Interleukin-10 (IL-10) is a cytokine produced by activated Th2 cells, B cells, keratinocytes and monocytes/macrophages. In vitro murine and human IL-10 inhibits cytokine synthesis by Th1 cells, natural killer cells and monocytes/macrophages. Several studies have suggested the potential application of IL-10 as an anti-inflammatory agent in the treatment of septic shock and as an immunosuppressive agent in certain T cell mediated autoimmune diseases.

BAG Family Molecular Chaperone Regulator 2 (BAG2) is a member of the Bag family whose members compete with Hip for binding to the Hsc70/Hsp70 ATPase domain and promote substrate release. BAG2 contains 1 BAG domain and is a important component of the HSC 70/CHIP chaperone-dependent ubiquitin ligase complex. In mammalian cells BAG1, BAG2, and BAG3 bind with high affinity to the ATPase domain of Hsc70 and inhibit its chaperone activity in a Hip-repressible manner.

Description:
Lipocalin-2, also known as Neutrophil Gelatinase-Associated Lipocalin (NGAL), is a secretory protein of the lipocalin superfamily. Lipocalin-2 contains a signal peptide that enables it to be secreted and form complexes with matrix metalloproteinase-9 (MMP-9) through disulphide bonds. Similar to other lipocalin family members, Lipocalin-2 is involved in diverse cellular processes, including the transport of small hydrophobic molecules, protection of MMP-9 from proteolytic degradation, and cell signalling. Furthermore, Lipocalin-2 can tightly bind to bacterial siderophore through a cell surface receptor, possibly serving as a potent bacteriostatic agent by sequestering iron, regulating innate immunity and protecting kidney epithelial cells from ischemia–reperfusion injury. This protein is mainly expressed in neutrophils and in lower levels in the kidney, prostate, and epithelia of the respiratory and alimentary tracts. Recent evidence also suggests its role as a biomarker for renal injury and inflammation.

Leptin functions via signalling pathways involving OB-R in hypothalamus. In mammals, leptin is mainly produced by the adipose tissue and encodes body fat reserves, acting as a short-term satiety signal. In fish, the presence of a leptin-like peptide was first evidenced by immuno-cross-reactivity [14], and its existence was certainly demonstrated after the finding by synteny of a leptin sequence in the pufferfish.

CST7 is a secreted protein and primarily expressed in peripheral blood cells and spleen.It is belongs to the cystatin family. The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. This gene encodes a glycosylated cysteine protease inhibitor with a putative role in immune regulation through inhibition of a unique target in the hematopoietic system.

Legumain is a lysosomal cysteine protease which is a member of the peptidase C13 family. Though it is found in many tissues, it is highly expressed in the kidney, heart, and placenta. Legumain has a strict specificity for hydrolysis of asparaginyl bonds and can also cleave aspartyl bonds slowly, especially under acidic conditions. Over-expression Legumain in tumors is significant for invasion and metastasis. In addition, Legumain may be involved in the processing of proteins for MHC class II antigen presentation in the lysosomal/endosomal system and negative regulation of neuron apoptosis.

Matrix Metalloproteinase-1 (MMP-1) is expressed by fibroblasts, keratinocytes, endothelial cells, monocytes and macrophages. MMP1 contains several distinct domains: a prodomain that is cleaved upon activation, a catalytic domain containing the zinc binding site, a short hinge region, and a carboxyl terminal (hemopexin like) domain. MMP-1 can degrade a broad range of substrates including types I, II, III, VII, VIII, and X collagens as well as casein, gelatin, alpha1 antitrypsin, myelin basic protein, L-Selectin, pro-TNF, IL1, IGFBP3, IGFBP5, pro-MMP2, and pro-MMP9. A significant role of MMP1 is the degradation of fibrillar collagens in extracellular matrix remodeling, characterized by the cleavage of the interstitial collagen triple helix into 3/4, 1/4 fragments. MMP1 may also be involved in enzyme cascades, cytokine regulation and cell surface molecule modulation.

Inter-Cellular Adhesion Molecule 1 (ICAM1) is a type of intercellular adhesion molecule continuously present in low concentrations in the membranes of leukocytes and endothelial cells. As an endothelial and leukocyte-associated transmembrane protein, ICAM1 is well known for its importance in stabilizing cell-cell interactions and facilitating leukocyte endothelial transmigration. The presence of heavy glycosylation and other structural characteristics lend ICAM1 binding sites for a number of immune-associated ligands. Notably, ICAM-1 binds to macrophage adhesion ligand-1 (Mac-1; ITGB2 / ITGAM), leukocyte function associated antigen-1 (LFA-1/integrin), and fibrinogen.ICAM-1 expressed by respiratory epithelial cells is also the binding site for rhinovirus, the causative agent of most common colds.

BMPR1A has also been designated as CD292 (cluster of differentiation 290). The bone morphogenetic protein (BMP) receptors are a family of transmembrane serine/threonine kinases that include the type I receptors BMPR1A (this protein) and BMPR1B and the type II receptor BMPR2. β2 microglobulin is also known as Beta-2-microglobulin (B2M), is a component of MHC class I molecules which belongs to the beta-2-microglobulin family. B2M is present on all nucleated cells (excludes red blood cells). B2M associates not only with the alpha chain of MHC class I molecules, but also with class I-like molecules such as CD1 and Qa. An additional function of B2M is association with the HFE protein, together regulating the expression of hepcidin in the liver which targets the iron transporter ferroportin on the cytoplasmic membrane of enterocytes and macrophages for degradation resulting in decreased iron uptake from food and iron release from recycled red blood cells respectively. Loss of this function causes iron excess and hemochromatosis. Defects in B2M are the cause of hypercatabolic hypoproteinemia (HYCATHYP).

RAC-alpha serine/threonine-protein kinase (AKT1) is also known PKB, Protein kinase B alpha, PKB alpha, Proto-oncogene c-Akt and RAC-PK-alpha, which belongs to the protein kinase superfamily, AGC Ser/Thr protein kinase family and RAC subfamily and is expressed in prostate cancer and levels increase from the normal to the malignant state (at protein level). AKT1 is one of 3 closely related serine/threonine-protein kinases (AKT1, AKT2 and AKT3) called the AKT kinase, and which regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis. AKT is responsible of the regulation of glucose uptake by mediating insulin-induced translocation of the SLC2A4/GLUT4 glucose transporter to the cell surface. AKT regulates also the storage of glucose in the form of glycogen by phosphorylating GSK3A at 'Ser-21' and GSK3B at 'Ser-9', resulting in inhibition of its kinase activity. Phosphorylation of GSK3 isoforms by AKT is also thought to be one mechanism by which cell proliferation is driven. AKT regulates also cell survival via the phosphorylation of MAP3K5 (apoptosis signal-related kinase).

Cholera toxin is protein complex secreted by the bacterium Vibrio cholerae. It is responsible for the massive, watery diarrhea characteristic of cholera infection. Cholera enterotoxin subunit B (CTXB) pentameric ring directs the A subunit to its target by binding to the GM1 gangliosides present on the surface of the intestinal epithelial cells. It can bind five GM1 gangliosides. It has no toxic activity by itself.

Syntaxin-8 is a single-pass type IV membrane protein which belongs to the syntaxin family. It contains one t-SNARE coil homology domain. STX8 is highly expressed in heart, also found in brain, kidney, liver, lung, placenta, skeletal muscle, spleen and pancreas. STX8 is involved in protein trafficking from early to late endosomes via vesicle fusion and exocytosis. It as a vesicle trafficking protein functions in the early secretory pathway, possibly mediating retrograde transport form cis-golgi membrane to the ER.
