Description:
Cadherin-11, also known as OSF-4, Osteoblast cadherin and CDH11, is a type II classical cadherin from the cadherin superfamily, integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Cadherins interact with themselves in a homophilic manner in connecting cells, may thus contribute to the sorting of heterogeneous cell types. Cadherin-11 contains five cadherin domains and is mainly expressed in brain. Mature cadherin proteins consists of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small, highly conserved C-terminal cytoplasmic domain. It is shown that cadherin-11 is a viable molecular target for therapeutic intervention in Glioblastoma multiforme.

CD27 (TNFRSF7) is a member of the TNF-receptor superfamily limited to cells of the lymphoid lineage and exists as both a dimeric glycoprotein on the cell surface and as a soluble protein in serum. As a T and B cell co-stimulatory molecule, the activity of CD27 is governed by its TNF-like ligand CD70 on lymphocytes and dendritic cells. The CD27-CD70 interaction is required for Th1 generation responses to differentiation signals and long-term maintenance of T cell immunity, and meanwhile, plays a key role in regulating B cell differentiation, activation and immunoglobulin synthesis. The CD27 receptor transduces signals and subsequently leads to the activation of NF-kappaB and MAPK8/JNK, mediated by the adaptor proteins TRAF2 and TRAF5. In addition, the proapoptotic protein SIVA is capable of binding the cytoplasmic tail of CD27 and exerts action in the process of apoptosis.

CCL4, (C-C motif chemokine 4; macrophage inflammatory protein 1beta) is a chemokine secreted at sites of inflammation by activated leukocytes, lymphocytes, vascular endothelial cells and pulmonary smooth cells. CCL4 attracts a variety of immune cells to sites of microbial infection as well as to pathologic inflammation, such as allergic asthma and ischemic myocardium. Full-length and truncated CCL4 exert biological activity through the receptor CCR5. The truncated form of CCL4 interacts additionally with CCR1 and CCR2b. The ability of CCL4 to bind CCR5 inhibits the cellular entry of M-tropic HIV. Both CCL4 forms block HIV-1 infection of T cells by inducing the downregulation of CCR5.

CD300A consists of a 163 amino acid (aa) extracellular domain (ECD) with one Ig-like V- type domain, a 21 amino acid transmembrane segment, and a 98 amino acid cytoplasmic domain with tyrosine residues. CD300A is expressed not only by natural killer (NK) cells but also by T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes and monocytes. CD300A is an inhibitory receptor which may contribute to the down-regulation of cytolytic activity in natural killer (NK) cells, and to the down-regulation of mast cell degranulation.

Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF) was initially characterised as a growth factorthat can support the in vitro colony formation of granulocyte-macrophage progenitors. It is produced by anumber of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cellsand fibroblasts) in response to cytokine of immune and inflammatory stimuli. Besides granulocyte-macrophageprogenitors, GM-CSF is also a growth factor for erythroid, megakaryocyte and eosinophil progenitors. Onmature hematopoietic, monocytes/ macrophages and eosinophils. GM-CSF has a functional role on nonhematopoitic cells. It can induce human endothelial cells to migrate and proliferate. Additionally, GM-CSF canalso stimulate the proliferation of a number of tumour cell lines, including osteogenic sarcoma, carcinoma andadenocarcinoma cell lines.

CD137 (4-1BB) is a member of the tumour necrosis factor (TNF) receptor family. CD137 is expressed by activated T cells, dendritic cells, NK cells, granulocytes and cells of blood vessel walls at sites of inflammation. CD137 has costimulatory activity for activated T cells. Crosslinking of CD137 enhances T cell proliferation, IL-2 secretion survival and cytolytic activity. CD137 was shown to enhance immune activity to eliminate tumours in mice.

CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulphide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

CD134 (OX40) is a T cell activation antigen structurally belonging to a lymphocyte-specific subgroup of the nerve growth factor and tumour necrosis factor receptor superfamily, which also includes the T cell antigen CD27, B cell antigen CD40, FAS antigen and the T cell activation antigen 4-1BB. The human CD134 protein is expressed only on activated CD4+ T blasts, and its ligand has been identified as gp34. Interactions between CD134 and its ligand in vivo are necessary for the differentiation of activated B cells into highly immunoglobulin-producing cells, however not involved in other pathways of antigen-driven differentiation of B cells such as development of memory cells in the germinal centers. In addition, the CD134 and gp34 system directly mediate adhesion of activated T cells to vascular endothelial cells, and contribute to growth stimulation of the virus-infected T cells.

FGF-basic is one of 23 known members of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. FGF-basic is a non-glycosylated heparin binding growth factor that is expressed in the brain, pituitary, kidney, retina, bone, testis, adrenal gland liver, monocytes, epithelial cells and endothelial cells. FGF-basic signals through FGFR 1b, 1c, 2c, 3c and 4. Recombinant human FGF-basic is a 17.2 kDa protein consisting of 154 amino acid residues. Recombinant murine FGF-basic is a 16.2 kDa protein consisting of 145 amino acid residues.
