STAT2 encoded by this gene is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. Ataxin-2 binding protein 1 (A2BP1) has an RNP motif that is highly conserved among RNA-binding proteins. This protein binds to the C-terminus of ataxin-2 and may contribute to the restricted pathology of spinocerebellar ataxia type 2 (SCA2). Ataxin-2 is the gene product of the SCA2 gene which causes familial neurodegenerative diseases. Ataxin-2 binding protein 1 and ataxin-2 are both localized in the trans-Golgi network. Twisted Gastrulation Protein Homolog 1 (TWSG1) is a 22 kDa secreted protein that belongs to the twisted gastrulation protein family. Human TWSG1 is synthesized as a 223 aa precursor that contains a 25 aa signal peptide and a 198 aa mature chain. TWSG1 may be involved in dorsoventral axis formation. TWSG1 seems to antagonize BMP signaling by forming ternary complexes with CHRD and BMPs, thereby preventing BMPs from binding to their receptors.TWSG1 can inhibit BMP activity by binding directly to BMP proteins, and can act the anti-BMP function, partly mediated by cleavage and degradation of CHRD, which releases BMPs from ternary complexes. TWSG1 may be an important modulator of BMP-regulated cartilage development, chondrocyte differentiation and thymocyte development.
