Description:
T cell immunoglobulin and mucin domain-3 (TIM3), also called hepatitis A virus cellular receptor 2 (HAVCR2), is a transmembrane glycoprotein of the TIM family of immune regulating molecules and plays an important role in the Th1-mediated immune response. TIM3 is expressed on the Th1 cells, CD8 T-cells, monocytes, and dendritic cells, but not on Th2 cells. TIM3 expressed by monocytes and dendritic cells facilitates phagocytosis of apoptotic cells and up-regulates cross-presentation of apoptotic cell-associated antigens through interaction with phosphatidylserine. Engagement of TIM3 by its ligand galectin-9 induces a range of immunosuppressive functions which enhance immune tolerance and inhibit anti-tumour immunity. Stimulation of TIM3 with an agonistic antibody promotes inflammation through the activation of innate immune cells. TIM3 is also regarded as a potential target molecule for immunotherapy. TIM3 and programmed cell death 1 (PD-1) as two important coinhibitory regulators of T cell responses, have been implicated with the T-cell dysfunction or exhaustion associated with chronic HBV infection including HBV-related HCC.

Human Chemokine (C-X-C motif) Ligand 4 (CXCL4) is expressed in megakaryocytes and stored in the alpha-granules of platelets. CXCL4 contains several heparin-binding sites at the C-terminal region and binds heparin with high affinity. The active CXCL4 protein is a tetramer. Human and mouse CXCL4 share 64% sequence identity. CXCL4 is chemotactic for neutrophils, fibroblasts and monocytes and plays a critical role in inflammation and wound repair. CXCL4 functions via a splice variant of the chemokine receptor CXCR3, known as CXCR3B. The major physiologic role of CXCL4 appears to be neutralisation of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. In contrast to other CXC chemokines, CXCL4 lacks chemotactic activity for polymorphonuclear granulocytes.

Kell blood group glycoprotein (KEL) is a single-pass type II membrane protein which belongs to the peptidase M13 family. It is expressed in Expressed at high levels in erythrocytes and testis, and, at lower levels, in skeletal muscle, tonsils, lymph node, spleen and appendix. KEL has been shown zinc endopeptidase with endothelin-3-converting enzyme activity. It cleaves EDN1, EDN2 and EDN3, with a marked preference for EDN3. It links via a single disulfide bond to the XK membrane protein that carries the Kx antigen.

Vaspin (Visceral Adipose-Specific SERPIN) is a newly described adipokine. Vaspin has three beta -sheets, nine alpha-helices, and one central loop; the structure is part of the set of distinctive features that are descriptive of Serpin family members. Vaspin is also a unique insulin sensitizing adipocytokine in obesity. A recent publication indicates that Vaspin mRNA expression in visceral fat is positively correlated with BMI and percent of body fat. and could be associated with parameters of obesity, insulin resistance, and glucose metabolism. These findings suggest a potential clinical use for Vaspin in ameliorating certain aberrations seen in the obesity metabolic syndrome.

Interleukin-23 receptor (IL-23R), is a member of the type I cytokine receptor family, type 2 subfamily. It is expressed by monocytes, Th1, Th0, NK and dendritic cells. Isoform 1 is specifically expressed in NK cells. IL-23 functions in innate and adaptive immunity and may participate in acute response to infection in peripheral tissues. IL-23 may be responsible for autoimmune inflammatory diseases and be important for tumourigenesis. IL-23R associates with IL-12RB1 to form the interleukin-23 receptor. It binds IL-23 and mediates T cells, NK cells and possibly certain macrophage/myeloid cells stimulation probably through activation of the Jak-Stat signalling cascade. Genetic variations in IL-23R are associated with inflammatory bowel disease type 17 (IBD17). IBD17 is a chronic, relapsing inflammation of the gastrointestinal tract with a complex etiology. Genetic variations in IL-23R also can cause susceptibility to psoriasis type 7.

CD99 is a type I transmembrane glycoprotein and the founding member of the CD99 family of molecules. The extracellular domain of CD99 contains no identifiable motifs, its cytoplasmic region, although short, does have signal transduction capability. Cells known to express CD99 include fibroblasts, neutrophils, T cells, double positive thymocytes, CD34+ stem cells, monocytes and endothelial cells. Two types of CD99 isoforms have been classified. Native human CD99 is referred to as the long, or type I isoform. The best studied type II isoform shows an Asp-Gly substitution for the C terminal 27 amino acids. The type I and II isoforms have distinctive signal transduction pathways (FAKsrc for type I PI3K plus srcERK1/2 for type II), and mediate clearly different biological outcomes. Homophilic interaction between CD99 on the neutrophil and CD99 on the endothelial cell regulates the transendothelial migration of neutrophils during inflammation. Human CD99 has 48% aa sequence identity to mouse CD99.

CD40 is a Type I Transmembrane Glycoprotein that belongs to the TNF Receptor Superfamily. CD40 is expressed in B cells, follicular dendritic cells, dendritic cells, activated monocytes, macrophages, endothelial cells, vascular smooth muscle cells, and several tumor cell lines. The extracellular domain of CD40 is characterized by Cysteine rich repeat regions. Interaction of CD40 with its ligand (CD40L) leads to aggregation of CD40 molecules, which in turn interact with cytoplasmic components to initiate signaling pathways. Several different TRAF proteins (adaptor proteins) have been identified to serves as mediators of the signal transduction. CD40 plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
