Description:
NOV, also called CCN3, is a secreted protein of CCN family members. CCN family members are highly conserved cysteine rich proteins sharing a common modular structure having 4 conserved domains, insulin-like growth factor-binding protein (IGFBP) domain, von Willebrand type C (VWC) domain, thrombospondin-1 (TSP-1) domain, and C-terminal (CT) domain (absent in CCN5). By specific interactions with these domains, CCN proteins modulate multiple signalling pathways including BMPs, Wnt, TGFs, Notch and integrins to regulate cell proliferation, differentiation, adhesion, migration, angiogenesis, and survival. CCN3 is firstly characterised as a promoter of progenitor activity of human hematopoietic stem cells, as knockdown of CCN3 can abrogate the function of primitive progenitors. Recent studies showed that CCN3 is also actively involved in the process of wound healing. CCN3 is highly expressed in granulation tissues of cutaneous wounds and capable of inducing synthetic responses of fibroblasts.

IL-3 is a hematopoietic growth factor that promotes the survival, differentiation and proliferation of committed progenitor cells of the megakaryocyte, granulocyte-macrophage, erythroid, eosinophil, basophil and mast cell lineages. Produced by T cells, mast cells and eosinophils, IL-3 enhances thrombopoieses, phagocytosis, and antibody-mediated cellular cytotoxicity. Its ability to activate monocytes suggests that IL-3 may have additional immunoregulatory roles. Many of the IL-3 activities depend upon co-stimulation with other cytokines. IL-3 is species-specific, variably glycosylated cytokine. Recombinant murine IL-3 is a 15.1 kDa globular protein containing 135 amino acid residues. Recombinant human IL-3 is a 15.0 kDa globular protein containing 133 amino acid residues.

IL-16 was initially identified as a chemotactic cytokine (1), but is now known to possess a wide range of activities. Later studies have more fully characterized IL-16 as an immunomodulatory cytokine that contributes to the regulatory process of CD4+ T cell recruitment and activation at sites of inflammation in association with asthma and several autoimmune diseases (2). The precursor of IL-16 (pro-IL-16) is thought to be cleaved towards the C-terminal region by Caspase-3, releasing a 20 kDa active form that binds to and signals through CD4 (3,4). Besides acting as a chemotactic cytokine, IL-16 is thought to also be involved in the regulation of T cell proliferation (5) and multiple infectious, immune-mediated, and autoimmune inflammatory disorders including irritable bowel syndrome, systemic lupus erythematosus, and neurodegenerative disorders (6). At least two isoforms of IL-16 are known to exist; the longer isoform (also known as NIL-16) is detected only in neurons of the cerebellum and hippocampus (7). This recombinant protein fragment is derrived from this longer isoform.

The Interferon- alpha/ beta Receptor 1 (IFN- alpha/ beta R1) is a receptor which binds Type I Interferons including Interferon- alpha and - beta . It is a cell surface receptor and heteromeric receptor composed of one chain with two subunits referred to as IFNAR1 and IFNAR2. IFN- alpha/ beta R1, in association with IFN- alpha/ beta R2, is required for propagating antiviral signal transduction triggered by IFN- alpha and IFN- beta . IFN- alpha/ beta R1 interacts very weakly or not at all with type 1 interferons and does not stably interact with IFN- alpha/ beta R2. Ligands associate with IFN- alpha/ beta R2, and this complex subsequently forms a stable ternary assembly with IFN- alpha/ beta R1. IFN- alpha/ beta R1 also associates with IFN- gamma R2 even in the absence of IFN- gamma stimulation. Human IFN- alpha/ beta R1 contains a nuclear localization signal in its extracellular domain that is required for receptor translocation to the nucleus following interaction with ligand. Interferon stimulation results in an immunologic response that is especially associated with viruses.

BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases such as Sjoegren syndrome, Rheumatoid arthritis (RA), Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE). BAFF has also increased levels in some lymphoid cancers.
