is downregulated, leading to inhibition of cell growth and cell polarization. EDF1 binds calmodulin thorough its IQ domain and regulates nitric oxide synthase activity through calmodulin sequestration in the cytoplasm. Though ubiquitously expressed, EDF1 is most abundant in adult liver, heart, adipose tissues, intestine and pancreas. In fetal tissues, EDF1 is most abundant in kidney. There are two isoforms of EDF1 that are produced as a result of alternative splicing events.

Description:
Chloride Intracellular Channel Protein 4 (CLIC4) is a 253 amino acid single-pass membrane protein that localizes to both the nucleus and the cytoplasm and contains one GST C-terminal domain. CLIC4 is expressed in various tissues and exhibits an intracellular vesicular pattern in Panc-1 cells (pancreatic cancer cells). CLIC4 acts as a monomer which is able to form selective ion channels in target proteins, thus facilitating the transport of chloride and other ions. CLIC4 is believed to have a role in apoptosis and is able to translocate to the nucleus under stress conditions. CLIC4 has alternate cellular functions like a potential role in angiogenesis or in maintaining apical-basolateral membrane polarity during mitosis and cytokinesis.

4-Hydroxyphenylpyruvate Dioxygenase (4HPPD) belongs to the 4HPPD family. 4HPPD is a key enzyme in the degradation of tyrosine, which catalyzes the second reaction in the catabolism of tyrosine the conversation of 4-hydroxyphenylpyruvate to homogentisate. 4HPPD exists in homodimer forms, which uses zinc as a cofactor to catalyze the third step in the conversion of L-phenylalanine to fumarate and acetoacetic acid. When the active 4HPPD enzyme concentration is low in the human body, it results in high levels of tyrosine concentration in the blood, which can cause mild mental retardation at birth, and degradation in vision as a patient grows older.

The ST6GAL1 gene encodes beta -Galactosamide alpha-2,6-Sialyltransferase 1. It is a type II membrane protein and is localized to the trans-Golgi network. It catalyzes 2,6-sialylation of Gal beta 1,4-GlcNAc structures on N-glycans. ST6GAL1 is highly expressed in the liver and other tissues. ST6GAL1 deficiency causes abnormalities in B-cell immunoreactivity. The expression and activity of ST6GAL1 are associated with tumor metastasis in breast and colon cancers. The majority of ST6GAL1 in the liver is cleaved and secreted into the serum and may be used as a biomarker for hepatitis diseases.

Immunoglobulin alpha Fc Receptor (IgA Fc Receptor) is a member of the immunoglobulin gene superfamily. It is a transmembrane glycoprotein present on the surface of myeloid lineage cells such as neutrophils, monocytes, macrophages, and eosinophils, where it mediates immunologic responses to pathogens through the charged arginin residue within its transmembrane domain. IgA Fc Receptor binds both IgA1 and IgA2 with similar affinity. The site of interaction between FCAR and IgA was identified in the first extracellular domain of FCAR and the C2/C3 junction of IgA. It interacts with IgA-opsonized targets and triggers several immunologic defense processes, including phagocytosis, antibody-dependent cell-mediated cytotoxicity, and stimulation of the release of inflammatory mediators. FCAR is also expressed on Kupffer cells in the liver, where it was suggested to provide a second line of defense.

Osteoactivin is an intracellular glycoprotein belongs to the NMB/pMEL-17 family, which is asscociated with cell endosomal/lysomal compartments. Human Osteoactivin is a 560 amino acid type I transmembrane protein, and one alternate splice form shows a 12 amino acid insert between amino acid 339-340. An additional 206 amino acid isoform shows a mutation at position 181 that results in a 26 amino acid substitution for the C-terminal 380 amino acids. Cells knowns to express Osteoactivin include fibroblast, osteoblasts, myeloid dendritic cell, melanocytes, plus fetal chondrocytes and stratum basale keratinocytes, macrophages/keratinocytes.

Mouse CD83 is a 30-35kDa member of the Siglec (or sialic-acid-binding immunoglobulin-like lectin) family of transmembrane proteins. CD83 is a primary marker for dendritic cells. It is also found on B cells, neutrophils, monocytes and macrophages. Except for dendritic cells, CD83 expression is often transient. CD83 binds to sialic acids on target cells. Membrane CD83 promotes T cell proliferation, particularly of CD8+ cytotoxic T cells. Soluble CD83 is immunosuppressive and blocks T cell activation. On monocytes, CD83 is suggested to drive monocytes into a fibrocyte phenotype. A lack of membrane-expressed CD83 leads to an unusual IL-4/ IL-10 producing CD4+ T cell phenotype.

Inducible Co-Stimulator Ligand (ICOSLG) belongs to the immunoglobulin superfamily. ICOSLG contains Ig-like C2-type (immunoglobulin-like) domain and 1 Ig-like V-type (immunoglobulin-like) domain. CD68 is a single-pass type I membrane protein which belongs to the LAMP family. CD68 is highly expressed by blood monocytes and tissue macrophages. It is also expressed in lymphocytes, fibroblasts and endothelial cells and in many tumour cell lines which could allow them to attach to selectins on vascular endothelium, facilitating their dissemination to secondary sites. CD68 plays a role in phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions. It is a commonly used marker for macrophages. However, a number of studies have shown that CD68 antibodies react with other hematopoietic and non-hematopoietic cell types, suggesting that CD68 may not be a macrophage-specific antigen. CD68 binds to tissue- and organ-specific lectins or selectins, allowing homing of macrophage subsets to particular sites. Rapid recirculation of CD68 from endosomes and lysosomes to the plasma membrane may allow macrophages to crawl over selectin-bearing substrates or other cells.

Human KLK1 precursor contains a singal peptide (residues 1 to 18), a short pro peptide (residues 19 to 24) and a mature chain (residues 25 to 262). CD152 [CTLA-4] and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4 + and CD8 + T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4 + T cells and 50% of CD8 + T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.
