Description:
UBE2M is a member of the E2 ubiquitin-conjugating enzyme family. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This protein is linked with a ubiquitin-like protein, NEDD8, which can be conjugated to cellular proteins, such as Cdc53/culin. UBE2M accepts the ubiquitin-like protein NEDD8 from the UBA3-NAE1 E1 complex and catalyses its covalent attachment to other proteins. The specific interaction with the E3 ubiquitin ligase RBX1, but not RBX2, suggests that the RBX1-UBE2M complex neddylates specific target proteins, such as CUL1, CUL2, CUL3 and CUL4. It involved in cell proliferation and is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation.

Calcitonin is a secreted protein which belongs to the calcitonin family. Calcitonin is cleaved into the following two chains: Calcitonin and Katacalcin. Katacalcin is a potent plasma calcium-lowering peptide. Calcitonin is a 32-amino acid linear polypeptide hormone. Calcitonin acts to reduce blood calcium (Ca2+), opposing the effects of parathyroid hormone (PTH). Its importance in humans has not been as well established as its importance in other animals, as its function is usually not significant in the regulation of normal calcium homeostasis. Calcitonin causes a rapid but short-lived drop in the level of calcium and phosphate in blood by promoting the incorporation of those ions in the bones.

Natural cytotoxicity triggering receptor 3 (NCR3) is also known as Activating natural killer receptor p30, Natural killer cell p30-related protein (NK-p30), CD antigen CD337, 1C7, LY117. NCR3 /CD337 /NKp30 belongs to the natural cytotoxicity receptor (NCR) family. NCR3 /NKp30 contains one Ig-like (immunoglobulin-like) domain. NCR3 /NKp30 is selectively expressed by all resting and activated NK cells and weakly expressed in spleen. NCR3 is homodimer in the unliganted form. NCR3 interacts with CD3Z. NCR3 interacts with and is activated by binding to NCR3LG1 or BAG6. Engagement of NCR3 by BAG6 also promotes dendritic cell (DC) maturation, both through killing those DCs that did not properly acquire a mature phenotype, and inducing NK cells to release TNFA and IFNG, which promotes DC maturation.

The protein specifically promotes pancreatic beta cell proliferation and beta cell mass expansion, thereby improving glucose tolerance. It promotes pancreatic beta cell proliferation without insulin resistance. Also it acts as a blood lipid regulator by regulating serum triglyceride levels and possibly by promoting ANGPTL3 cleavage. It interacts with ANGPTL3. It predominantly expressed in liver and also expressed in adipose tissues. The ability of the protein to induce pancreatic beta cell proliferation is promising in diabetes therapy. Betatrophin treatment could supply or replace insulin injections by increasing the number of insulin-producing cells in diabetes.

Junctional Adhesion Molecule B (JAM-B) is a single-pass type I membrane protein that belongs to the juctional adhesion molecules family. JAM-B includes a signal sequence (aa 1-28), an extracellular region (aa 29-238) with one Ig-like C2-type domain and one Ig-like V-type domain, a transmembrane segment (aa 239-259), and a cytoplasmic domain (aa 260 - 298). JAMB is localized to the tight junctions between endothelial cells or epithelial cells. JAM-B is prominently expressed in the heart, placenta, lung, foreskin and lymph node. It is also present on the endothelia of other vessels. JAM-B acts as an adhesive ligand for interacting with a variety of immune cell types and may play a role in lymphocyte homing to secondary lymphoid organs.
