Dermatopontin, also known as Tyrosine-rich acidic matrix protein, TRAMP and DPT, is a secreted protein which belongs to the dermatopontin family. DPT is expressed in various tissues, such as fibroblasts, heart, skeletal muscle, brain and pancreas. It seems to mediate adhesion by cell surface integrin binding. DPT may serve as a communication link between the dermal fibroblast cell surface and its extracellular matrix environment. DPT can enhance TGFB1 activity through interaction with decorin. In addition, DPT accelerates collagen fibril formation, stabilizes collagen fibrils against low-temperature dissociation and inhibits cell proliferation.

R-spondin-1 is also known as Roof plate-specific Spondin 1 (RSPO1) and cysteinerich and single thrombospondin domain containing protein 3 (Cristin 3), which is a secreted protein which belongs to the R-Spondin family and encodes a secreted activator protein with two cystein-rich, furin-like domains and one thrombospondin type 1 domain. All Rspondins regulate Wnt/β-catenin signaling, but have distinct expression patterns. Like other R-Spondins, R-Spondin-1 contains two adjacent cysteinerich furinlike domains (aa 34-135) with one potential N-glycosylation site, followed by a thrombospondin (TSP1) motif (aa 147-207) and a region rich in basic residues (aa 211-263). Only the furinlike domains are needed for β-catenin stabilisation. A putative nuclear localisation signal at the C-terminus may allow some expression in the nucleus.

Kit ligand (KITLG) is also known as stem cell factor (SCF), mast cell growth factor (MGF), steel factor (SF), which belongs to the SCF family, and is a widely expressed 28 - 40 kDa type I transmembrane glycoprotein. KITLG is the ligand for the receptor-type protein-tyrosine kinase KIT. SCF / MGF plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. KITLG / SCF binding can activate several signaling pathways. KITLG / SF Promotes phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase, and subsequent activation of the kinase AKT1. KITLG / SCF and KIT also transmit signals via GRB2 and activation of RAS, RAF1 and the MAP kinases MAPK1/ERK2 and/or MAPK3/ERK1. S100 calcium-binding protein P (S100P),a member of the S-100 family, is also known as protein S100-P, migration-inducing gene 9 protein (MIG9) and protein S100-E,which contains two EF-hand domains. As for subunit structure,S100P is homodimer and heterodimer with S100A1. S100P may function as calcium sensor and contribute to cellular calcium signaling. In a calcium-dependent manner, S100P functions by interacting with other proteins, such as EZR and PPP5C, and indirectly plays a role in physiological processes like the formation of microvilli in epithelial cells. Neurexophilin-1 (NXPH1) is a member of Neurexophilin family. NXPH1 consist of 271 amino acis. It contains a 21 amino acid signal peptide, 86 amino acid propeptide, and 164 amino acid mature protein. NXPH1 is expressed in subpopulations of neurons within the cerebral cortex, cerebellum and olfactory bulb that are thought to be inhibitory interneurons. In humans, NXPH2 and NXPH3 are most similar to NXPH1, sharing 84% and 64% aa identity within the mature region, respectively. By contrast, NXPH4 dost not bind a-neurexins. Genetic deletion of NXPH1 or NXPH3 produces no anatomical effect.

Semaphorin-5A (SEMA5A) is a member of the Semaphorin family of axon guidance molecules. SEMA5A is a 140 kDa protein. Class 5 Semaphorins are type I transmembrane glycoproteins with an N- terminal Sema domain and multiple juxtamembrane type 1 Thrombospondin (TSP) repeats within their extracellular domains. SEMA5A is expressed in neuroepithelial cells surrounding retinal axons, oligodendrocytes, the base of limb buds, the mesoderm surrounding cranial vessels , and the cardiac atrial septum and endocardial cushions, Human SEMA5A cDNA encodes a signal sequence, a extracellular domain (ECD), a transmembrane sequence and an cytoplasmic portion. SEMA5A mutations have been implicated in the genetic syndrome,cri-du-chat,while some polymorphisms may increase risk for neurodegenerative diseases such as Parkinson. The expression of SEMA5A may be upregulated in metastatic cancer cells and downregulated in autism.
