These proteins act through adaptor molecules such as MyD88 and TIRAP to activate various kinases and transcription factors. TLR7, like TLRs 3, 8, and 9, is localized in intracellular acidic compartments such as the phagolysosome and will recognize some single-stranded RNA viruses such as vesicular stomatitis virus (VSV) and influenza virus. TLR4, the major signaling receptor for lipopolysaccharide (LPS), requires the binding of MD-2 to its extracellular region for maximal response to LPS. The specificity of this response is determined by the species of MD-2; e.g., human MD-2 can cause mouse TLR4 to react to LPS analogs that are normally antagonistic to human but not mouse TLR4.

Anthrax Toxin Receptor 1 (ANTXR1) is a single-pass type I membrane protein that belongs to the ATR family. ANTXR1 contains one VWFA domain and binds PA through the VWA domain. ANTXR1 is highly expressed in tumor endothelial cells. ANTXR1 plays a role in cell attachment and migration. ANTXR1 interacts with extracellular matrix proteins and the actin cytoskeleton, it mediates adhesion of cells to type 1 collagen and gelatin, reorganization of the actin cytoskeleton and promotes cell spreading. DR4 Antibody: Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel death domain containing receptor was recently identified and designated DR4 (for death receptor 4). The ligand for this novel death receptor has been identified and termed TRAIL, which is a new member in the TNF family. DR4 is also called TRAIL receptor-1 (TRAIL-R1). DR4 is expressed in most of human tissues including spleen, peripheral blood leukocytes, small intestine and thymus. Like TNFR1, Fas and DR3, DR4 mediates apoptosis and NF-kappa B activation in many tissues and cells.

Cluster of Differentiation 31 (CD31) is also known as Platelet endothelial cell adhesion molecule (PECAM-1), is a 130-kDa transmembrane glycoprotein expressed by endothelial cells, platelets, macrophages and Kupffer cells, granulocytes, T / NK cells, lymphocytes, megakaryocytes, osteoclasts, neutrophils, certain tumors, and is the only known member of the CAM family on platelets. CD31 is found on the surface of platelets, monocytes, neutrophils, and some types of T-cells, and makes up a large portion of endothelial cell intercellular junctions. The encoded protein is a member of the immunoglobulin superfamily and is likely involved in leukocyte migration, angiogenesis, and integrin activation. CD31 plays a key role in removing aged neutrophils from the body. CD31 mediates the homotypic or heterotypic cell adhesion by binding to itself or the leukocyte integrin αvβ3, and thus plays a role in neutrophil recruitment in inflammatory responses, transendothelial migration of leukocytes, as well as in cardiovascular development. In addition, it has been shown that CD31 expression is up-regulated by LPS stimulation, and might function as a feedback negative regulator of LPS inflammatory response in macrophages.

Chemokine (C-C Motif) Ligand 28 (CCL28) is a novel chemokine that shares the most homology with CCL27/CTACK. CCL28 shows chemotactic activity for resting CD4, CD8 T-cells and eosinophils. It Binds to CCR3 and CCR10 and induces calcium mobilization in a dose-dependent manner. CCR10 (GPR2 orphan receptor) is also the receptor for CCL27/CTACK. Interleukin-16 (IL-16) is a CD8+ T cell-derived cytokine that induces chemotaxis of CD4+ T cells and CD4+ monocytes and eosinophils. Analysis by gel filtration suggests that, under physiological conditions, human IL-16 exists predominantly as a noncovalently linked multimer, but that some IL-16 may exist as a monomer. Tumor Necrosis Factor Ligand Superfamily Member 15 (TNFSF15) is a new member of the tumor necrosis factor family. TNFSF15 is predominantly an endothelial cell-specific gene, and recombinant TNFSF15 is a potent inhibitor of endothelial cell proliferation, angiogenesis and tumor growth. TNFSF15 exerts two activities on endothelial cells: early G1 arrest of G0/G1-cells responding to growth stimuli and programmed cell death of proliferating cells. These activities are highly specific to endothelial cells. TNFSF15 is also able to regulate the expression of several important genes involved in angiogenesis. These findings are consistent with the view that TNFSF15 functions as an autocrine cytokine to inhibit angiogenesis and stabilize the vasculature.
