Description:
Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

Signal Transducer and Activator of Transcription 6 (STAT6) is a member of the STAT family of transcription factors. At least seven STATs exist: STAT1, 2, 3, 4, 5a, 5b, and 6. They are responsible for an array of cellular activities including regulating growth, survival, differentiation, motility, and the immune response. STAT6 plays a central role in exerting IL4 mediated biological responses. It is found to induce the expression of BCL2L1/BCL-X(L), which is responsible for the anti-apoptotic activity of IL4. Knockout studies in mice suggested the roles of this gene in differentiation of T helper 2 (Th2) cells, expression of cell surface markers, and class switch of immunoglobulins. STAT6 has been shown to interact with EP300, CREB-binding protein, NFKB1, Nuclear receptor coactivator 1, IRF4 and SND1.

Tachykinin 3 (TAC3) is a secreted protein that belongs to the Tachykinin family. Tachykinins are active peptides that excite neurons and evoke behavioral responses; they are potent vasodilators and secretagogues, and contract many smooth muscles in pregnancy. TAC3 is primarily expressed in the central and peripheral nervous systems and functions as a neurotransmitter. It is also expressed in the outer syncytiotrophoblast of the placenta and may be associated with pregnancy-induced hypertension and pre-eclampsia. TAC3 acts as the ligand for the neurokinin-3 receptor, mutations in this gene are associated with normosmic hypogonadotropic hypogonadism.

Pro-neuregulin-1,Neuregulin-1 beta 1（NRG1） is a single-pass type I membrane protein and belongs to the neuregulin family .It contains 1 EGF-like domain and 1 Ig-like C2-type (immunoglobulin-like) domain. Direct ligand for ERBB3 and ERBB4 tyrosine kinase receptors. The protein concomitantly recruits ERBB1 and ERBB2 coreceptors, resulting in ligand-stimulated tyrosine phosphorylation and activation of the ERBB receptors. The multiple isoforms perform diverse functions such as inducing growth and differentiation of epithelial, glial, neuronal, and skeletal muscle cells; inducing expression of acetylcholine receptor in synaptic vesicles during the formation of the neuromuscular junction; stimulating lobuloalveolar budding and milk production in the mammary gland and inducing differentiation of mammary tumor cells; stimulating Schwann cell proliferation; implication in the development of the myocardium such as trabeculation of the developing heart. Isoform 10 may play a role in motor and sensory neuron development.

Apoptosis Regulator BAX (BAX) belongs to the Bcl-2 family. BAX exists as a homodimer and is expressed in a wide variety of tissues. The Bax gene encodes different isoforms including Bax alpha (21 kDa) and Bax beta (24 kDa). Although both isoforms contain BH1, BH2 and BH3 domains, Bax beta has a unique carboxyl terminus and does not contain a hydrophobic transmembrane domain. BAX accelerates programmed cell death by binding to, and antagonizing the apoptosis repressor BCL2 or its adenovirus homolog E1B 19k protein. BAX also promotes activation of CASP3, and thereby apoptosis.
