Description:
Chemokine (C C Motif) Ligand 26 (CCL26) is a novel small cytokine belonging to the CC chemokine family, which involved in immunoregulatory and inflammatory processes. CCL26 is expressed constitutively in thymus, but only transiently in phytohemagglutinin-stimulated peripheral blood mononuclear cells. It specifically binds and induces chemotaxis in T cells and elicits its effects by interacting with the chemokine receptor CCR4. Eotaxin-3/CCL26, along with Eotaxin-1 and Eotaxin-2, selectively activates the CC chemokine receptor 3 (CCR3). The Eotaxin-3-CCR3 interaction may play an important role in allergic diseases such as atopic dermatitis and bronchial asthma. The full-length cDNA for Eotaxin-3 encodes a protein of 94 amino acids with a putative signal peptide of either 23 or 26 amino acid residues. Both the 71 and 68 amino acid residue variants of recombinant Eotaxin-3 demonstrate equal potency in inducing chemotaxis of a human CCR3-transfected cell line. Unlike most other CC chemokines, Eotaxin-3 maps to human chromosome 7q11.2, within 40 kilobases of the Eotaxin-2 loci. Eotaxin-3 and Eotaxin-2 are unique in that they are the only chemokines identified to date that map to chromosome 7.

Activin proteins that belong to the transforming growth factor-beta (TGF- beta ) superfamily, exert their biological actions by binding to heteromeric receptor complexes of type I and type II serine/threonine kinase receptors. Advanced Glycosylation End Product-Specific Receptor (AGER) belongs to the immunoglobulin superfamily of cell surface molecules. It lies within the major histocompatibility complex (MHC) class III region on chromosome 6. Besides AGEs, AGER is also able to bind other ligands which is thought to result in pro-inflammatory gene activation. It is known that AGER serve as a mediator of both acute and chronic vascular inflammation in certain conditions such as atherosclerosis and in particular as a complication of diabetes. Furthermore, it plays an important role in regulating the production/expression of TNF-alpha, oxidative stress, and endothelial dysfunction in type 2 diabetes.

Progranulin (PGRN) is a widely expressed pluripotent growth factor which plays a role in processes such as development, wound repair and inflammation by activating signaling cascades that control cell cycle progression and cell motility. Its function in the central nervous system is of interest, as mutations in the PGRN gene were found in cases of frontotemporal degeneration (FTLD). In addition, PGRN has also been linked to tumorigenesis. Progranulin is a biomarker for FTLD, other types of Alzheimer‘s Disease (AD) and potentially for MCI (Mild Cognitive Impairment). Additionally, PGRN is described as a new ligand of TNF receptors and a potential therapeutic against inflammatory disease like arthritis.

Tumour necrosis factor (TNF, cachexin or cachectin and formerly known as tumour necrosis factor-alpha) is a cytokine involved in systemic inflammation and is a member of a group of cytokines that stimulate the acute phase reaction. The receptor TNF-R1 is activated by both the membrane-bound and soluble trimeric forms of TNF-alpha, whereas the receptor TNF-R2 only responds to the membrane-bound form of TNF-alpha (MultimericTNF-alpha (Prod. No. AG-40B-0019). Since the MultimericTNF-alpha mimics the membrane-bound form (forms oligomers higher than trimer), it is the only TNF-alpha protein that can activate the TNF-R2. For TNF-R1 activation, either "normal" TNF-alpha or MultimericTNF-alpha can be used."

Semenogelin-1 (SEMG1) is the predominant protein in semen; it is a secretory protein involved in the formation of a gel matrix entrapping the accessory gland secretions and ejaculated spermatozoa. The prostate-specific antigen (PSA) protease processes SEMG1 into smaller peptides, each possibly having a separate function. In the proteolysis process, Alpha-inhibin-92 and alpha-inhibin-31 are produced; they inhibit the secretion of pituitary follicle-stimulating hormone. At the same time, it breaks down the gel matrix, allowing the spermatozoa to move more freely.

Lymphatic Vessel Endothelial Hyaluronic Acid Receptor 1 is a single-pass type I membrane protein. LYVE-1 is a CD44 homolog found primarily on lymphatic endothelial cells 1. LYVE-1 mainly expressed in endothelial cells lining lymphatic vessels. While LYVE-1 functions is a Ligand-specific transporter trafficking between intracellular organelles (TGN) and the plasma membrane. LYVE-1 plays a role in autocrine regulation of cell growth mediated by growth regulators containing cell surface retention sequence binding (CRS). It may act as an hyaluronan (HA) transporter, either mediating its uptake for catabolism within lymphatic endothelial cells themselves, or its transport into the lumen of afferent lymphatic vessels for subsequent re-uptake and degradation in lymph nodes.
