Description:   Vascular cell adhesion protein 1 (VCAM1) is also known as CD106, INCAM-100 and L1CAM, is a cell surface sialoglycoprotein belonging to the immunoglobulin superfamily. VCAM1 / CD106 contains 7 Ig-like C2-type (immunoglobulin-like) domains. CD106 / VCAM-1 is expressed on inflammed vascular endothelium, as well as on macrophage-like and dendritic cell types in both normal and inflammed tissue. L1CAM / VCAM-1 is Important in cell-cell recognition and appears to function in leukocyte-endothelial cell adhesion. CD106 / VCAM1 interacts with the beta-1 integrin VLA4 on leukocytes, and mediates both adhesion and signal transduction. The VCAM1 / VLA4 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation. INCAM-100 / VCAM1 binds to ECMV-D capsid proteins and acts as a receptor for this virus.

Description:   Clusterin shares homology with the small heat shock protein family of molecular chaperones. The mature secreted form of the protein is a glycosylated, 80-kDa disulphide-linked heterodimer of alpha and beta subunits (produced by internal cleavage). Clusterin is expressed in virtually all tissues and found in all human fluids. Description:   Human CD137 (4-1BB) is a costimulatory molecule of the tumour necrosis factor (TNF) receptor superfamily. The glycoprotein 4-1BB is expressed mainly on activated CD4 + and CD8 + T cells and binds to a high-affinity ligand (4-1BBL) expressed on several antigen-presenting cells such as macrophages and activated B cells. Upon ligand binding, 4-1BB is associated with the tumour receptor-associated factors (TRAF), the adaptor protein and mediates downstream signalling events including the activation of NF-kappaB and cytokine production. 4-1BB signalling either by binding to 4-1BBL or by antibody ligation delivers signals for T cell activation and growth as well as monocyte proliferation and B cell survival, and plays a important role in the amplification of T cell-mediated immune responses. In contrast, it can also enhance activation-induced T cell apoptosis when triggered by engagement of the TCR/CD3 complex. In addition, the 4-1BB/4-1BBL costimulatory pathway has been shown to augment secondary CTL responses to several viruses and increase antitumour immunity. 4-1BB is therefore a promising candidate for immunotherapy of human cancer.

Description:   CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

Description:   Nectin-4 (PVRL4) is a type I transmembrane glycoprotein which belongs to the nectin family of Ig superfamily proteins. It contains two Ig-like C2-type domains and one Ig-like V-type domain. PVRL4 seems to be involved in cell adhesion through trans-homophilic and -heterophilic interactions, the latter including specifically interactions with nectin-1. It does not act as receptor for alpha-herpesvirus entry into cells. It is predominantly expressed in placenta, the embryo and breast carcinoma. But it is not detected in normal breast epithelium. The soluble form is produced by proteolytic cleavage at the cell surface (shedding), probably by ADAM17. Mutations in this gene are the cause of ectodermal dysplasia-syndactyly syndrome type 1, an autosomal recessive disorder.

Description:   Leukocyte surface antigen CD47 is also known as Antigenic surface determinant protein OA3, Integrin-associated protein (IAP) and Protein MER6. CD47 contains 1 Ig-like V-type (immunoglobulin-like) domain. CD47 is very broadly distributed on normal adult tissues. CD47 has a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins and plays an important role in memory formation and synaptic plasticity in the hippocampus by similarity. CD47 is the receptor for SIRPA, binding to which prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells. Description:   Tryptophan 2,3-dioxygenase (TDO) is a heme-containing dioxygenase catalyzing the addition of molecular oxygen across the 2,3-double bond of the indole ring of tryptophan to form N-formylkynurenine (NFK). In Anopheles gambiae, TDO is the only enzyme able to catalyze the first and rate-limiting step in L-Trp catabolism through the kynurenine pathway. Tryptophan is an essential amino acid for protein synthesis and also the precursor for production of a number of neurotransmitters, such as serotonin and melatonin; in mosquitoes, the kynurenine pathway is essential for eye pigmentation. Conceivably, the tryptophan-using pathways should be regulated in a coordinated manner in mosquitoes as well as in other species and TDO activation/inactivation processes could play an essential role in these phenomena.
