Angiopoietin-related protein 4 (ANGPTL4) is also known as Hepatic fibrinogen / angiopoietin - related protein (HFARP), ARP4, PGAR. ANGPTL4 contains one fibrinogen C-terminal domain. ANGPTL4 is a homooligomer , and the homooligomer undergoes proteolytic processing to release its carboxyl fibrinogen-like domain, which circulates as a monomer. The homooligomer unprocessed form is able to interact with the extracellular matrix. ANGPTL4 may act as a regulator of angiogenesis and modulate tumorigenesis. ANGPTL4 inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. ANGPTL4 may exert a protective function on endothelial cells through an endocrine action.

IL-33 is structurally related to IL-1, which induces helper T cells to produce type 2 cytokines and acts through the receptor IL1RL-1. It is a 254 amino acids cytokine that is expressed in brain, placenta, lung, skeletal muscle and kidney. Kallikrein7, also named as stratum corneum chymotryptic enzyme (SCCE), is a secreted protein of the Kallikrein-related peptidase (KLK) family. This family contains fifteen homologous secreted serine endopeptidases and plays a significant role in various physiological processes, including skin desquamation, semen liquefaction, neural plasticity, and body fluid homeostasis. In skin KLK5, KLK 7 and KLK14 are able to degrade corneodesmosomes, which leads to desquamation of skin surface cells. KLK activation is believed to be mediated through highly organised proteolytic cascades, regulated through a series of feedback loops, inhibitors, auto-degradation and internal cleavages. Studies have shown that one potential physiological activator for KLK7 is KLK5. Along with KLK14, these three kallikreins form a proteolytic cascade in the stratum corneum. KLK7 is primarily expressed in the skin but is also detected at relatively high levels in oesophagus, heart, liver, central nervous system, kidney, pancreas, mammary and salivary glands.

Cadherin-3 (CDH3) is a single-pass type I membrane protein that belongs to the cadherin superfamily. CDH3 is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. CDH3 is expressed in some normal epithelial tissues and in some carcinoma cell lines. CDH3 preferentially interacts with themselves in a homophilic manner in connecting cells. CDH3 is involved in loss of heterosygosity events in breast and prostate cancer. Mutations in CDH3 have been associated with congential hypotrichosis with juvenile macular dystrophy.

Programmed Death-1 (PD-1), firstly cloned from mouse T cell hybridoma 2B4.11, is one member of CD28/CTLA-4 superfamily. PD-1 belongs to type I transmembrane protein and acts as an important immunosuppressive molecule. The cytoplamsic tail of PD-1 contains two structural motifs, an immunoreceptor tyrosine-based inhibitory motif (ITIM) and an immunoreceptor tyrosine-based switch motif (ITSM) formed by two tyrosine residues which make the difference in PD-1 signal mediating. Mouse PD-1 is expressed in thymus and shares about 69% aa sequence identity with human PD-1. Recently, programmed death-1 (PD-1) with its ligands, programmed death ligand B7H1 (PD-L1) and B7DC (PD-L2), was found to regulate T-cell activation and tolerance, upon ligand binding, inhibiting T-cell effector functions in an antigen-specific manner. PD-1 gene knocked out mice would induce some autoimmune diseases, which suggests that PD-1 acts as a co-inhibitory molecule actively participating in maintaining peripheral tolerance. Thus, PD-1 may be a useful target for the immunologic therapy of carcinoma,infection,autoimmune diseases as well as organ transplantation.

Anexin A5 is a phospholipid binding protein, which binds with high affinity and selectivity to PS in the presence of calcium. PS is predominantly located in membrane leaflets, which face the cytosol. However, recent findings show that each cell type has the molecular machinery to expose PS at its cell surface. This machinery is activated during the execution of apoptosis. Once PS is exposed at the cell surface it exhibits procoagulant and proinflammatory activities. Anexin A5 will bind to the PS-exposing apoptotic cell and can inhibit the procoagulant and proinflammatory activities of the dying cell. Anexin A5 has also been identified as an anticoagulant protein in the blood coagulation cascade, by acting as an inhibitor of prothrombin activation. The presence of antibodies to Anexin A5 is associated with systemic lupus erythematosus (SLE), recurrent spontaneous abortions and systemic sclerosis (SSc).

CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumour growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.

Interleukin-1 Receptor Accessory Protein (IL-1RAcP) is a member of the interleukin-1 receptor family. It contains three Ig-like C2-type domains in the extracellular region and a long cytoplasmic domain implicated in signal transduction. IL-1RAcP acts as a non-ligand binding accessory component of the receptors for IL1 alpha, IL1 beta and IL33. IL-1RAcP mediates interleukin-1-dependent activation of NF-kappa-B. It is part of the membrane-bound form of the IL-1 receptor. IL-1 RAcP takes part in the Signaling ways by the formation of a ternary complex containing IL1R1, TOLLIP, MYD88, and IRAK1 or IRAK2. In addition, IL-1RAcP modulates the response to interleukins by associating with soluble IL1R1 and enhancing interleukin-binding to the decoy receptor.
