Hyccin is a 521 amino acid cytoplasmic protein that is widely expressed with highest levels found in heart, brain, placenta, spleen and testis. Belonging to the FAM126 family, hyccin may play a role in the ∫-catenin/Lef signaling pathway. Hyccin is likely involved in the process of myelination of the central and peripheral nervous system. Defects in the gene encoding hyccin are the cause of leukodystrophy hypomyelinating type 5 (HLD5), which is characterized by congenital cataract, progressive neurologic impairment and diffuse myelin deficiency. Individuals affected by HLD5 experience progressive pyramidal and cerebellar dysfunction along with muscle weakness in the lower limbs. Hyccin exists as two alternatively spliced isoforms and is encoded by a gene located on human chromosome 7.

Emx1 and Emx2 are human homologs to the Drosophila developmental genes empty spiracles expressed in anterior body regions during early Drosophila embryogenesis. Emx1 and Emx2 are homeobox proteins expressed in the developing vertebrate brain. Emx2 is expressed in the dorsal telencephalon and small diencephalic regions, while Emx1 expression is exclusively confined to pyramidal neurons of the dorsal telencephalon. In the embryonic brain, Emx1 is expressed in both proliferating and differentiating neurons while Emx2 is expressed only in proliferating neurons. OTX1 and OTX2 are human homologs of the Drosophila developmental genes orthodenticle. In development, the sequence of expression begins with OTX2 at day ten post coitum followed by OTX1, Emx2 and finally Emx1. The genes encoding human Emx1 and Emx2 map to chromosomes 2p13.2 and 10q26.11, respectively.

Serving as an adaptor or scaffold molecule, beta Arrestin 1 is essential for mitogenic signaling. It mediates agonist dependent desensitization and internalization of G protein coupled receptors (GPCRs, e.g., beta 2 adrenergic receptor). After binding to their ligand and interacting with heterotrimeric G proteins, GPCRs are phosphorylated by G protein receptor kinases (GRKs) on serine residues. Beta Arrestin 1 has important roles in the cytoplasm and at the plasma membrane in the desensitization and internalization of G protein coupled receptors (GPCRs) and is increasingly appreciated to play an important role in the endocytosis and signaling of GPCRs. Beta Arrestin 1 in the cytosol is phosphorylated by ERK1 and 2 on serine 412 in a negative feedback mechanism and binds to the phosphorylated receptors at the plasma membrane. Serine 412 is then dephosphorylated and the GPCRs are internalized, leading to activation of the Ras, Raf, ERK1 and 2 signaling pathway.
